    La esquizofrenia es un constructo clínico sin un sustrato cerebral claramente identificado. Creemos que el estudio de la conectividad cerebral funcional puede permitir progresar en la resolución de este problema por i) su relación con la representación cortical de las percepciones y con la coordinación de los grupos neurales implicados en tareas cognitivas ii) su marcada relación con la trasmisión inhibitoria, alterada en las psicosis de modo no uniforme y iii) su asociación con variables genéticas relevantes para la esquizofrenia. Además, la actual definición exclusivamente clínica de la esquizofrenia puede permitir incluir grupos biológicamente heterogéneos, por lo que la caracterización de la conectividad funcional puede contribuir a identificar un grupo de pacientes basado en una plausible alteración cerebral común, con manifestaciones clínicas y cognitivas características, que pueda servir para la identificación de nuevas dianas terapéuticas. Estudiaremos 240 pacientes con esquizofrenia, 240 familiares sanos y 240 controles sanos con electroencefalografía (EEG) para valorar los patrones de conectividad funcional y con exámenes clínicos y cognitivos. Estudiaremos parámetros conectivos cerebrales funcionales con EEG a partir de registros de electroencefalografía como medida funcional. Usaremos medidas de conectividad global basadas en la teoría de grafos. Nuestra hipótesis es que existirá un grupo caracterizado por alteraciones conectivas funcionales , con mayor probabilidad de presentar ciertas variantes genéticas de riesgo y quizá algún patrón cínico distintivo. Este subtipo de pacientes puede servir de base para una clasificación basada en la disfunción cerebral en las psicosis, en el que resultaría de interés explorar la utilidad de terapias orientadas a paliar la disfunción inhibitoria. Este proyecto se coordina con otro orientado a valorar la variación genética relevante para el estudio de la disfunción conectiva. (Spanish)

    Schizophrenia is a clinical construct without a clearly identified brain substrate. We believe that the study of functional brain connectivity may allow progress in solving this problem by i) its relationship with the cortical representation of perceptions and the coordination of neural groups involved in cognitive tasks ii) its marked relationship with inhibitory transmission, altered in psychosis in a nonuniform way and iii) its association with genetic variables relevant to schizophrenia. In addition, the current exclusively clinical definition of schizophrenia may allow to include biologically heterogeneous groups, so the characterisation of functional connectivity can help identify a group of patients based on a plausible common brain disorder, with characteristic clinical and cognitive manifestations, which may serve to identify new therapeutic targets. We will study 240 patients with schizophrenia, 240 healthy family members and 240 healthy controls with electroencephalography (EEG) to assess functional connectivity patterns and clinical and cognitive tests. We will study functional cerebral connective parameters with EEG from electroencephalography records as a functional measure. We will use global connectivity measures based on graph theory. Our hypothesis is that there will be a group characterised by functional connective alterations, with a greater probability of presenting certain genetic variants of risk and perhaps some distinctive cynic pattern. This subtype of patients can serve as a basis for a classification based on brain dysfunction in psychosis, in which it would be interesting to explore the usefulness of therapies aimed at alleviating inhibitory dysfunction. This project is coordinated with another project aimed at assessing the genetic variation relevant to the study of connective dysfunction. (English)
