LAS ENFERMEDADES DE ORIGEN VIRAL COMO LA SHARKA (PLUM POX VIRUS, PPV) Y LA VIRUELA (APPLE CHLOROTIC LEAF SPOT VIRUS, ACLSV), SUPONEN EN ESTOS MOMENTOS EL MAYOR FACTOR LIMITANTE PARA EL CULTIVO DE ALBARICOQUERO EN ESPAÑA Y EN LA REGION DE MURCIA, LA PRINCIPAL REGION PRODUCTORA A NIVEL NACIONAL CON ALREDEDOR DEL 65% DEL TOTAL. EL DESARROLLO DE NUEVAS VARIEDADES RESISTENTES A ESTOS VIRUS ES LA SOLUCION DEFINITIVA AL PROBLEMA. SI BIEN EN EL CASO DE LA RESISTENCIA A PPV NUESTRO GRUPO HA OBTENIDO NUEVAS VARIEDADES RESISTENTES, EN EL CASO DE ACLSV SOLO SE CONOCEN DOS FUENTES DE RESISTENCIA, IDENTIFICADAS RECIENTEMENTE POR NUESTRO GRUPO. NINGUNA DE ESTAS VARIEDADES ES RESISTENTE A LAS DOS VIROSIS. AUNQUE SE HAN REALIZADO DIVERSOS ESTUDIOS SOBRE DIFERENTES ASPECTOS DE ACLSV Y SU RELACION CON LA ENFERMEDAD DENOMINADA VIRUELA DEL ALBARICOQUERO, NUMEROSAS INCOGNITAS PERMANECEN SIN CLARIFICAR EN TORNO A LAS BASES GENETICAS Y MOLECULARES DE LA RESISTENCIA. EN ESTOS MOMENTOS DESPUES DE LA AMPLIA EXPANSION DE LAS NUEVAS VARIEDADES DE ALBARICOQUERO DEL CEBAS-CSIC RESISTENTES A PPV, ES NECESARIA LA INTRODUCCION DE NUEVAS VARIEDADES RESISTENTES TAMBIEN A ACLSV. ESTE PROYECTO TIENE PUES COMO OBJETIVO EL ESTUDIO DE LAS BASES GENETICAS Y MOLECULARES DE LA INTRODUCCION DE RESISTENCIA MULTIPLE A PPV Y ACLSV EN ALBARICOQUERO. SE ANALIZARAN LA TRANSMISION DE LA RESISTENCIA A ACLSV, LA UTILIZACION DE GENOTIPOS HOMOZIGOTICOS RESISTENTES A PPV Y LA INTEGRACION DE GENES DE RESISTENCIA A PPV Y ACLSV. POR OTRO LADO DESDE UN PUNTO DE VISTA MOLECULAR SE IDENTIFICARAN LOS QTLS LIGADOS A LA RESISTENCIA A ACLSV Y LA INTEGRACION DE QTLS LIGADOS A LA RESISTENCIA A ACLSV Y PPV. FINALMENTE ESTOS RESULTADOS GENOMICOS SE VALIDARAN A NIVEL TRANSCRIPTOMICO MEDIANTE RNA-SEQ Y QPCR PARA PROFUNDIZAR EN LA EXPRESION GENICA DE LA RESISTENCIA A ACLSV EN ALBARICOQUERO. DEBEMOS DESTACAR QUE LA CONSECUCION DE ESTOS OBJETIVOS SERIA IMPOSIBLE DE ABORDAR EN EL PERIODO DEL PROYECTO (3 AÑOS) SI NO CONTARAMOS CON EL MATERIAL VEGETAL NECESARIO. PARA TAL FIN, DISPONEMOS DE VARIEDADES RESISTENTES A ACLSV, RESISTENTES A PPV, DIVERSOS CRUZAMIENTOS MAS UNA POBLACION DE 130 DESCENDIENTES PARA ESTUDIAR LA HERENCIA DE LA RESISTENCIA A ACLSV, INCLUYENDO SU MAPA DE LIGAMIENTO CON 90 MARCADORES POSICIONADOS, ASI COMO CRUZAMIENTOS ENTRE VARIEDADES RESISTENTES A PPV Y A ACLSV. A TODO ESTO DEBEMOS AÑADIR LA DISPONIBILIDAD DE LAS HERRAMIENTAS Y TECNICAS NECESARIAS QUE FACILITARAN LA CONSECUCION DE LOS OBJETIVOS PREVISTOS. (Spanish)

