LA PRODUCCION DE LECHE DE OVEJA EN ESPAÑA CONSTITUYE EL 21% DEL TOTAL DE LA UE, SIENDO CASTILLA Y LEON LA REGION MAS PRODUCTORA Y CON MAYOR CENSO DE NUESTRO PAIS. ESTA PRODUCCION ESTA DESTINADA CASI EN SU TOTALIDAD A LA ELABORACION DE QUESO DE ALTA CALIDAD, POR LO QUE EL AUMENTO DEL RENDIMIENTO QUESERO, ALTAMENTE DEPENDIENTE DEL CONTENIDO DE PROTEINA Y GRASA DE LA LECHE, ASI COMO SUS PROPIEDADES DE COAGULACION (MCP), SON DE GRAN INTERES PARA ESTE SECTOR. EL PRESENTE PROYECTO, PROTMILKOMA, PRETENDE UTILIZAR LA TECNICA DE SECUENCIACION MASIVA PARALELA DEL TRANSCRIPTOMA, O RNASEQ, PARA LA DETECCION DE VARIANTES GENETICAS CON EFECTOS SOBRE CARACTERES DE PRODUCCION LECHERA, PRINCIPALMENTE EL PORCENTAJE DE PROTEINA (PP) Y LAS MCP. ADEMAS, EL PROYECTO PROPONE CARACTERIZAR LA MICROBIOTA DE LA GLANDULA MAMARIA SANA DE LA OVEJA, CUYA COMPOSICION Y EQUILIBRIO PUEDEN AFECTAR AL BIENESTAR Y PRODUCTIVIDAD DEL ANIMAL, ASI COMO A LA CALIDAD Y PROPIEDADES TECNOLOGICAS DE LA LECHE PRODUCIDA. PARA ELLO SE PLANTEAN TRES OBJETIVOS ESPECIFICOS. EN EL PRIMERO SE ANALIZARA EL TRANSCRIPTOMA MAMARIO DE OVEJAS CON FENOTIPO DIVERGENTE PARA PP, REALIZANDOSE UN EXPERIMENTO DE RNASEQ EN DOS RAZAS OVINAS, CHURRA Y ASSAF (4 ANIMALES DE ALTO PP VS 4 AN. BAJO PP, EN CADA RAZA). DE TODAS LAS VARIANTES IDENTIFICADAS CON FRECUENCIAS EXTREMAS ENTRE LOS DOS GRUPOS, ESPECIALMENTE LAS LOCALIZADAS EN LOS GENES DE LAS CASEINAS Y PROTEINAS DEL LACTOSUERO Y LAS DE LOS GENES IDENTIFICADOS COMO EXPRESADOS DIFERENCIALMENTE (DEGS), SE SELECCIONARA UN GRUPO DE SNPS PARA SU INCLUSION EN UN CHIP DE BAJA DENSIDAD, EL MILKPROTEIN-CHIP (3.2K), CUYO GENOTIPADO EN POBLACIONES COMERCIALES (600 CHURRAS Y 600 ASSAF) SERVIRA PARA CONFIRMAR POSIBLES ASOCIACIONES CON CARACTERES LECHEROS. EN EL SEGUNDO OBJETIVO, DE FORMA PARALELA, SE REALIZARA UN EXPERIMENTO DE RNASEQ PARA COMPARAR EL TRANSCRIPTOMA MAMARIO DE OVEJAS ASSAF CON FENOTIPO EXTREMO DIVERGENTE PARA CARACTERES DE COAGULABILIDAD DE LA LECHE Y DE RENDIMIENTO QUESERO (4 AN. DE OPTIMAS MCP VS 4 AN. DE DEFICIENTES MCP). PARA ELLO, SE REALIZARA UNA DETERMINACION DE VARIOS PARAMETROS RELACIONADOS CON LAS MCP (RCT, K20, A30, A45, Y A90, RENDIMIENTO QUESERO, CONTENIDO EN CITRATO DE LA LECHE Y PH). DE NUEVO, DE LA VARIABILIDAD DIVERGENTE IDENTIFICADA EN ESTE EXPERIMENTO SE SELECCIONARAN SNPS (LOCALIZADOS EN LOS GENES DE LAS PROTEINAS DE LA LECHE, EN LOS DEGS Y LOS GENES DE LA RUTA DE SINTESIS DEL CITRATO) PARA INCLUIRLOS EN EL MILKPROTEIN-CHIP. LOS GENOTIPOS DE ESTE CHIP GENERADOS EN 400 OVEJAS ASSAF CON MEDIDAS DISPONIBLES PARA LOS CARACTERES TECNOLOGICOS EN ESTUDIO PERMITIRAN IDENTIFICAR VARIANTES ASOCIADAS CON DICHOS FENOTIPOS. FINALMENTE, EL TERCER OBJETIVO CARACTERIZARA LA MICROBIOTA MAMARIA DE LAS OVEJAS ASSAF MUESTREADAS EN EL OBJETIVO 2 MEDIANTE EL ANALISIS DE LA VARIABILIDAD DEL GEN 16S RRNA MEDIANTE SECUENCIACION, QUE SE USARA PARA LA ASIGNACION TAXONOMICA Y CUANTIFICACION DE LA MICROBIOTA MAMARIA. MEDIANTE POSTERIOR ANALISIS DE ASOCIACION SE PRETENDE IDENTIFICAR CIERTOS PERFILES MICROBIANOS QUE PUDIERAN TENER UN EFECTO POSITIVO PARA EL RENDIMIENTO QUESERO DE LA LECHE. ADEMAS, LAS MUESTRAS CON MAYOR VARIABILIDAD MICROBIANA SERAN SOMETIDAS A UNA SECUENCIACION COMPLETA DEL DNA BACTERIANO UTILIZANDO UN ENFOQUE METAGENOMICO. EN CONJUNTO, PROTMILKOMA, PRETENDE, A TRAVES DE LAS MAS MODERNAS HERRAMIENTAS Y TECNOLOGIAS GENOMICAS, PROPORCIONAR SOLUCIONES PRACTICAS AL SECTOR OVINO LECHERO. (Spanish)

