LA TENDENCIA ACTUAL HACIA EL CONSUMO DE PRODUCTOS SALUDABLES, LA PRODUCCION SOSTENIBLE DE ALIMENTOS Y EL RESPETO POR EL MEDIO AMBIENTE ESTAN IMPULSANDO UNA REDUCCION EN EL USO DE COMPUESTOS QUIMICOS EN LA INDUSTRIA ALIMENTARIA, TANTO AQUELLOS QUE SE AÑADEN COMO CONSERVANTES COMO LOS QUE SE UTILIZAN COMO DESINFECTANTES. EN ESTE CONTEXTO, LOS SISTEMAS DE BIOCONSERVACION DE ALIMENTOS RESULTAN DE ESPECIAL INTERES, ASI COMO LOS SISTEMAS DE LIMPIEZA E HIGIENIZACION DE LAS INSTALACIONES QUE IMPLIQUEN DESINFECTANTES CUYO IMPACTO AMBIENTAL SEA MINIMO. LOS BACTERIOFAGOS CONSTITUYEN UNA OPCION PROMETEDORA, YA QUE SON ANTIMICROBIANOS NATURALES QUE INHIBEN ESPECIFICAMENTE BACTERIAS, Y POR TANTO, SON TOTALMENTE INOCUOS PARA OTROS SERES VIVOS Y PARA EL MEDIO AMBIENTE. SU POTENCIAL PARA PREVENIR Y ELIMINAR BACTERIAS PATOGENAS A LO LARGO DE LA CADENA ALIMENTARIA YA HA SIDO DEMOSTRADO. ADEMAS, CIERTAS PROTEINAS DERIVADAS DE BACTERIOFAGOS TIENEN ACTIVIDAD ANTIMICROBIANA (ENDOLISINAS) Y OTRAS (EXOPOLISACARIDO DESPOLIMERASAS) PRESENTAN LA CAPACIDAD DE DEGRADAR EL POLISACARIDO QUE CONSTITUYE LA MATRIZ EXTRACELULAR DE LAS BIOPELICULAS BACTERIANAS, LAS CUALES DIFICULTAN LOS PROCESOS DE LIMPIEZA Y DESINFECCION EN LAS INSTALACIONES ALIMENTARIAS. _x000D_ HASTA AHORA LA PRODUCCION Y COMERCIALIZACION DE PRODUCTOS DERIVADOS DE BACTERIOFAGOS HA ESTADO LIMITADA POR LA FALTA DE ESTUDIOS QUE PERMITIESEN ACORTAR LOS PROCESOS NECESARIOS PARA SU OBTENCION A BAJO COSTE. EL TRABAJO PREVIO REALIZADO EN NUESTRO LABORATORIO NOS HA PERMITIDO SELECCIONAR BACTERIOFAGOS Y PROTEINAS FAGICAS CON POTENCIAL APLICACION INDUSTRIAL (VB_SAUM-PHIIPLA-RODI (PHIIPLA-RODI), LYSH5, DPO7). EN ESTE CONTEXTO, UNO DE LOS OBJETIVOS DE ESTE PROYECTO ES OPTIMIZAR LA PROPAGACION, PURIFICACION, ALMACENAMIENTO E INACTIVACION DEL BACTERIOFAGO PHIIPLA-RODI QUE INFECTA A STAPHYLOCOCCUS AUREUS. UN SEGUNDO OBJETIVO ESTA ORIENTADO A COMPLETAR LA CARACTERIZACION DE LA ACTIVIDAD ENZIMATICA DE LA EXOPOLISACARIDO DESPOLIMERASA DPO7, CON OBJETO DE AMPLIAR SU ESPECTRO DE ACCION SOBRE BIOPELICULAS MIXTAS FORMADAS POR S. AUREUS EN COMBINACION CON OTROS MICROORGANISMOS. AMBOS OBJETIVOS ESTAN ENCAMINADOS A EVITAR LA CONTAMINACION POR S. AUREUS EN LA INDUSTRIA ALIMENTARIA, YA QUE LA ERRADICACION DE ESTA BACTERIA DEPENDE EN GRAN MEDIDA DE LA UTILIZACION DE SISTEMAS EFICACES DE DESINFECCION DE LAS INSTALACIONES ALIMENTARIAS JUNTO CON EL USO DE AGENTES ANTIMICROBIANOS QUE INHIBAN SU CRECIMIENTO EN LOS ALIMENTOS. EL TERCER OBJETIVO PLANTEA EL DISEÑO DE FORMULACIONES BASADAS EN TECNICAS DE MICRO Y NANOENCAPSULACION QUE FACILITEN LA UTILIZACION DE BACTERIOFAGOS Y PROTEINAS FAGICAS COMO AGENTES DE BIOCONTROL EN LA INDUSTRIA ALIMENTARIA. FINALMENTE, EL ULTIMO OBJETIVO ABORDA, COMO PASO PREVIO A LA UTILIZACION MASIVA DE PRODUCTOS DERIVADOS DE BACTERIOFAGOS, EL ESTUDIO DE LA RESPUESTA BACTERIANA QUE PROVOCAN, TANTO A NIVEL TRANSCRIPCIONAL