    LA RESPUESTA AL ESTRES ESTA BIEN CARACTERIZADA EN SUS GRANDES RASGOS EN PECES Y PARTICULARMENTE EN LA DORADA (SPARUS AURATA), TELEOSTEO MARINO DE ALTO VALOR COMERCIAL, CUYO CULTIVO PUEDE IMPLICAR PROCESOS DE ESTRES TIPICOS DE LA PRACTICA ACUICOLA (I.E. DENSIDAD Y MANIPULACION). DE ESTE MODO, EL OBJETIVO GENERAL ES MEJORAR LOS PROCESOS DE GESTION ACUICOLA EN LOS PECES EN EL MODELO DE LA DORADA. ESTE OBJETIVO SE QUIERE CONSEGUIR MEDIANTE HERRAMIENTAS DE DETECCION Y DE ATENUACION DEL ESTRES EN TODOS LOS PUNTOS RELEVANTES DEL EJE HIPOTALAMO-PITUITARIO-INTERRENAL (HPI) Y SUS RESPUESTAS. SIN EMBARGO, UN ELEMENTO IMPORTANTE EN RELACION A LA CAPACIDAD DE MONITORIZACION DE LOS ESTADOS DE ESTRES ES QUE NO SE DISPONE EN LA ACTUALIDAD DE ALGUNOS DE LOS MARCADORES RELEVANTES EN LA CARACTERIZACION DE DICHA RESPUESTA. DE ESTE MODO, SE TERMINARIA DE COMPLETAR LAS HERRAMIENTAS METODOLOGICAS (I.E. CLONACION DE TODOS LOS RECEPTORES DE CORTISOL, QUE CONSTITUIRIA EL OBJETIVO ESPECIFICO 1), ASI COMO EL EMPLEO DE LAS NUEVAS APROXIMACIONES TECNOLOGICAS COMO EL USO DE MICROARRAYS PARA EL ESTUDIO DEL HPI. ASI, EN ESTE ESTUDIO SE ABORDARAN DOS SITUACIONES EXPERIMENTALES DE INTERES PARA LA PRACTICA ACUICOLA Y QUE CONSTITUYEN DOS DE LOS OBJETIVOS ESPECIFICOS DEL MISMO: I) ESTRES CRONICO POR DENSIDAD CON SUSTANCIAS QUE ATENUEN LA ACTIVACION DEL EJE HPI (OBJETIVO ESPECIFICO 2) Y II) ESTRES INDUCIDO POR TRANSPORTE Y USO DE ANESTESICOS BAJO DOSIS DE SEDACION (OBJETIVO ESPECIFICO 4) DADA ADEMAS LA RECIENTE PROLIFERACION DE AGENTES POTENCIALMENTE ANTI-ESTRES. SEGUN EL PARADIGMA CLASICO, EL ORDEN DE ACTIVACION TENDRIA UNA DIRECCION DESDE EL CENTRO (HIPOTALAMO) HACIA LA PERIFERIA. SIN EMBARGO, LA INDUCCION NO NECESARIAMENTE SE PRODUCE EN ESTE ORDEN, SINO QUE PUEDE INICIARSE EN LA PERIFERIA, EN PARTICULAR BAJO CONDICIONES DE PRESENCIA DE PATOGENOS, PARASITOS O CONTAMINANTES. EN ESTOS CASOS NO SOLAMENTE LA INDUCCION DE LA RESPUESTA DEL EJE EMPIEZA EN LA PERIFERIA SINO QUE YA SE DESARROLLAN RESPUESTAS LOCALES EN ESPECIAL EN LAS MUCOSAS DEL INTESTINO, PIEL O BRANQUIAS QUE DESPUES, Y VIA MEDIADORES EN SANGRE, ACTIVAN EL EJE HPI. EL PRESENTE PROYECTO INCORPORA DOS DISEÑOS EXPERIMENTALES QUE CONTEMPLAN ESAS NUEVAS APROXIMACIONES Y QUE CONSTITUYEN DOS DE LOS OBJETIVOS ESPECIFICOS DEL MISMO: I) EL TRATAMIENTO CON NANOPARTICULAS (OBJETIVO ESPECIFICO 3), Y II) EL TRATAMIENTO CON LIPOPOLISACARIDOS (LPS) DE BACTERIAS (OBJETIVO ESPECIFICO 5). EL SISTEMA DE ESTRES SERA ESTUDIADO MEDIANTE LA DETERMINACION DE: I) NIVELES HORMONALES (CORTISOL) Y METABOLICOS (GLUCOSA, LACTATO, ETC) EN PLASMA; II) NIVELES DE METABOLITOS Y ACTIVIDADES ENZIMATICAS EN HIGADO; III) EXPRESION DE ACTIVIDADES ENZIMATICAS DE ESTRES OXIDATIVO EN HIGADO Y BRANQUIA; IV) EXPRESION DE CRH, CRH-BP Y TRH EN HIPOTALAMO POR RT-Q-PCR; V) EXPRESION DE POMC A Y POMC B EN HIPOFISIS POR RT-Q-PCR; VI) EXPRESION DE RECEPTORES GLUCOCORTICOIDE Y MINERALCORTICOIDE EN DIFERENTES ORGANOS DEL EJE HPI ASI COMO EN ORGANOS METABOLICOS (HIGADO) Y PERIFERICOS (BRANQUIA, INTESTINO, PIEL) MEDIANTE RT-Q-PCR. FINALMENTE SE UTILIZARA LA EXPRESION GENICA EN MICROARRAY PARA LA CONSECUCION DE LOS OBJETIVOS 2 Y 4. LAS APLICACIONES FINALES DE ESTE ESTUDIO SON IMPORTANTES, YA QUE DE LOS RESULTADOS PODREMOS OBTENER INFORMACION SUSCEPTIBLE DE SER UTILIZADA PARA UNA OPTIMIZACION DEL CULTIVO DE S. AURATA EN RELACION A LOS PROCESOS DE ESTRES. LOS AUTORES PRETENDEMOS TANTO LA PUBLICACION CIENTIFICA COMO INFORMES TECNICOS PARA EL SECTOR. (Spanish)

