    LOS CITRICOS CONSTITUYEN LA PRINCIPAL FRUTA PARA CONSUMO EN FRESCO EN ESPAÑA. ADEMAS, ESPAÑA ES EL PRIMER PAIS EXPORTADOR A NIVEL MUNDIAL DE ESTOS FRUTOS PARA CONSUMO EN FRESCO. ESTE HECHO EXPLICA LA IMPORTANCIA DE LA CONSERVACION POSTCOSECHA DE LOS CITRICOS. EL HONGO PENICILLIUM DIGITATUM ES EL PRINCIPAL PATOGENO POST-COSECHA DE LOS CITRICOS. EL CONTROL DE ESTE PATOGENO SE LLEVA A CABO CON FUNGICIDAS SINTETICOS, QUE CADA VEZ PRESENTAN MAYORES INCONVENIENTES DEBIDO A LA PREOCUPACION CRECIENTE DE LOS CONSUMIDORES POR PROBLEMAS RELACIONADOS CON LA SALUD Y LA CONTAMINACION AMBIENTAL, LA EXIGENCIA DE LOS MERCADOS DEMANDANTES DE PRODUCTOS HORTOFRUTICOLAS CON ¿RESIDUOS CERO¿ Y LA APARICION DE CEPAS RESISTENTES A LOS FUNGICIDAS EXISTENTES EN EL MERCADO, POR LO QUE LA BUSQUEDA DE ALTERNATIVAS SIGUE SIENDO UNA PRIORIDAD. EN ESTE CONTEXTO, ESTE PROYECTO PLANTEA EL OBJETIVO GENERAL DE PROFUNDIZAR EN EL CONOCIMIENTO DE LOS MECANISMOS DE PATOGENESIS/VIRULENCIA DE P. DIGITATUM, ASI COMO EN LOS MECANISMOS DE RESISTENCIA DE LOS FRUTOS CITRICOS A LA INFECCION POR ESTE PATOGENO, CON EL FIN DE ENCONTRAR ALTERNATIVAS AL USO DE FUNGICIDAS SINTETICOS PARA EL CONTROL DE PODRIDOS DURANTE LA POSTCOSECHA. EL PROYECTO SE PLANTEA COMO CONTINUACION DE DOS PROYECTOS ANTERIORES LIDERADOS RESPECTIVAMENTE POR LOS DOS IPS SOLICITANTES DE ESTE PROYECTO. PARA ESTUDIAR LOS MECANISMOS DE PATOGENICIDAD DISPONEMOS DE UN MUTANTE DE P. DIGITATUM CON VIRULENCIA MUY REDUCIDA, QUE RECUPERA LA VIRULENCIA AL ESCINDISRSE EL CASETE DE RESISTENCIA A HIGROMICINA CON EL QUE FUE OBTENIDO A PARTIR DE UNA CEPA SILVESTRE. ADEMAS, RESULTADOS PREVIOS DE NUESTRO GRUPO INDICAN QUE EL METABOLISMO DE IONES METALICOS ES ESENCIAL PARA LA PATOGENICIDAD DE ESTE HONGO, YA QUE UN QUELANTE DE IONES METALICOS PREVIENE COMPLETAMENTE EL DESARROLLO DE LA INFECCION. ADEMAS, LA ADICION DE IONES MELALICOS, COMO HIERRO Y COBRE, REVIERTE LA PROTECCION. POR OTRA PARTE, RESULTADOS DE NUESTRO GRUPO DEMUESTRAN QUE LA APLICACION DE LUZ AZUL LED EN FRUTOS COSECHADOS AUMENTA SU RESISTENCIA A LA INFECCION POR P. DIGITATUM Y QUE LAS FOSOFOLIPASAS JUEGAN UN PAPEL IMPORTANTE EN LA RESISTENCIA INDUCIDA. A PESAR DE QUE EN OTROS SISTEMAS VEGETALES SE CONOCE CON DETALLE LA REGULACION HORMONAL DE LA RESPUESTA DE DEFENSA FRENTE A DISTINTOS PATOGENOS, EN EL CASO DE LOS FRUTOS CITRICOS EL PAPEL DE LAS HORMONAS EN LA RESISTENCIA A LA INFECCION POR P. DIGITATUM ES APENAS CONOCIDO. CON ESTOS ANTECEDENTES NOS PROPONEMOS ABORDAR LOS SIGUIENTES OBJETIVOS PARCIALES: 1) IDENTIFICAR MEDIANTE APROXIMACIONES ¿OMICAS¿ LA CAUSA DE LA PERDIDA DE VIRULENCIA EN UN MUTANTE DE P. DIGITATUM; 2) ESTUDIAR EL PAPEL DEL HIERRO EN P. DIGITATUM Y SU VIRULENCIA Y 3) ESTUDIAR EL PAPEL DE LAS HORMONAS ETILENO, ACIDO ABSCISICO Y JASMONICO, Y SU INTERACCION, EN LA RESPUESTA DE LOS FRUTOS CITRICOS A LA INFECCION POR P. DIGITATUM Y EN LA RESISTENCIA INDUCIDA FRENTE A ESTE PATOGENO POR LA LUZ AZUL. TENIENDO EN CUENTA QUE LA FOSFOLIPASA A2 ES RELEVANTE EN LA EFICACIA DE LA LUZ AZUL, SE HARA ESPECIAL HINCAPIE EN EL ESTUDIO DE CAMBIOS HORMONALES Y EN EXPRESION GENICA GLOBAL QUE SE PRODUCE AL INHIBIR ESTA ENZIMA DURANTE LA INDUCCION DE RESISTENCIA. LOS RESULTADOS OBTENIDOS PUEDEN ABRIR NUEVAS ALTERNATIVAS DE CONTROL NO CONTAMINANTES Y FACTIBLES PARA SU UTILIZACION INDUSTRIAL. (Spanish)