COMO FENOTIPICO, A FIN DE GARANTIZAR QUE SU USO NO PROPICIARA UN INCREMENTO EN LA VIRULENCIA DE DICHAS BACTERIAS. THE CURRENT TREND TOWARDS THE CONSUMPTION OF HEALTHY PRODUCTS, THE SUSTAINABLE PRODUCTION OF FOOD AND RESPECT FOR THE ENVIRONMENT ARE DRIVING A REDUCTION IN THE USE OF CHEMICAL COMPOUNDS IN THE FOOD INDUSTRY, BOTH THOSE THAT ARE ADDED AS PRESERVATIVES AND THOSE USED AS DISINFECTANTS. IN THIS CONTEXT, FOOD BIOCONSERVATION SYSTEMS ARE OF PARTICULAR INTEREST, AS WELL AS CLEANING AND HYGIENISATION SYSTEMS FOR PLANTS INVOLVING DISINFECTANTS WITH MINIMAL ENVIRONMENTAL IMPACT. BACTERIOPHAGES ARE A PROMISING OPTION, AS THEY ARE NATURAL ANTIMICROBIALS THAT SPECIFICALLY INHIBIT BACTERIA, AND ARE THEREFORE TOTALLY HARMLESS TO OTHER LIVING BEINGS AND TO THE ENVIRONMENT. ITS POTENTIAL TO PREVENT AND ELIMINATE PATHOGENOUS BACTERIA ALONG THE FOOD CHAIN HAS ALREADY BEEN DEMONSTRATED. IN ADDITION, CERTAIN PROTEINS DERIVED FROM BACTERIOPHAGUS HAVE ANTIMICROBIAL ACTIVITY (ENDOLISINS) AND OTHERS (EXOPOLISACARIDO DEPOLIMERASAS) HAVE THE ABILITY TO DEGRADE THE POLYSACCHARIDE THAT CONSTITUTES THE EXTRACELLULAR MATRIX OF BACTERIAL BIOPELICULAS, WHICH HINDER CLEANING AND DISINFECTION PROCESSES IN FOOD FACILITIES. _x000D_ NOW THE PRODUCTION AND COMMERCIALISATION OF BACTERIOFAGED PRODUCTS HAVE BEEN LIMITED BY THE FALTE OF STUDIES that would make it possible to accept the necessities for their low cost management. THE PREVIOUS WORK CARRIED OUT IN OUR LABORATORY HAS ALLOWED US TO SELECT BACTERIOPHAGES AND PROTEINS FAGICAS WITH POTENTIAL INDUSTRIAL APPLICATION (VB_SAUM-PHIIPLA-RODI (PHIIPLA-RODI), LYSH5, DPO7). IN THIS CONTEXT, ONE OF THE OBJECTIVES OF THIS PROJECT IS TO OPTIMISE THE PROPAGATION, PURIFICATION, STORAGE AND INACTIVATION OF THE BACTERIOFAGO PHIIPLA-RODI THAT INFECTS STAPHYLOCOCCUS AUREUS. A SECOND OBJECTIVE IS AIMED AT COMPLETING THE CHARACTERISATION OF THE ENZYME ACTIVITY OF EXOPOLISACARIDO DESPOLIMERASE DPO7, IN ORDER TO EXPAND ITS SPECTRUM OF ACTION ON MIXED BIOPELICULES FORMED BY S. AUREUS IN COMBINATION WITH OTHER MICROORGANISMS. BOTH OBJECTIVES ARE AIMED AT AVOIDING CONTAMINATION BY S. AUREUS IN THE FOOD INDUSTRY, AS THE ERADICATION OF THIS BACTERIUM DEPENDS TO A LARGE EXTENT ON THE USE OF EFFECTIVE SYSTEMS OF DISINFECTION OF FOOD FACILITIES TOGETHER WITH THE USE OF ANTIMICROBIAL AGENTS THAT INHIBIT THEIR GROWTH IN FOOD. THE THIRD OBJECTIVE PROPOSES THE DESIGN OF FORMULATIONS BASED ON MICRO AND NANOENCAPSULATION TECHNIQUES THAT FACILITATE THE USE OF BACTERIOPHAGUS AND PROTEINS FAGICAS AS BIOCONTROL AGENTS IN THE FOOD INDUSTRY. FINALLY, THE LAST OBJECTIVE ADDRESSES, AS A STEP PRIOR TO THE MASSIVE USE OF PRODUCTS DERIVED FROM BACTERIOFAGOS, THE STUDY OF THE BACTERIAL RESPONSE THAT THEY CAUSE, BOTH AT A TRANSCRIPTIONAL AND PHENOTIC LEVEL, IN ORDER TO ENSURE THAT THEIR USE DID NOT LEAD TO AN INCREASE IN THE VIRULENCE OF THESE BACTERIA.