THE PRODUCTION OF SHEEP’S MILK IN SPAIN CONSTITUTES 21 % OF THE TOTAL EU, WITH CASTILE AND LEON BEING THE MOST PRODUCING REGION AND WITH THE HIGHEST CENSUS OF OUR COUNTRY. THIS PRODUCTION IS DESTINED ALMOST ENTIRELY TO THE MANUFACTURE OF HIGH QUALITY CHEESE, SO THE INCREASE IN THE CHEESE YIELD, HIGHLY DEPENDENT ON THE PROTEIN AND FAT CONTENT OF THE MILK, AS WELL AS ITS COAGULATION PROPERTIES (MCP), ARE OF GREAT INTEREST FOR THIS SECTOR. THIS PROJECT, PROTMILKOMA, AIMS TO USE THE PARALLEL MASS SEQUENCE TECHNIQUE OF THE TRANSCRIPTOME, OR RNASEQ, FOR THE DETECTION OF GENETIC VARIANTS WITH EFFECTS ON DAIRY PRODUCTION CHARACTERS, MAINLY THE PERCENTAGE OF PROTEIN (PP) AND MCP. IN ADDITION, THE PROJECT PROPOSES TO CHARACTERISE THE MICROBIOTA OF THE HEALTHY BREAST GLAND OF THE SHEEP, WHOSE COMPOSITION AND BALANCE CAN AFFECT THE WELFARE AND PRODUCTIVITY OF THE ANIMAL, AS WELL AS THE QUALITY AND TECHNOLOGICAL PROPERTIES OF THE MILK PRODUCED. TO THIS END, THREE SPECIFIC OBJECTIVES ARE SET. IN THE FIRST ONE THE MAMMARY TRANSCRIPTOME OF SHEEP WITH DIVERGENT PHENOTYPE FOR PP WAS ANALYSED, PERFORMING AN EXPERIMENT OF RNASEQ IN TWO SHEEP BREEDS, CHURRA AND ASSAF (4 ANIMALS HIGH PP VS 4 AN. UNDER PP, IN EACH BREED). FROM ALL VARIANTS IDENTIFIED WITH EXTREME FREQUENCIES BETWEEN THE TWO GROUPS, ESPECIALLY THOSE LOCATED IN THE GENES OF THE CASEINS AND WHEY PROTEINS AND THOSE OF THE GENES IDENTIFIED AS DIFFERENTIALLY EXPRESSED (DEGS), A GROUP OF SNPS WILL BE SELECTED FOR INCLUSION IN A LOW-DENSITY CHIP, THE MILKPROTEIN-CHIP (3.2K), WHOSE GENOTYPING IN COMMERCIAL POPULATIONS (600 CHURRAS AND 600 ASSAF) WILL SERVE TO CONFIRM POSSIBLE ASSOCIATIONS WITH DAIRY CHARACTERS. IN THE SECOND OBJECTIVE, IN PARALLEL, A RNASEQ EXPERIMENT WAS PERFORMED TO COMPARE THE MAMMARY TRANSCRIPTOME OF ASSAF SHEEP WITH DIVERGING EXTREME PHENOTYPE FOR MILK COAGULABILITY AND CHEESE YIELD CHARACTERS (4 AN. OF OPTIMAL MCP VS 4 AN. OF POOR PCM). TO DO THIS, A DETERMINATION OF SEVERAL PARAMETERS RELATED TO MCPS (RCT, K20, A30, A45, AND A90, CHEESE YIELD, MILK CITRATE CONTENT AND PH) WILL BE PERFORMED. AGAIN, THE DIVERGING VARIABILITY IDENTIFIED IN THIS EXPERIMENT WILL SELECT SNPS (LOCATED IN THE GENES OF MILK PROTEINS, IN THE SDRS AND THE GENES OF THE CITRATE SYNTHESIS ROUTE) TO BE INCLUDED IN THE MILKPROTEIN-CHIP. THE GENOTYPES OF THIS CHIP GENERATED IN 400 ASSAF SHEEP WITH MEASURES AVAILABLE FOR THE TECHNOLOGICAL CHARACTERS UNDER STUDY WILL ALLOW TO IDENTIFY VARIANTS ASSOCIATED WITH THESE PHENOTYPES. FINALLY, THE THIRD OBJECTIVE WILL CHARACTERISE THE MAMMARY MICROBIOTA OF THE ASSAF SHEEP SAMPLED IN OBJECTIVE 2 BY ANALYSING THE VARIABILITY OF THE 16S RRNA GENE BY SEQUENCING, TO BE USED FOR TAXONOMIC ALLOCATION AND QUANTIFICATION OF THE MAMMARY MICROBIOTA. THE AIM IS TO IDENTIFY MICROBIAL PROFILES THAT COULD HAVE A POSITIVE EFFECT ON MILK’S CHEESE YIELD. IN ADDITION, SAMPLES WITH GREATER MICROBIAL VARIABILITY WILL BE SUBJECTED TO COMPLETE SEQUENCING OF BACTERIAL DNA USING A METAGENOMIC APPROACH. TOGETHER, PROTMILKOMA AIMS, THROUGH THE MOST MODERN TOOLS AND GENOMIC TECHNOLOGIES, TO PROVIDE PRACTICAL SOLUTIONS TO THE DAIRY SHEEP SECTOR. (English)
