DISTINTOS EXOPOLISACARIDOS SON UTILIZADOS PARA FORMAR LA MATRIZ DEL BIOFILM (CELULOSA, ALGINATO, ACIDO COLANICO), PERO SORPRENDENTEMENTE UNA AMPLIA VARIEDAD DE BACTERIAS FILOGENETICAMENTE MUY DISTANTES RECURREN AL MISMO EXOPOLISACARIDO, -1,6-N-ACETYLGLUCOSAMINE (PNAG), PARA ESTE PROPOSITO. ESTA OBSERVACION SUGIERE QUE LA MATRIZ FORMADA POR PNAG CONFIERE VENTAJAS ESPECIALES FRENTE A OTRO TIPO DE MATRICES, Y SUBRAYA LA NECESIDAD DE ESTUDIAR ESTE EXOPOLISACARIDO EN PROFUNDIDAD. EN ESTE PROYECTO, PROPONEMOS UTILIZAR LA BACTERIA PATOGENA STAPHYLOCOCCUS AUREUS COMO MODELO PARA ENTENDER LAS SINGULARIDADES QUE EL PNAG TIENE PARA LA FORMACION DEL BIOFILM. PARA ELLO, PROPONEMOS ESTUDIAR LOS MECANISMOS MOLECULARES QUE REGULAN LA SINTESIS Y ACTIVACION DE LA PNAG SINTASA, EL MECANISMO DE TRANSDUCCION DE SEÑAL QUE CONECTA LAS CONDICIONES AMBIENTALES CON LA PRODUCCION DE PNAG Y EL NUMERO Y LOCALIZACION DE PNAG SINTASAS EN LA MEMBRANA DE LA BACTERIA. ADEMAS, UTILIZAREMOS LA EXPRESION HETEROLOGA DE PNAG EN SALMONELLA ENTERITIDIS, UNA BACTERIA QUE CARECE DEL MISMO, PARA DESCIFRAR LAS PROPIEDADES QUE CONFIERE LA PRESENCIA DE PNAG A LA BACTERIA.     BIOFILMS ARE CONSORTIA OF BACTERIA THAT GROW ATTACHED TO BIOTIC OR ABIOTIC SURFACES AND SURROUNDED BY AN EXTRACELLULAR MATRIX THAT THEY THEMSELVES HAVE SYNTHESISED. COMMUNITY LIFE HAS MANY ADVANTAGES OVER SINGLE-CELL LIFE: IT FACILITATES WATER RETENTION BY PROVIDING A HYDRATED ENVIRONMENT, ALLOWS TO CONCENTRATE NUTRIENTS AND EXTRACELLULAR DEGRADATIVE ENZYMES, ACTS AS A PROTECTIVE BARRIER AGAINST THE ACTION OF PREDATORS, ANTIMICROBIAL SUBSTANCES OR THE IMMUNE SYSTEM AND FACILITATES THE EXCHANGE OF GENETIC MATERIAL. MANY OF THESE PROPERTIES DEPEND ON THE MATRIX SURROUNDING THE COMMUNITY OF BACTERIA. THE MOST COMMON AND IMPORTANT COMPONENT OF THE BIOFILM MATRIX IS EXOPOLISACARIDS. DIFFERENT EXOPOLISACARIDS ARE USED TO FORM THE BIOFILM MATRIX (CELLULOSE, ALGINATE, COLANIC ACID), BUT SURPRISINGLY A WIDE VARIETY OF PHYLOGENETICALLY DISTANT BACTERIA RESORT TO THE SAME EXOPOLISACARIDO, -1,6-N-ACETYLGLUCOSAMINE (PNAG), FOR THIS PURPOSE. THIS OBSERVATION SUGGESTS THAT THE MATRIX FORMED BY PNAG CONFERS SPECIAL ADVANTAGES OVER OTHER TYPES OF MATRICES, AND UNDERLINES THE NEED TO STUDY THIS EXOPOLISACARIDO IN DEPTH. IN THIS PROJECT, WE PROPOSE TO USE THE BACTERIUM PATOGENA STAPHYLOCOCCUS AUREUS AS A MODEL TO UNDERSTAND THE SINGULARITIES THAT PNAG HAS FOR THE FORMATION OF THE BIOFILM. FOR THIS, WE PROPOSE TO STUDY THE MOLECULAR MECHANISMS THAT REGULATE THE SYNTHESIS AND ACTIVATION OF PNAG SYNTHASE, THE MECHANISM OF SIGNAL TRANSDUCTION THAT CONNECTS THE ENVIRONMENTAL CONDITIONS WITH THE PRODUCTION OF PNAG AND THE NUMBER AND LOCATION OF PNAG SYNTHASE IN THE MEMBRANE OF THE BACTERIA. IN ADDITION, WE WILL USE THE HETEROLOGOUS EXPRESSION OF PNAG IN SALMONELLA ENTERITIDIS, A BACTERIUM THAT LACKS IT, TO DECIPHER THE PROPERTIES THAT THE PRESENCE OF PNAG CONFERS ON THE BACTERIA. FINALLY, WE WILL DESIGN AND VALIDATE THE USE OF PNAG TO INDUCE THE FORMATION OF MIXED BIOFILM FOR BIOTECHNOLOGICAL APPLICATIONS. (English)
