This new approach will be validated by the development of new small molecule-type compounds presenting selective inhibitory activity of the enzyme O-GlcNAc-hydrolase (OGA)._x000D_ The present project focuses on the research of the fundamental pathological characteristics of Alzheimer’s disease (hereinafter referred to as “AS”): the formation of intracellular balls of Tau protein (t) hyperphosphorylated, which make up one of the most recognizable and, at the same time, most destructive characteristics of this disease. Throughout the research activities that will be addressed in the project, the scientific knowledge of this condition will be deepened so that the results can be applied to the development of new drugs aimed at modifying the course of Alzheimer’s disease through innovative therapeutic routes._x000D_ It is important to note the degree of innovation introduced by the project when considering the validation of an innovative therapeutic strategy for Alzheimer’s disease. Specifically, an approximation of small molecules (small molecules) is proposed in order to prevent the formation of neurofibrillary balls that consist of accumulations of hyperphosphorylate protein associated with microtubules, with the ultimate aim of modifying the course of Alzheimer’s disease._x000D_ To achieve the general objectives of the INOA project, it will be necessary to carry out important research and development actions to enable the discovery of new compounds with selective inhibitory activity of the enzyme O-GlcNAc-hydrolase or OGA.     El proyecto INOA plantea el objetivo general de abordar una serie de actividades de investigación aplicada que permitan validar un innovador abordaje terapéutico de la enfermedad de Alzheimer, así como el posterior descubrimiento de potenciales candidatos a fármacos. Esta nueva aproximación será validada mediante el desarrollo de nuevos compuestos de tipo molécula pequeña (small molecule) que presenten actividad inhibitoria selectiva de la enzima O-GlcNAc-hidrolasa (OGA)._x000D_ El presente proyecto se focaliza en la investigación de las características patológicas fundamentales de la enfermedad de Alzheimer (en adelante “EA”): la formación de ovillos intracelulares de proteína Tau (t) hiperfosforilada, que conforman una de las características más reconocibles y, al mismo tiempo, más destructivas de esta enfermedad. A lo largo de las actividades de investigación que se abordarán en el proyecto, se profundizará en el conocimiento científico de esta condición para que los resultados puedan ser aplicados al desarrollo de nuevos fármacos orientados a la modificación del curso de la enfermedad de Alzheimer a través de rutas terapéuticas innovadoras._x000D_ Es importante señalar el grado de innovación que introduce el proyecto al plantear la validación de una innovadora estrategia terapéutica para la enfermedad de Alzheimer. Concretamente, se plantea una aproximación de moléculas pequeñas (small molecules) para lograr prevenir la formación de ovillos neurofibrilares que se componen de acúmulos de proteína t hiperfosforilada asociada a los microtúbulos, con el fin último de modificar el curso de la enfermedad de Alzheimer._x000D_ Para el cumplimiento de los objetivos generales del proyecto INOA, será necesario llevar a cabo importantes actuaciones de investigación y desarrollo para posibilitar el descubrimiento de nuevos compuestos con actividad inhibitoria selectiva de la enzima O-GlcNAc-hidrolasa u OGA.
