EL LIMONENO ADEMAS DE TENER GRAN USO COMO AROMA EN ALIMENTACION (SABOR A LIMON) POSEE ACTIVIDAD ANTICANCERIGENA, ANTIINFLAMATORIA E INMUNOMODULADORA. BIOMASS, APART FROM BEING JUST A RENEWABLE SOURCE OF ENERGY, REPRESENTS AN IMPORTANT CARBON SOURCE FOR THE PRODUCTION OF A WIDE RANGE OF PRODUCTS WITH HIGH ADDED VALUE THROUGH A PROCESS OF BIOREFINERY. AMONG THOSE CHEMICAL COMPOUNDS HAVING A GROWING INTEREST, TERPENOIDS ARE SECONDARY METABOLITES WITH GREAT INDUSTRIAL APPLICATION AS AROMAS IN FOOD, COSMETICS AND PHARMACY, AND SOME OF THEM HAVE IMPORTANT BIOLOGICAL ACTIVITIES. ONE OF THE MOST IMPORTANT TERPENOIDS IS LIMONENE, WHICH ALSO SERVES AS A PRECURSOR FOR THE PRODUCTION OF MANY OTHER_x000D_ DERIVATIVES OF INDUSTRIAL INTEREST. LIMONENE, IN ADDITION TO ITS BROAD USE AS AN AROMA IN FOOD (LEMON FLAVOR), HAS ANTICANCER, ANTIINFLAMMATORY AND IMMUNOMODULATORY ACTIVITY. OTHER TERPENOIDS, SUCH AS LIMONENE 1,2 EPOXIDE, CAN BE PRODUCED FROM LIMONENE, WHICH IS ALSO WIDELY USED AS AROMA AND FRAGRANCE AND COULD HAVE PHARMACOLOGICAL APPLICATIONS DUE TO ITS ANXIOLYTIC EFFECT IN MICE. OTHER IMPORTANT TERPENOIDS ARE THE XANTHOPHYLLS ZEAXANTHIN AND ASTAXANTHIN. ZEAXANTHIN IS A YELLOW ALCOHOL IMPORTANT FOR VISION, AS IT EXERTS A PROTECTIVE ACTION AGAINST VARIOUS EYE DISEASES. FOR ITS PART, ASTAXANTHIN IS A RED-ORANGE PIGMENT WITH STRONG ANTIOXIDANT PROPERTIES AND IS USED AS A SUPPLEMENT IN FEED._x000D_ THE GROWING DEMAND FOR NATURAL PRODUCTS HAS CONSIDERABLY INCREASED THE INTEREST IN THE PRODUCTION OF THESE COMPOUNDS THROUGH FERMENTATION PROCESSES, WHICH USUALLY REQUIRES GENETIC ENGINEERING PROCESSES TO ACHIEVE ADEQUATE YIELDS FOR INDUSTRIAL PURPOSES. THE OBJECTIVE OF THIS SUBPROJECT IS THE VALORIZATION OF AGRICULTURAL WASTES (MAIZE STRAW AND BEET PULP) OBTAINED FROM THE LEADING MAIZE AND BEET PRODUCER GEOGRAPHIC AREA (LEON AND NEIGHBORING AREAS) BY MEANS OF THEIR BIOTRANSFORMATION IN NATURAL TERPENOIDS OF INTEREST. TO THIS END, THE USE OF BIOTECHNOLOGICAL PLATFORMS CONSISTING OF GENETICALLY ENGINEERED MICROORGANISMS IS PROPOSED. THE BACTERIA STREPTOMYCES LIVIDANS WILL BE MODIFIED FOR THE PRODUCTION OF LIMONENE AND LIMONENE 1,2 EPOXIDE BY MEANS OF PROGRESSIVE OVEREXPRESSION OF THE GENE ENCODING 4S-LIMONENE SYNTHASE FROM MENTHA SPICATA AND THE GENE ENCODING LIMONENE 1,2 MONOOXYGENASE (LIMB) FROM RHODOCOCCUS ERYTHROPOLIS DCL14. THE YEAST XANTHOPHYLLOMYCES DENDRORHOUS WILL BE ENGINEERED TO IMPROVE ASTAXANTHIN PRODUCTION THROUGH THE OVEREXPRESSION OF THE ORTHOLOG GENES ENCODING PHYTOENE SYNTHASE (CRTYB GENE) AND ASTAXANTHIN SYNTHASE (CRTS GENE). ON THE OTHER HAND, IN ORDER TO OBTAIN ZEAXANTHIN-OVERPRODUCING MUTANTS, A MODIFIED STRAIN OF X. DENDRORHOUS, WHICH ACCUMULATES THE PRECURSOR TERPENOID B-CAROTENE, WILL BE ENGINEERED BY MEANS OF THE OVEREXPRESSION OF SEVERAL ORTHOLOG BIOSYNTHETIC GENES AND THE GENE ENCODING B-CAROTENE C3-HYDROXYLASE (CRTZ GENE) FROM BREVUNDIMONAS SP. SD212. ONCE THESE BIOTECHNOLOGICAL PLATFORMS HAVE BEEN OBTAINED, OPTIMIZATION OF THE PRODUCTION PROCESS IS PROPOSED SO AS TO OBTAIN ENOUGH TITERS FOR FURTHER RECOVERY AND PURIFICATION STEPS. THESE NATURAL TERPENOIDS WILL REPRESENT THE BUILDING BLOCKS FOR THE SUBSEQUENT POLYMERIZATION, A PROCESS THAT WILL BE CARRIED OUT BY THE RESEARCH TEAM OF SUBPROJECT 1. (English)