THE CONTROL OF THIS PATHOGEN IS CARRIED OUT WITH SYNTHETIC FUNGICIDES, WHICH ARE INCREASINGLY INCONVENIENT DUE TO THE GROWING CONCERN OF CONSUMERS ABOUT HEALTH AND ENVIRONMENTAL POLLUTION PROBLEMS, THE DEMAND OF THE DEMANDING MARKETS FOR FRUIT AND VEGETABLE PRODUCTS WITH ZERO RESIDUES AND THE APPEARANCE OF STRAINS RESISTANT TO FUNGICIDES EXISTING ON THE MARKET, SO THE SEARCH FOR ALTERNATIVES REMAINS A PRIORITY. IN THIS CONTEXT, THIS PROJECT PROPOSES THE GENERAL OBJECTIVE OF DEEPENING THE KNOWLEDGE OF THE MECHANISMS OF PATHOGENESIS/VIRULENCE OF P. DIGITATUM, AS WELL AS THE MECHANISMS OF RESISTANCE OF CITRIC FRUITS TO INFECTION BY THIS PATHOGEN, IN ORDER TO FIND ALTERNATIVES TO THE USE OF SYNTHETIC FUNGICIDES FOR THE CONTROL OF ROTS DURING POST-HARVEST. THE PROJECT IS PROPOSED AS A CONTINUATION OF TWO PREVIOUS PROJECTS LED RESPECTIVELY BY THE TWO IPS APPLICANTS FOR THIS PROJECT. TO STUDY THE MECHANISMS OF PATHOGENICITY WE HAVE A MUTANT OF P. DIGITATUM WITH VERY REDUCED VIRULENCE, WHICH RECOVERS VIRULENCE TO ESCINDISRSE THE CASSETTE OF RESISTANCE TO HYGROMYCIN WITH WHICH IT WAS OBTAINED FROM A WILD STRAIN. IN ADDITION, PREVIOUS RESULTS OF OUR GROUP INDICATE THAT THE METABOLISM OF METAL IONS IS ESSENTIAL FOR THE PATHOGENICITY OF THIS FUNGUS, SINCE A CHELATING OF METAL IONS COMPLETELY PREVENTS THE DEVELOPMENT OF THE INFECTION. IN ADDITION, THE ADDITION OF MELALIC IONS, SUCH AS IRON AND COPPER, REVERSES THE PROTECTION. ON THE OTHER HAND, RESULTS FROM OUR GROUP SHOW THAT THE APPLICATION OF BLUE LED LIGHT ON HARVESTED FRUITS INCREASES ITS RESISTANCE TO INFECTION BY P. DIGITATUM AND THAT PHOSOPHOLIPAS PLAY AN IMPORTANT ROLE IN INDUCED RESISTANCE. ALTHOUGH IN OTHER PLANT SYSTEMS THE HORMONAL REGULATION OF THE DEFENSE RESPONSE AGAINST DIFFERENT PATHOGENS IS KNOWN IN DETAIL, IN THE CASE OF CITRIC FRUITS THE ROLE OF HORMONES IN THE RESISTANCE TO INFECTION BY P. DIGITATUM IS HARDLY KNOWN. AGAINST THIS BACKGROUND, WE INTEND TO ADDRESS THE FOLLOWING PARTIAL OBJECTIVES: 1) IDENTIFY BY APPROXIMATIONS THE CAUSE OF THE LOSS OF VIRULENCE IN A P. DIGITATUM MUTANT; 2) STUDY THE ROLE OF IRON IN P. DIGITATUM AND ITS VIRULENCE AND 3) STUDY THE ROLE OF HORMONES ETHYLENE, ABSCISIC AND JASMONICO ACID, AND THEIR INTERACTION, IN THE RESPONSE OF CITRIC FRUITS TO INFECTION BY P. DIGITATUM AND IN THE RESISTANCE INDUCED AGAINST THIS PATHOGEN BY BLUE LIGHT. CONSIDERING THAT PHOSPHOLIPASE A2 IS RELEVANT IN THE EFFICACY OF BLUE LIGHT, SPECIAL HARA HINCAPIES IN THE STUDY OF HORMONAL CHANGES AND IN GLOBAL GENIC EXPRESSION THAT OCCURS BY INHIBITING THIS ENZYME DURING THE INDUCTION OF RESISTANCE. THE RESULTS OBTAINED CAN OPEN NEW CONTROL ALTERNATIVES THAT ARE NON-POLLUTING AND FEASIBLE FOR INDUSTRIAL USE. (English)
