ALTHOUGH INFLAMMATION EXERTS A FUNDAMENTAL PROTECTIVE AND RESTORATIVE FUNCTION AGAINST DAMAGE TO THE BODY (E.G., INFECTIONS OR TRAUMA), ITS ANOMALY INDUCEMENT CAN CAUSE DISEASE AND DEATH (E.G., AUTOIMMUNITY, ISCHEMIAS ASSOCIATED WITH ATHEROSCLEROSIS AND EVEN FAVORING CANCER). SPECIFICALLY, VASCULAR INFLAMMATION UNDERLIES THE MAIN FORMS OF MORBIDITY AND MORTALITY IN THE WORLD AS ACUTE DAMAGE TO VARIOUS ORGANS, SEPSIS, MYOCARDIAL INFARCTION OR STROKE. BECAUSE NEUTROPHILS, THE MOST ABUNDANT MYELOID CELLS IN THE BODY, ARE ESSENTIAL MEDIATORS OF BOTH THE DEFENSE AGAINST PATHOGENS AND VASCULAR DAMAGE, THEY HAVE BEEN THOROUGHLY STUDIED FOR MORE THAN A CENTURY. INTERESTINGLY, HOWEVER, TWO OF THE FUNDAMENTAL PROPERTIES OF ANY BIOLOGICAL PROCESS, SPACE AND TIME, HAVE BEEN LARGELY IGNORED WHEN STUDYING THE BIOLOGY OF INFLAMMATORY CELLS. THE SPATIAL PARAMETER IS IMPORTANT BECAUSE THE INFLAMMATORY RESPONSES ARE USUALLY RESTRICTED TO THE SPECIFIC FOCUS IN WHICH THE DAMAGE OCCURS, WHICH ALLOWS PREVENTING SYSTEMIC DAMAGE TO OTHER TISSUES. THE TIME PARAMETER IS ALSO ESSENTIAL BECAUSE INFLAMMATION HAS EVOLVED TO ANTICIPATE THE EXPOSURE OF THE ORGANISM TO EXTERNAL PATHOGENS. THIS TEMPORARY PROPERTY OF INFLAMMATION UNDERLIES THE WELL-KNOWN SUSCEPTIBILITY OF HUMANS AND OTHER MAMMALS TO ACUTE INFLAMMATORY PROCESSES AT CERTAIN TIMES OF THE DAY (I.E. CIRCADIANS). FOR THESE REASONS, WE PROPOSE THAT THE DETAILED UNDERSTANDING OF BOTH PARAMETERS WOULD ALLOW AN UNPRECEDENTED CONTROL OF THE INFLAMMATORY PROCESS SO AS TO PROMOTE PROTECTIVE RESPONSES, WHILE PREVENTING THOSE THAT CAUSE DISEASES. OUR OBSERVATIONS ALREADY PUBLISHED OR OBTAINED IN PREPARATION OF THIS PROPOSAL SUGGEST FOR THE FIRST TIME THAT A DEFINED GROUP OF MOLECULES, CELLULAR DOMAINS, AND ACCESSORY CELLS REGULATE THE SPATIAL AND TEMPORAL PERCEPTION OF NEUTROPHILS. _x000D_ in THIS PROPOSES we intend to DEFIN the MOLECULAR MEDIATORS AND CELLARS who endow the NEUTROphiles of SPACE AND TIME PERCEPTION AND underpin their capacity to identify INFLAMATORY DISEASE. IN THE FIRST OBJECTIVE WE WILL USE NOVEL PROTEOMIC ASSAYS ALONG WITH IN VIVO IMAGE OF INFLAMMATORY PROCESSES TO SUBCELLULAR RESOLUTION TO IDENTIFY THE MOLECULAR RECEPTORS AND TRANSDUCERS THAT ALLOW NEUTROPHIL TO DETECT ACTIVATING SIGNALS IN ITS ENVIRONMENT, AND THAT ULTIMATELY ORIGINATE LOCALISED VASCULAR DAMAGE. IN A SECOND OBJECTIVE WE PROPOSE TO DEFINE THE INTRICATE INSTRUCTORS OF CIRCADIAN TIME IN NEUTROPHILS, AND WE WILL LOOK FOR ACCESSORY CELLS IN THE PERIVASCULAR SPACE THAT ALLOW THE TEMPORAL COORDINATION OF NEUTROPHILS WITH THEIR ENVIRONMENT. WE WILL ALSO DETERMINE HOW THE LOSS OF TEMPORAL REGULATION IN NEUTROPHILS AFFECTS SUSCEPTIBILITY TO INFLAMMATORY DISEASE. IN A THIRD OBJECTIVE WE WILL CARRY OUT INTEGRATIVE PHYSIOLOGICAL ANALYSIS TO EXPLORE HOW THE SPATIAL AND TEMPORAL DYNAMICS OF NEUTROPHILS IN TURN REGULATES THE PHYSIOLOGY OF CERTAIN TISSUES IN THE ORGANISM TO PRESERVE HOMEOSTASIS AND PREVENT INFLAMMATION.
