Las catinonas sintéticas es uno de los grupos más relevantes y numerosos de nuevas sutancias psicoativos (NSP). Todas ellas tienen estructuras químicas similares pero suficientemente peculiares para presentar un perfil de mecanismo de acción distinto y probablemente característico según el derivado. Se dispone únicamente de datos sobre la farmacología humana de la mefedrona (investigación previa de este grupo), aunque otras catinonas sintéticas poseen gran relevancia. La a-PVP y la metilona son consideradas por sus efectos como las alternativas legales a la cocaína y la MDMA, respectivamente. No existen estudios publicados en humanos de a-PVP ni metilona. En este proyecto se pretende estudiar el perfil de efectos y farmacocinética de ambas catinonas, a-PVP y metilona, y así completar la información de las catinonas sintéticas junto con la mefedrona. Objetivos 1. Conocer el potencial de abuso, los efectos subjetivos, fisiológicos, la farmacocinética y el metabolismo de la a-PVP en humanos tras su administración a dosis única. 2. Conocer el potencial de abuso, los efectos subjetivos, fisiológicos, la farmacocinética y el metabolismo de la metilona en humanos tras su administración a dosis única. Métodos Dos ensayos clínicos, uno para cada objetivo. El diseño de ambos ensayos será aleatorio, doble ciego, cruzados y controlados con placebo en sujetos sanos con previa experiencia en el consumo de anfetaminas, MDMA, catinonas sintéticas y/o cocaína. Se pretende comparar el perfil de efectos y potencial de abuso de la a-PVP y la metilona, con los propios de los psicoestimulantes clásicos de referencia, cocaína (comparador activo) para a-PVP y MDMA y mefedrona (comparadores activos) para metilona. They all have similar chemical structures but sufficiently peculiar to present a distinct and probably characteristic action mechanism profile depending on the derivative. Only data are available on the human pharmacology of mephedrone (previous research of this group), although other synthetic cathinones are of great relevance. A-PVP and methylone are considered for their effects as legal alternatives to cocaine and MDMA, respectively. There are no published human studies of a-PVP or methylone. This project aims to study the effects profile and pharmacokinetics of both catinones, a-PVP and methylone, and thus complete the information on synthetic cathinones together with mephedrone. Objectives 1. To know the potential for abuse, subjective, physiological effects, pharmacokinetics and metabolism of a-PVP in humans after single-dose administration. 2. To know the potential for abuse, subjective, physiological effects, pharmacokinetics and metabolism of methylone in humans after single-dose administration. Methods Two clinical trials, one for each goal. The design of both trials will be randomised, double-blind, cross-crossed, and placebo-controlled in healthy subjects with previous experience in the use of amphetamines, MDMA, synthetic cathinones or cocaine. The aim is to compare the profile of effects and potential for abuse of a-PVP and methylone with those of classical reference psychostimulants, cocaine (active comparator) for a-PVP and MDMA and mephedrone (active comparisons) for methylone.
