SE TRATA DE COMPUESTOS USADOS EN PRODUCTOS DE CUIDADO PERSONAL (PCPS), INCLUYENDO FRAGANCIAS SINTETICAS, FILTROS SOLARES, ANTIMICROBIANOS Y NUEVOS PLASTIFICANTES Y RETARDANTES IGNIFUGOS. EL PROYECTO CONLLEVA UNA PRIMERA ETAPA DE PRIORIZACION EN BASE A MUESTREOS DE BIOTA, FASE DISUELTA Y PARTICULADA EN EL AREA DE ESTUDIO SELECCIONADA (BAHIA DE CADIZ). PARA ELLO SE UTILIZARA UNA COMBINACION DE MUESTREOS PUNTUALES DE AGUA Y SEDIMENTO, SISTEMAS DE MUESTREADORES PASIVOS PREVIAMENTE DESARROLLADOS POR EL GRUPO DE INVESTIGACION, Y EXPOSICION EN JAULAS DE ORGANISMOS FILTRADORES (ALMEJA JAPONESA, RUDITAPES PHILIPPINARUM). TRAS LA FINALIZACION DE ESTA ETAPA Y PROCESADO DE LA INFORMACION OBTENIDA SE SELECCIONARAN LOS 5-6 COMPUESTOS MAS RELEVANTES CON EL FIN DE EVALUAR SU RIESGO AMBIENTAL UTILIZANDO DOS ESPECIES MARINAS DE INTERES COMERCIAL (R. PHILIPPINARUM Y DORADA, SPARUS AURATA) EN ENSAYOS CONTROLADOS DE LABORATORIO USANDO SISTEMAS DE FLUJO CONTINUO. PARA ELLO, ADEMAS DE DETERMINAR LOS FACTORES DE BIOCONCENTRACION DE ESTOS CONTAMINANTES EN LOS ORGANISMOS EXPUESTOS, SE APLICARAN TECNICAS Y HERRAMIENTAS INNOVADORAS (OMICAS Y BIOINFORMATICAS) CON LAS QUE IDENTIFICAR EFECTOS BIOLOGICOS Y ATRIBUIR MODOS DE ACCION (MOAS) ESENCIALES PARA LOS GRUPOS DE COMPUESTOS QUIMICOS SELECCIONADOS. MAS ESPECIFICAMENTE, SE PRETENDE REUNIR UNA MASA CRITICA DE DATOS TRANSCRIPTOMICOS, PROTEOMICOS Y METABOLOMICOS ENCAMINADA A LA COMPRENSION DE LAS VIAS DE TOXICIDAD DE PCPS. LA INTEGRACION DE DATOS DE NIVELES DE CONCENTRACION, BIOACUMULACION Y EFECTOS MOLECULARES MEDIANTE METODOS BIO Y QUIMIOINFORMATICOS SOFISTICADOS PERMITIRA EL DESARROLLO DE NUEVAS ESTRATEGIAS DE EVALUACION DEL RIESGO AMBIENTAL (ERAS) BASADAS EN LA MODELIZACION Y PREDICCION DE TOXICIDADES PARA OTROS COMPUESTOS NO ESTUDIADOS DE CARACTERISTICAS ESTRUCTURALES SIMILARES. DE ESTA FORMA SE PRETENDE REDUCIR EN UN FUTURO CONFLICTOS PRACTICOS Y ETICOS ACERCA DE LA EXPERIMENTACION IN VIVO, TAL COMO SE DESTACA EN DIFERENTES INFORMES EUROPEOS E INTERNACIONALES (ECETOC, OECD, US EPA) QUE HAN LLEGADO A LA CONCLUSION DE QUE LA FORMA TRADICIONAL DE EVALUACION DE IMPACTO AMBIENTAL ES INEFICIENTE E INVIABLE PARA EL CONJUNTO TOTAL DE COMPUESTOS QUIMICOS EXISTENTES Y AUN POR DESARROLLAR. FOR THIS PURPOSE, A COMBINATION OF WATER AND SEDIMENT SAMPLING POINTS, PASSIVE SAMPLER SYSTEMS PREVIOUSLY DEVELOPED BY THE RESEARCH GROUP, AND EXPOSURE IN CAGES OF FILTERING ORGANISMS (JAPANESE CLAM, RUDITAPES PHILIPPINARUM) WILL BE USED. AFTER THE COMPLETION OF THIS STAGE AND PROCESSING OF THE INFORMATION OBTAINED, THE 5-6 MOST RELEVANT COMPOUNDS WILL BE SELECTED IN ORDER TO ASSESS THEIR ENVIRONMENTAL RISK USING TWO MARINE SPECIES OF COMMERCIAL INTEREST (R. PHILIPPINARUM AND GOLDEN, SPARUS AURATA) IN CONTROLLED LABORATORY TRIALS USING CONTINUOUS FLOW SYSTEMS. TO THIS END, IN ADDITION TO DETERMINING THE BIOCONCENTRATION FACTORS OF THESE POLLUTANTS IN THE EXPOSED ORGANISMS, INNOVATIVE TECHNIQUES AND TOOLS (OMICA AND BIOINFORMATICS) WILL BE APPLIED TO IDENTIFY BIOLOGICAL EFFECTS AND ATTRIBUTE MODES OF ACTION (MOAS) ESSENTIAL FOR THE GROUPS OF CHEMICAL COMPOUNDS SELECTED. MORE SPECIFICALLY, IT IS INTENDED TO GATHER A CRITICAL MASS OF TRANSCRIPTOMIC, PROTEOMIC AND METABOLOMIC DATA AIMED AT UNDERSTANDING PCPS TOXICITY PATHWAYS. THE INTEGRATION OF DATA ON CONCENTRATION LEVELS, BIOACCUMULATION AND MOLECULAR EFFECTS USING SOPHISTICATED BIO AND QUIMIOINFORMATION METHODS WILL ALLOW THE DEVELOPMENT OF NEW ENVIRONMENTAL RISK ASSESSMENT STRATEGIES (ERA) BASED ON THE MODELLING AND PREDICTION OF TOXICITIES FOR OTHER COMPOUNDS NOT STUDIED OF SIMILAR STRUCTURAL CHARACTERISTICS. IN THIS WAY IT IS INTENDED TO REDUCE IN A FUTURE PRACTICAL AND ETHICAL CONFLICTS ABOUT IN VIVO EXPERIMENTATION, AS HIGHLIGHTED IN DIFFERENT EUROPEAN AND INTERNATIONAL REPORTS (ECETOC, OECD, US EPA) THAT HAVE REACHED THE CONCLUSION THAT THE TRADITIONAL FORM OF ENVIRONMENTAL IMPACT ASSESSMENT IS INEFFICIENT AND UNFEASIBLE FOR THE TOTAL SET OF EXISTING CHEMICAL COMPOUNDS AND STILL TO BE DEVELOPED.
