    Por su papel como precursoras de metástasis las células tumorales circulantes (CTCs) están adquiriendo un papel cada más relevante en la práctica clínica por su utilidad diagnóstica y prognóstica. El objetivo de este proyecto es caracterizar los genes y rutas de señalización más importantes para la supervivencia (resistencia a “anoikis” o muerte por ausencia de adhesión a sustrato) de las células tumorales circulantes (CTCs) de melanoma en el contexto de la adaptación metabólica a la disponibilidad de glucosa, un factor determinante en la progresión tumoral. Buscaremos identificar fármacos que bloqueen dichas rutas y validarlos como estrategias terapéuticas contra el desarrollo de melanoma metastático. Metodología: 1. Caracterizacíon de líneas celulares de melanoma humano por su sensibilidad a anoikis y desarrollo de líneas resistentes a anoikis en condiciones variables de disponibilidad de glucosa. 2. Análisis comparativo de los perfiles transcriptómicos y de respuesta a fármacos en células sensibles y resistentes a anoikis en situaciones variables de glucemia. 3. Identificación y caracterización de los genes y rutas de señalización clave para la supervivencia de CTCs y validación de los los compuestos identificados en este estudio como terapias anti-metástasis. 4. Análisis de la expresión de genes candidatos en tumores primarios y metastásicos por su potencial como marcadores de progresión tumoral y dianas terapéuticas. (Spanish)

    Because of their role as precursors of metastasis, circulating tumor cells (CTCs) are acquiring an increasingly relevant role in clinical practice because of their diagnostic and prognostic utility. The objective of this project is to characterise the genes and signaling routes most important for survival (resistance to “anoikis” or death due to lack of substrate adhesion) of circulating tumor cells (CTCs) of melanoma in the context of metabolic adaptation to glucose availability, a determining factor in tumor progression. We will seek to identify drugs that block these routes and validate them as therapeutic strategies against the development of metastatic melanoma. Methodology: 1. Characterisation of human melanoma cell lines for their sensitivity to anoikis and development of anoiki-resistant lines under variable glucose availability conditions. 2. Comparative analysis of transcriptomic and drug response profiles in sensitive and anoiki-resistant cells in variable glycemia situations. 3. Identification and characterisation of genes and key signaling pathways for CTC survival and validation of the compounds identified in this study as anti-metastasis therapies. 4. Analysis of the expression of candidate genes in primary and metastatic tumors for their potential as markers of tumor progression and therapeutic targets. (English)
