ESTOS ESFUERZOS SE TRADUCEN EN UNA MAYOR TECNIFICACION DE LAS EXPLOTACIONES, DONDE EL EMPLEO DE TECNOLOGIAS REPRODUCTIVAS INNOVADORAS OCUPA UN LUGAR IMPORTANTE. ENTRE ESTAS TECNOLOGIAS DESTACA LA TRANSFERENCIA DE EMBRIONES (TE) POR SUS TRANSCENDENTALES APLICACIONES, INCLUYENDO EL MOVIMIENTO DE RECURSOS GENETICOS (EMBRIONES) CON MINIMO RIESGO DE TRANSMISION DE ENFERMEDADES, CON REDUCIDO COSTE DEL TRANSPORTE Y SIN PROBLEMAS DE BIENESTAR ASOCIADOS AL MISMO. PESE A QUE LA TE HA SIDO DEMANDADA POR EL SECTOR DESDE HACE DECADAS, SU UTILIZACION ES PRACTICAMENTE INEXISTENTE DEBIDO, PRINCIPALMENTE, A LA NECESIDAD DE EMPLEAR TECNICAS QUIRURGICAS PARA LAS TES A LAS RECEPTORAS. SIN EMBARGO, LOS PROGRESOS REALIZADOS EN LOS ULTIMOS AÑOS INDICAN QUE EL EMPLEO COMERCIAL DE LA TE A CORTO PLAZO ES INCUESTIONABLE. EL GRUPO SOLICITANTE DEL PRESENTE PROYECTO ES PIONERO DE LA TECNICA DE TRANSFERENCIA NO QUIRURGICA EN LA PROFUNDIDAD DE UN CUERNO UTERINO, TANTO CON EMBRIONES FRESCOS COMO CRIOPRESERVADOS. ESTO HA SIDO POSIBLE GRACIAS AL DESARROLLO DE DIFERENTES PROYECTOS DEL PLAN ESTATAL DE I+D+I Y DE CONTRATOS DE INVESTIGACION CON EMPRESAS. COMO RESULTADOS DE ESAS INVESTIGACIONES, HOY EN DIA SE DISPONE DE UN METODO PRACTICO Y EFICIENTE PARA TRANSFERIR POR VIA NO QUIRURGICA EMBRIONES FRESCOS, CULTIVADOS DURANTE 24 H O VITRIFICADOS. LA UTILIZACION DE EMBRIONES CULTIVADOS DURANTE 24 H PERMITE TAN SOLO EL TRANSPORTE NACIONAL O, EN EL MEJOR DE LOS CASOS, INTRACONTINENTAL, DE LOS EMBRIONES DESDE SU ORIGEN HASTA LA GRANJA RECEPTORA. LA UTILIZACION DE EMBRIONES VITRIFICADOS PRESENTA ALGUNOS INCONVENIENTES, INCLUYENDO LA NECESIDAD DE ELEVAR SUSTANCIALMENTE EL NUMERO DE EMBRIONES POR TRANSFERENCIA Y LA UTILIZACION DE NITROGENO LIQUIDO (N2L) PARA SU CONSERVACION Y TRANSPORTE, LO CUAL DIFICULTA EL TRANSPORTE AEREO (POR LAS ESTRICTAS NORMAS REGULADAS POR LA IATA) Y LA LOGISTICA EN LA GRANJA DE DESTINO, DONDE NO SIEMPRE SE PUEDE DISPONER DE TANQUES Y DE N2L PARA REALIZAR LA MANIPULACION PRE-CALENTAMIENTO. POR ELLO, EXISTE UN INTERES CRECIENTE POR LA CONSERVACION DE EMBRIONES EN AUSENCIA DE N2L POR PERIODOS DE TIEMPO COMPATIBLES CON SU TRANSPORTE INTERNACIONAL. NOSOTROS HEMOS COMPROBADO QUE LAS MORULAS PERMANECEN VIABLES A 37ºC DURANTE 48 H SIN ALCANZAR EL ESTADIO DE BLASTOCISTO ECLOSIONADO, LO CUAL ES FUNDAMENTAL POR RAZONES SANITARIAS. ESA TEMPERATURA LIMITA LA CONSERVACION DE LOS BLASTOCISTOS A TAN SOLO 24 HORAS, AUNQUE DICHO PERIODO PUEDE AUMENTARSE A 48 HORAS UTILIZANDO TEMPERATURAS DE 25ºC. EN ESTE ESCENARIO, NUESTRA PROPUESTA SE DIRIGE A LA CONSERVACION DE MORULAS Y BLASTOCISTOS PORCINOS DURANTE 72 H MEDIANTE EL EMPLEO DE VARIOS MEDIOS DE CONSERVACION Y TEMPERATURAS QUE DESACELEREN EL DESARROLLO EMBRIONARIO, MANTENIENDO SU VIABILIDAD. POR OTRO LADO, PARA MEJORAR LA EFICIENCIA DE LOS PROGRAMAS DE TE, EL PRESENTE PROYECTO PROPONE EL USO DE INFUSIONES DE PLASMA SEMINAL Y CITOQUINAS DURANTE EL ESTRO, TANTO EN LAS CERDAS DONANTES COMO EN LAS RECEPTORAS. LOS RESULTADOS DEL PROYECTO TENDRAN UN ELEVADO IMPACTO CIENTIFICO, POR LA NOVEDAD DE LOS OBJETIVOS, SOCIAL, YA QUE POR PRIMERA VEZ SE DISPONDRIA DE UN SISTEMA PRACTICO PARA COMERCIALIZAR EMBRIONES PORCINOS NO CRIOPRESERVADOS DE FORMA SENCILLA Y SEGURA, Y ECONOMICO, POR LAS VENTAJAS SEÑALADAS ANTERIORMENTE. THESE EFFORTS TRANSLATE INTO A GREATER TECHNOLOGICALISATION OF FARMS, WHERE THE USE OF INNOVATIVE REPRODUCTIVE TECHNOLOGIES OCCUPIES AN IMPORTANT PLACE. AMONG THESE TECHNOLOGIES STANDS OUT THE TRANSFER OF EMBRYOS (TE) FOR THEIR TRANSCENDENTAL APPLICATIONS, INCLUDING THE MOVEMENT OF GENETIC RESOURCES (EMBRYONS) WITH MINIMAL RISK OF TRANSMISSION OF DISEASES, WITH REDUCED COST OF TRANSPORT AND WITHOUT ASSOCIATED WELFARE PROBLEMS. ALTHOUGH IT HAS BEEN DEMANDED BY THE SECTOR FOR DECADES, ITS USE IS PRACTICALLY NON-EXISTENT, MAINLY DUE TO THE NEED TO USE SURGICAL TECHNIQUES FOR TES TO RECEIVERS. HOWEVER, THE PROGRESS MADE OVER THE LAST FEW YEARS INDICATES THAT THE SHORT-TERM COMMERCIAL EMPLOYMENT OF TE IS UNQUESTIONABLE. THE APPLICANT GROUP FOR THIS PROJECT IS A PIONEER OF THE TECHNIQUE OF NON-SURGICAL TRANSFER IN THE DEPTH OF A UTERINE HORN, BOTH WITH FRESH AND CRYOPRESERVATED EMBRYOS. THIS HAS BEEN POSSIBLE THANKS TO THE DEVELOPMENT OF DIFFERENT PROJECTS OF THE STATE R & D & I PLAN AND RESEARCH CONTRACTS WITH COMPANIES. AS A RESULT OF THESE INVESTIGATIONS, TODAY THERE IS A PRACTICAL AND EFFICIENT METHOD TO TRANSFER FRESH EMBRYOS, CULTIVATED FOR 24 HOURS OR VITRIFIED BY VIA NON-SURGICAL. THE USE OF EMBRYOS CULTIVATED FOR 24 HOURS ALLOWS ONLY THE NATIONAL OR, AT BEST, INTRACONTINENTAL TRANSPORT OF EMBRYOS FROM THEIR ORIGIN TO THE RECEIVING FARM. THE USE OF VITRIFIED EMBRYOS HAS SOME DRAWBACKS, INCLUDING THE NEED TO SUBSTANTIALLY INCREASE THE NUMBER OF EMBRYOS BY TRANSFER AND THE USE OF LIQUID NITROGEN (N2L) FOR THEIR CONSERVATION AND TRANSPORT, WHICH HINDERS AIR TRANSPORT (DUE TO THE STRICT RULES REGULATED BY IATA) AND LOGISTICA IN THE DESTINATION FARM, WHERE TANKS AND N2L CANNOT ALWAYS BE AVAILABLE FOR PRE-HEATING HANDLING. THEREFORE, THERE IS AN INCREASING INTEREST IN THE CONSERVATION OF EMBRYOS IN THE ABSENCE OF N2L FOR PERIODS OF TIME COMPATIBLE WITH THEIR INTERNATIONAL TRANSPORT. WE HAVE FOUND THAT MORULAS REMAIN VIABLE AT 37 °C FOR 48 H WITHOUT REACHING THE BLASTOCYST STAGE HATCHED, WHICH IS ESSENTIAL FOR HEALTH REASONS. THIS TEMPERATURE LIMITS BLASTOCYST CONSERVATION TO ONLY 24 HOURS, ALTHOUGH THIS PERIOD CAN BE INCREASED TO 48 HOURS USING TEMPERATURES OF 25 °C. IN THIS SCENARIO, OUR PROPOSAL IS DIRECTED TO THE CONSERVATION OF MORULAS AND PORCINE BLASTOCYSTS FOR 72 H THROUGH THE USE OF VARIOUS MEANS OF CONSERVATION AND TEMPERATURES THAT SLOW THE EMBRYONIC DEVELOPMENT, MAINTAINING ITS VIABILITY. ON THE OTHER HAND, TO IMPROVE THE EFFICIENCY OF TEA PROGRAMS, THIS PROJECT PROPOSES THE USE OF SEMINAL PLASMA INFUSIONS AND CYTOKINES DURING ESTRUS, BOTH IN DONOR BRISTLES AND RECIPIENTS. THE RESULTS OF THE PROJECT WILL HAVE A HIGH SCIENTIFIC IMPACT, DUE TO THE NOVELTY OF THE OBJECTIVES, SOCIAL, SINCE FOR THE FIRST TIME A PRACTICAL SYSTEM WOULD BE AVAILABLE TO MARKET PIG EMBRYOS NOT CRYOPRESERVED IN A SIMPLE AND SAFE WAY, AND ECONOMICALLY, DUE TO THE ADVANTAGES MENTIONED ABOVE.
