ASIMISMO, EN BIOPSIAS DE PIEL ESTUDIAREMOS LAS PROPIEDADES TOPOGRAFICAS DE LA SUPERFICIE DE LAS CELULAS VIVAS Y A NANOESCALA MEDIANTE MICROSCOPIA DE FUERZA ATOMICA. FINALMENTE, SE EVALUARAN LOS EFECTOS EN LA INFLAMACION DE LA PIEL PRODUCIDOS TRAS LA ADMINISTRACION EN DIETA DE SPPDP11, LA LEVADURA QUE MEJORES RESULTADOS ARROJE EN LOS ESTUDIOS PREVIOS Y NANOPARTICULAS LIPIDICAS CONTENIENDO ARGININA Y NANOPARTICULAS DE SEDA, AMBAS SELECCIONADAS POR SUS CONOCIDAS PROPIEDADES ANTI-INFLAMATORIAS Y CICATRIZANTES EN MAMIFEROS. EN CUANTO AL EMPLEO DE NANOPARTICULAS EN PECES, TODAVIA ESTA EN UNA FASE MUY INICIAL PERO ENTENDER SU INTERACCION CON LOS SISTEMAS BIOLOGICOS ES DE GRAN INTERES PARA PROMOVER QUE SU APLICACION SEA SEGURA. THE AIM OF THE PRESENT PROPOSAL IS TO MODULATE THE INFLAMMATION OF THE SKIN OF GILTHEAD SEABREAM (SPARUS AURATA) THROUGH THE DIET. WE PROPOSE TO CHARACTERIZE THE SKIN USING POWERFUL TECHNIQUES OF IMAGE ANALYSIS THAT ARE THE ULTRASOUND -ECHOGRAPHY- AND THE COMPUTED TOMOGRAPHY-SCANNER, MICRO-CT-, THANKS TO THE EQUIPMENTS RECENTLY ACQUIRED BY THE ANIMAL EXPERIMENTATION SERVICE OF OUR UNIVERSITY. THIS WILL ALLOW US TO ASSESS ITS USEFULNESS FOR THE STUDY OF CHANGES IN THE SKIN IN RELATION TO THE BODY ZONE, INFLAMMATION OR DIET SUPPLIED. THESE ARE NON-LETHAL AND NON-INVASIVE TECHNIQUES, WHICH WILL PERMIT US TO OBTAIN REAL-TIME IMAGES OF THE HEALING PROCESS, AS WELL AS ALLOWING US TO DETERMINE THE HARDNESS OR RESISTANCE OF THE SKIN IN THOSE AREAS, THE THICKNESS OR THE PRESENCE OF FAT IN THE FISH BODY. FURTHERMORE, WE WILL STUDY THE TOPOGRAPHIC PROPERTIES OF THE SURFACE OF LIVING CELLS AT NANOSCALE BY ATOMIC FORCE MICROSCOPY IN SKIN BIOPSIES. IT IS INTENDED TO KNOW THE CHANGES THAT OCCUR IN THE MICROBIOTA OF THE SKIN AND IN THE KERATINOCYTES (BY MEANS OF TRANSCRIPTOMIC STUDIES) AFTER CONTACTING SKIN CELLS WITH A PROBIOTIC (OBTAINED FROM THE SKIN OF HEALTHY GILTHEAD SEAMBREAM SPECIMENS, SHEWANELLA PUTREFACIENS, SPPDP11) OR VIBRIO HARVEYI (PATHOGENIC BACTERIA TAKEN AS A MODEL FOR CAUSING SEVERE SKIN ULCERS), WHICH WILL ALLOW US TO ADVANCE IN THE HOST-PROBIOTIC AND HOST-PATHOGEN RELATIONSHIPS, HELPING TO DEVELOP POSSIBLE EFFECTIVE TREATMENTS. THIS PROPOSAL INTENDS TO OPTIMIZE A TECHNIQUE TO PRODUCE INTRADERMAL EDEMA AND TO EVALUATE THE EFFECTS THAT THE EXTRACELLULAR PRODUCTS OF SPPDP11, V. HARVEYI AND MARINE YEASTS PRODUCE IN SUCH EDEMA. WE INTEND TO STUDY 3 STRAINS OF MARINE AND OLEAGINOUS EXTREMOPHILIC YEASTS, WHOSE GENOME IS SEQUENCED AND THAT HAVE GREAT BIOTECHNOLOGICAL POTENTIAL, WITH THE INTENTION OF FINDING BENEFICIAL EFFECTS AND TO EVALUATE THEIR USE IN AQUACULTURE, TO KNOW THEIR ANTI-INFLAMMATORY AND ANTIOXIDANTS PROPERTIES AND THEIR REPERCUSSION IN THE NUTRITIONAL COMPOSITION OF FISH. FINALLY, THE EFFECTS ON SKIN INFLAMMATION PRODUCED AFTER SPPDP11 DIET, THE BEST-YIELDING YEAST AND LIPID NANOPARTICLES CONTAINING ARGININE AND SILK NANOPARTICLES, BOTH SELECTED FOR THEIR KNOWN ANTI-INFLAMMATORY AND HEALING PROPERTIES IN MAMMALS, WILL BE STUDIED. REGARDING THE USE OF NANOPARTICLES IN FISH, IT IS STILL AT A VERY EARLY STAGE BUT UNDERSTANDING THEIR INTERACTIONS WITH THE BIOLOGICAL SYSTEMS IS OF GREAT INTEREST FOR PROMOTING A SAFE APPLICATION. WE WILL WORK FOR STRENGTHEN THE IMMUNE SYSTEM OF FARMED FISH IN ORDER TO REDUCE THE USE OF CHEMICALS, CONTRIBUTING TO A MORE SUSTAINABLE USE OF SEAS AND OCEANS. (English)
