Objetivos: (1) Desde una perspectiva poblacional, analizar el N-glicoma sérico en adultos y sus posibles variaciones en función de variables demográficas, hábitos tóxicos y de estilo de vida y enfermedades prevalentes en la población. El estudio tiene como finalidad, además de profundizar en el conocimiento mecanístico de enfermedades frecuentes, muchas de ellas de base inflamatoria (inflamación crónica de bajo grado), el establecimiento de biomarcadores en esas enfermedades. (2) Estudio específico de la glicación de algunas proteínas séricas, especialmente las inmunoglobulinas y concretamente la inmunoglobulina E (IgE) en relación con consumo de alcohol, enfermedad alérgica o infección por helmintos, así como sus diferencias en esas entidades y su correlación con aspectos clínicos. Metodología: (1) El proyecto se implementa dentro de un estudio basado en un muestreo aleatorio de la población general adulta de un municipio (n=2000), realizado por el mismo grupo con vistas a investigación multi-objetivo en marcadores de enfermedades inflamatorias frecuentes en la población. (2) Análisis del los N-glicanos liberados tras acción enzimática (análisis del N-glicoma) o de glicopéptidos tras digestión con endoproteinasas (estudio de la glicosilación de la IgE) mediante HPLC y espectrometría de masas (MALDI-TOF). Objectives: (1) From a population perspective, analyse serum N-glycoma in adults and its possible variations according to demographic variables, toxic habits and lifestyle and diseases prevalent in the population. The study aims, in addition to deepening the mechanistic knowledge of frequent diseases, many of them of inflammatory basis (low-grade chronic inflammation), the establishment of biomarkers in these diseases. (2) Specific study of the glycation of certain serum proteins, especially immunoglobulins and specifically immunoglobulin E (IgE) in relation to alcohol consumption, allergic disease or helminth infection, as well as their differences in these entities and their correlation with clinical aspects. Methodology: (1) The project is implemented as part of a study based on random sampling of the general adult population of a municipality (n=2000), carried out by the same group with a view to multi-objective research into markers of frequent inflammatory diseases in the population. (2) Analysis of N-glycans released after enzymatic action (N-glycoma analysis) or glycopeptides after digestion with endoproteinases (IgE glycosylation study) using HPLC and mass spectrometry (MALDI-TOF). Objectifs: (1) D’un point de vue de la population, analyser le N-glycome sérique chez l’adulte et ses variations possibles en fonction des variables démographiques, des habitudes toxiques, du mode de vie et des maladies prédominantes dans la population. L’étude vise, en plus d’approfondir les connaissances mécanistes sur les maladies fréquentes, beaucoup d’entre elles de base inflammatoire (inflammation chronique de faible grade), l’établissement de biomarqueurs dans ces maladies. Ziele: (1) Aus Bevölkerungssicht analysieren Sie Serum-N-Glykom bei Erwachsenen und seine möglichen Variationen nach demografischen Variablen, toxischen Gewohnheiten und Lebensweise und Krankheiten, die in der Bevölkerung vorherrschen. Die Studie zielt neben der Vertiefung des mechanistischen Wissens über häufige Krankheiten, viele von ihnen von entzündlicher Basis (geringgradige chronische Entzündung), die Etablierung von Biomarkern bei diesen Krankheiten. (2) Spezifische Untersuchung der Glykation bestimmter Serumproteine, insbesondere Immunglobulinen und insbesondere Immunglobulin E (IgE) in Bezug auf Alkoholkonsum, allergische Erkrankung oder Helminth-Infektion, sowie deren Unterschiede in diesen Einheiten und ihre Korrelation mit klinischen Aspekten. Methodik: (1) Das Projekt wird im Rahmen einer Studie durchgeführt, die auf Zufallsstichproben der allgemeinen erwachsenen Bevölkerung einer Gemeinde (n=2000) beruht, die von derselben Gruppe im Hinblick auf die multiobjektive Erforschung von Markern häufig entzündlicher Erkrankungen in der Bevölkerung durchgeführt wird. Doelstellingen: (1) Vanuit het oogpunt van de populatie, analyseer serum N-glycoma bij volwassenen en de mogelijke variaties op basis van demografische variabelen, toxische gewoonten en levensstijl en ziekten die in de populatie voorkomen. De studie is gericht op het verdiepen van de mechanistische kennis van frequente ziekten, veel van hen van ontstekingsbasis (low-grade chronische ontsteking), de oprichting van biomarkers in deze ziekten. (2) Specifieke studie naar de glycatie van bepaalde serumproteïnen, met name immunoglobulinen en specifiek immunoglobuline E (IgE) in verband met alcoholconsumptie, allergische ziekte of helminthinfectie, alsmede hun verschillen in deze entiteiten en hun correlatie met klinische aspecten.