THIS OBJECTIVE IS TO BE ACHIEVED BY MEANS OF DETECTION AND STRESS MITIGATION TOOLS IN ALL THE RELEVANT POINTS OF THE HYPOTHALAME-PITUITARY-INTERRENAL AXIS (HPI) AND ITS ANSWERS. HOWEVER, AN IMPORTANT ELEMENT IN RELATION TO THE MONITORING CAPACITY OF STRESS STATES IS THAT AT PRESENT SOME OF THE RELEVANT MARKERS ARE NOT AVAILABLE IN THE CHARACTERISATION OF THAT RESPONSE. IN THIS WAY, THE METHODOLOGICAL TOOLS (I.E. CLONING OF ALL CORTISOL RECEPTORS, WHICH WOULD CONSTITUTE THE SPECIFIC OBJECTIVE 1), AS WELL AS THE USE OF NEW TECHNOLOGICAL APPROACHES SUCH AS THE USE OF MICROARRAYS FOR THE STUDY OF HPI, WOULD BE COMPLETED. THUS, THIS STUDY WILL ADDRESS TWO EXPERIMENTAL SITUATIONS OF INTEREST FOR ACUICOLA PRACTICE AND WHICH CONSTITUTE TWO OF THE SPECIFIC OBJECTIVES OF THIS STUDY: CHRONIC STRESS BY DENSITY WITH SUBSTANCES THAT ATTENUATE THE ACTIVATION OF THE HPI AXIS (SPECIFIC OBJECTIVE 2) AND II) STRESS INDUCED BY TRANSPORT AND USE OF ANESTHETICS LOW DOSES OF SEDATION (SPECIFIC OBJECTIVE 4) GIVEN THE RECENT PROLIFERATION OF POTENTIALLY ANTI-STRESS AGENTS. ACCORDING TO THE CLASSICAL PARADIGM, THE ORDER OF ACTIVATION WOULD HAVE A DIRECTION FROM THE CENTER (HYPOTALAMOM) TO THE PERIPHERY. HOWEVER, THE INDUCEMENT DOES NOT NECESSARILY OCCUR IN THIS ORDER, BUT CAN BE INITIATED AT THE PERIPHERY, IN PARTICULAR UNDER CONDITIONS OF PRESENCE OF PATHOGENS, PARASITES OR CONTAMINANTS. IN THESE CASES NOT ONLY THE INDUCTION OF THE AXIS RESPONSE BEGINS AT THE PERIPHERY BUT LOCAL RESPONSES ARE ALREADY DEVELOPED ESPECIALLY IN THE MUCOUS MEMBRANES OF THE INTESTINE, SKIN OR GILLS THAT AFTERWARD, AND VIA BLOOD MEDIATORS, ACTIVATE THE HPI AXIS. THIS PROJECT INCORPORATES TWO EXPERIMENTAL DESIGNS THAT CONTEMPLATE THESE NEW APPROACHES AND WHICH CONSTITUTE TWO OF THE SPECIFIC OBJECTIVES OF THE PROJECT: TREATMENT WITH NANOPARTICULAE (SPECIFIC OBJECTIVE 3), AND II) TREATMENT WITH LIPOLYSACCHARIDES (LPS) OF BACTERIA (SPECIFIC OBJECTIVE 5). THE STRESS SYSTEM WILL BE STUDIED BY DETERMINING: HORMONE (CORTISOL) AND METABOLIC LEVELS (GLUCOSE, LACTATE, ETC.) IN PLASMA; LEVELS OF METABOLITES AND ENZYMATIC ACTIVITIES IN LIVERS; EXPRESSION OF ENZYMATIC ACTIVITIES OF OXIDATIVE STRESS IN LIVER AND GILL; EXPRESSION OF CRH, CRH-BP AND TRH IN HYPOTHALAME BY RT-Q-PCR; EXPRESSION OF POMC A AND POMC B IN HYPOPHYSIS BY RT-Q-PCR; EXPRESSION OF GLUCOCORTICOID AND MINERALCORTICOID RECEPTORS IN DIFFERENT ORGANS OF THE HPI AXIS AS WELL AS IN METABOLIC ORGANS (LIVER) AND PERIPHERAL ORGANS (BRANCH, INTESTINE, SKIN) USING RT-Q-PCR. FINALLY, THE GENIC EXPRESSION IN MICROARRAY WILL BE USED FOR THE ACHIEVEMENT OF OBJECTIVES 2 AND 4. THE FINAL APPLICATIONS OF THIS STUDY ARE IMPORTANT, SINCE FROM THE RESULTS WE WILL BE ABLE TO OBTAIN INFORMATION THAT CAN BE USED TO OPTIMISE THE CULTIVATION OF S. AURATA IN RELATION TO STRESS PROCESSES. THE AUTHORS INTEND BOTH SCIENTIFIC PUBLICATION AND TECHNICAL REPORTS FOR THE SECTOR. (English)
