DEFINIR ESTAS DOS CARACTERISTICAS DEL RECEPTOR CB1 PUEDE CONDUCIR A IDENTIFICAR QUE CASCADA DE SEÑALIZACION O QUE INTERACCIONES CORRIENTE ABAJO DEL RECEPTOR SON NECESARIAS BLOQUEAR O CO-ACTIVAR CON OTROS FARMACOS PARA LOGRAR EL PROCESO BIOLOGICO PERSEGUIDO: PROLIFERACION, MIGRACION O DIFERENCIACION. EL PRESENTE PROYECTO PROPONE CARACTERIZAR LA LOCALIZACION Y EL INTERACTOMA DEL RECEPTOR CB1 TANTO EN EL DESARROLLO IN VITRO DE OLIGODENDROCITOS DE LA MEDULA COMO TRAS UNA LESION MEDULAR PARA RESOLVER UN PROBLEMA SIN SOLUCION ACTUAL: LA GENERACION DE UNA TERAPIA FARMACOLOGICA QUE INDUZCA SELECTIVAMENTE LOS PROCESOS BIOLOGICOS NECESARIOS PARA CONSEGUIR REMIELINIZAR LOS AXONES QUE SE HAN PRESERVADO EN UN AMBIENTE DE INFLAMACION CRONIFICADA, COMO SUCEDE TRAS UNA LESION MEDULAR. THE PROREMIELINISING ACTIVITY OF CANNABINOIDS HAS SHOWN THERAPEUTIC POTENTIAL IN VARIOUS PATHOLOGIES. REGARDLESS OF THE ETIOLOGY OF THESE, AXONAL REMIELINIZACION REQUIRES THE PROLIFERATION AND MIGRATION OF THE PRECURSORS OF OLIGODENDROCYTES (OPCS) TO THE DESMIELINIZADA ZONE AND THEIR DIFFERENTIATION TO PREMIELINIZANT OLIGODENDROCYTES. OUR RESULTS AND THOSE OF OTHER GROUPS SHOW THAT CANNABINOIDS ACT AT ALL THESE STAGES. _x000D_ our WORK HYPOTESIS is that the DIFFERENTIAL ACTIONS DERIVATED OF RECEPTOR CB1 DEPENDED YOUR DEVELOPMENT DIFFERENT LOCALISATION AND interacts TO THE LONG OF THE DISTINTS STATE DEVELOPMENT OF OPCS. DEFINING THESE TWO CB1 RECEPTOR CHARACTERISTICS MAY LEAD TO IDENTIFYING WHICH SIGNALING CASCADE OR WHICH DOWNSTREAM INTERACTIONS OF THE RECEPTOR ARE NECESSARY TO BLOCK OR CO-ACTIVATE WITH OTHER PHARMACOS TO ACHIEVE THE DESIRED BIOLOGICAL PROCESS: PROLIFERATION, MIGRATION OR DIFFERENTIATION. THIS PROJECT PROPOSES TO CHARACTERISE THE LOCATION AND INTERACTION OF THE CB1 RECEPTOR BOTH IN THE IN VITRO DEVELOPMENT OF OLIGODENDROCYTES OF THE MARROW AND AFTER A SPINAL INJURY TO SOLVE A PROBLEM WITH NO CURRENT SOLUTION: THE GENERATION OF A PHARMACOLOGICAL THERAPY THAT SELECTIVELY INDUCES THE BIOLOGICAL PROCESSES NECESSARY TO ACHIEVE REMIELINISING THE AXONS THAT HAVE BEEN PRESERVED IN AN ENVIRONMENT OF CHRONIFIED INFLAMMATION, AS HAPPENS AFTER A MEDULLARY INJURY.
