    Los pacientes con dolor crónico (DC) sufren depresión y déficits cognitivos, desconociéndose las causas fisiopatológicas. El DC actúa como estrés crónico y en estudios preclínicos, nuestro grupo ha demostrado que esta asociación produce un empeoramiento en la neurogénesis adulta (NRGA) que puede ser causa de trastornos depresivos y déficits cognitivos. Esta posibilidad ha sido escasamente investigada en clínica por lógicos problemas metodológicos. El proyecto se desarrollará con una doble vertiente: clínica y preclínica. Pacientes con DC y depresión serán evaluados valorando la estabilización de memorias remotas relacionadas con NRGA. Este paradigma ha podido ser demostrado experimentalmente, por tanto realizaremos estudios de NRGA en ratas expuestas a DC que desarrollan conductas depresivas y déficit cognitivo. El LC proyecta al hipocampo y es posible que este input (noradrenérgico) impacte en el proceso de NRGA en respuesta al dolor. La actividad del LC puede ser medida mediante pupilometría incruenta. Valoraremos cambios pupilométricos producidos por la comorbilidad de depresión-dolor crónico-déficits cognitivo. Pretendemos evaluar consecuencias clínicas de esta comorbilidad como reflejo de cambios en la NRGA producidos por la inhibición de la inervación noradrenérgica. Investigaremos NRGA en animales con DC tras inhibición farmacogenética de la vía LC-hipocampo mediante la técnica DREADDs (designer receptors exclusively activated by designer drugs). (Spanish)

DC acts as chronic stress and in preclinical studies, our group has shown that this association results in a worsening of adult neurogenesis (NRGA) that can cause depressive disorders and cognitive deficits. This possibility has been scarcely investigated in the clinic due to logical methodological problems. The project will be developed with a double dimension: clinic and preclinical. Patients with DC and depression will be evaluated by assessing the stabilisation of remote NRGA-related memories. This paradigm has been experimentally demonstrated, therefore we will conduct studies of NRGA in rats exposed to DC that develop depressive behaviors and cognitive deficit. LC projects the hippocampus and this input (noradrenergic) may impact the NRGA process in response to pain. The activity of the LC can be measured by bloodless pupilometry. We will evaluate pupilometric changes produced by the comorbidity of depression-chronic pain-cognitive deficits. We intend to evaluate clinical consequences of this comorbidity as a reflection of changes in NRGA caused by inhibition of noradrenergic inervation. We will investigate NRGA in animals with DC after pharmacogenetic inhibition of the LC-hypocampus pathway using the DREADDs (designer receptors exclusively activated by designer drugs) technique.