DISEASES OF VIRAL ORIGIN SUCH AS SHARKA (PLUM POX VIRUS, PPV) AND SMALLPOX (APPLE CHLOROTIC LEAF SPOT VIRUS, ACLSV) ARE CURRENTLY THE BIGGEST LIMITING FACTOR FOR APRICOT CULTIVATION IN SPAIN AND IN THE REGION OF MURCIA, THE MAIN PRODUCING REGION NATIONWIDE WITH AROUND 65 % OF THE TOTAL. THE DEVELOPMENT OF NEW VARIETIES RESISTANT TO THESE VIRUSES IS THE DEFINITIVE SOLUTION TO THE PROBLEM. ALTHOUGH IN THE CASE OF RESISTANCE TO PPV OUR GROUP HAS OBTAINED NEW RESISTANT VARIETIES, IN THE CASE OF ACLSV ONLY TWO SOURCES OF RESISTANCE ARE KNOWN, RECENTLY IDENTIFIED BY OUR GROUP. NEITHER OF THESE VARIETIES IS RESISTANT TO BOTH VIROSIS. ALTHOUGH SEVERAL STUDIES HAVE BEEN CARRIED OUT ON DIFFERENT ASPECTS OF ACLSV AND ITS RELATION TO THE DISEASE CALLED APRICOT POX, NUMEROUS INCOGNITES REMAIN UNCLARIFIED AROUND THE GENETIC AND MOLECULAR BASES OF RESISTANCE. NOW AFTER THE WIDE EXPANSION OF THE NEW APRICOT VARIETIES OF CEBAS-CSIC RESISTANT TO PPV, IT IS NECESSARY TO INTRODUCE NEW VARIETIES RESISTANT TO ACLSV AS WELL. THIS PROJECT THEREFORE AIMS TO STUDY THE GENETIC AND MOLECULAR BASES OF THE INTRODUCTION OF MULTIPLE RESISTANCE TO PPV AND ACLSV IN APRICOT. THE TRANSMISSION OF RESISTANCE TO ACLSV, THE USE OF PPV-RESISTANT HOMOZIGOTIC GENOTYPES AND THE INTEGRATION OF PPV AND ACLSV RESISTANCE GENES WILL BE ANALYSED. ON THE OTHER HAND, FROM A MOLECULAR POINT OF VIEW, THE QTLS LINKED TO THE RESISTANCE TO ACLSV AND THE INTEGRATION OF QTLS LINKED TO THE RESISTANCE TO ACLSV AND PPV WILL BE IDENTIFIED. FINALLY, THESE GENOMIC RESULTS WILL BE VALIDATED AT THE TRANSCRIPTOMIC LEVEL USING RNA-SEQ AND QPCR TO DEEPEN THE GENIC EXPRESSION OF RESISTANCE TO ACLSV IN APRICOT. WE MUST EMPHASISE THAT THE ACHIEVEMENT OF THESE OBJECTIVES WOULD BE IMPOSSIBLE TO ADDRESS IN THE PERIOD OF THE PROJECT (3 YEARS) IF WE DID NOT HAVE THE NECESSARY PLANT MATERIAL. TO THIS END, WE HAVE VARIETIES RESISTANT TO ACLSV, RESISTANT TO PPV, VARIOUS CROSSES PLUS A POPULATION OF 130 DESCENDANTS TO STUDY THE HERITAGE OF RESISTANCE TO ACLSV, INCLUDING ITS BINDING MAP WITH 90 MARKERS POSITIONED, AS WELL AS CROSSES BETWEEN VARIETIES RESISTANT TO PPV AND ACLSV. WE MUST ADD TO ALL THIS THE AVAILABILITY OF THE NECESSARY TOOLS AND TECHNIQUES TO FACILITATE THE ACHIEVEMENT OF THE INTENDED OBJECTIVES. (English)
