POR OTRO LADO EXISTEN EVIDENCIAS QUE LIGAN LA PRESENCIA DE POLI-P A LA CAPACIDAD DE UNION DE METALES PESADOS EN MICROORGANISMOS Y LA ADMINISTRACION ORAL DE LACTOBACILLUS PLANTARUM, UNA ESPECIE POLI-P+ CON CAPACIDAD DE ABSORCION DE METALES, DISMINUYE LA TOXICIDAD DE LA INGESTION AGUDA DE CADMIO Y PLOMO EN MODELOS ANIMALES. LA INVESTIGACION DE NUESTRO EQUIPO SOBRE LA SINTESIS Y ACUMULACION DE POLI-P EN BACTERIAS LACTICAS HA EVIDENCIADO QUE UN NUMERO ELEVADO DE ESPECIES DE LACTOBACILLUS SON CAPACES DE ACUMULARLO. NUESTROS RESULTADOS TAMBIEN MUESTRAN QUE CEPAS DE LACTOBACILLUS CASEI DEFICIENTES EN LA SINTESIS DE POLI-P SON MAS SENSIBLES AL MERCURIO. POR TANTO TODO APUNTA A QUE EL POLI-P SINTETIZADO POR LACTOBACILLUS ES UNA MOLECULA CON CAPACIDAD FUNCIONAL EN DOS ASPECTOS DIFERENTES: SALUD INTESTINAL Y DETOXIFICACION DE METALES. EL FIN DE LA PRESENTE PROPUESTA ES DETERMINAR EL POTENCIAL DEL POLI-P PRODUCIDO POR LACTOBACILLUS EN APLICACIONES DE PROBIOTICOS EN ALIMENTACION FUNCIONAL. CON ESTE FIN SE HAN PROPUESTO LOS SIGUIENTES OBJETIVOS ESPECIFICOS:_x000D_ _x000D_ 1. DETERMINAR LA PRODUCCION DE POLI-P Y SU GRADO DE POLIMERIZACION, TANTO DEL POLI-P INTRACELULAR COMO DEL ACUMULADO EN EL MEDIO DE CULTIVO, DE UNA COLECCION DE CEPAS DE LACTOBACILLUS._x000D_ _x000D_ 2. UTILIZAR L. CASEI Y L. PLANTARUM COMO MODELOS PARA EL ESTUDIO DEL PAPEL FISIOLOGICO DEL POLI-P MEDIANTE LA CONSTRUCCION DE MUTANTES DEFICIENTES EN EL GEN DE LA POLIFOSFATO QUINASA (PPK), EL ENZIMA RESPONSABLE DE LA SINTESIS DE POLI-P Y CEPAS QUE SOBREEXPRESEN PPK Y ESTUDIAR SU EFECTO EN LA ACUMULACION DE POLI-P._x000D_ _x000D_ 3. DETERMINAR EL EFECTO PROTECTOR DEL POLI-P FRENTE A ESTIMULOS INFLAMATORIOS EN UN MODELO DE CULTIVO DE CELULAS EPITELIALES INTESTINALES, UTILIZANDO TANTO LAS CEPAS DE LACTOBACILLUS CON MEJOR PRODUCCION DE POLI-P COMO LOS DOS MODELOS ANTERIORES, INCLUYENDO SUS MUTANTES, CON EL FIN DE CORRELACIONAR EL EFECTO OBSERVADO CON EL CONTENIDO EN POLI-P._x000D_ _x000D_ 4. DETERMINAR LOS NIVELES DE TOXICIDAD DE METALES EN LAS CEPAS DE LACTOBACILLUS SELECCIONADAS Y SUS DERIVADAS MODIFICADAS Y LA CAPACIDAD DE UNION (ADSORCION Y TRANSPORTE INTRACELULAR) UTILIZANDO EL MERCURIO COMO MODELO. SE CORRELACIONARAN LOS DATOS OBTENIDOS CON LOS CONTENIDOS DE POLI-P. EVENTUALMENTE SE REALIZARAN PRUEBAS CON OTROS METALES (CADMIO Y PLOMO) PARA DETERMINAR EL POTENCIAL DEL POLI-P EN SU DETOXIFICACION._x000D_ _x000D_ MEDIANTE ESTOS OBJETIVOS SE PRETENDE VALIDAR Y CUANTIFICAR LA ACCION DEL POLI-P COMO UNA MOLECULA EFECTORA DE PROBIOTICOS. A SU VEZ SE CREARIAN LAS BASES PARA LA SELECCION DE AGENTES CON POTENCIAL EN LA DETOXIFICACION DE METALES A NIVEL INTESTINAL. (Spanish)

    POLYPHOSPHATE (POLY-P) WHICH IS EXCRETED BY A PROBIOTICA STRAIN OF LACTOBACILLUS BREVIS HAS BEEN DESCRIBED AS AN EFFECTOR MOLECULA WITH THE ABILITY TO REGULATE ASPECTS OF INTESTINAL HOMEOSTASIS. ON THE OTHER HAND, THERE IS EVIDENCE LINKING THE PRESENCE OF POLY-P TO THE CAPACITY OF UNION OF HEAVY METALS IN MICROORGANISMS AND ORAL ADMINISTRATION OF LACTOBACILLUS PLANTARUM, A POLY-P+ SPECIES WITH METAL ABSORPTION CAPACITY, DECREASES THE TOXICITY OF ACUTE CADMIUM AND LEAD INTAKE IN ANIMAL MODELS. OUR TEAM’S RESEARCH ON THE SYNTHESIS AND ACCUMULATION OF POLYP IN LACTICA BACTERIA HAS SHOWN THAT A LARGE NUMBER OF LACTOBACILLUS SPECIES ARE ABLE TO ACCUMULATE IT. OUR RESULTS ALSO SHOW THAT STRAINS OF LACTOBACILLUS CASEI DEFICIENT IN POLYP SYNTHESIS ARE MORE SENSITIVE TO MERCURY. THEREFORE EVERYTHING SUGGESTS THAT THE POLY-P SYNTHESISED BY LACTOBACILLUS IS A MOLECULA WITH FUNCTIONAL CAPACITY IN TWO DIFFERENT ASPECTS: INTESTINAL HEALTH AND METAL DETOXIFICATION. THE PURPOSE OF THIS PROPOSAL IS TO DETERMINE THE POTENTIAL OF POLYP PRODUCED BY LACTOBACILLUS IN PROBIOTIC APPLICATIONS IN FUNCTIONAL FEEDING. The following SPECIFIC OBJECTIVES have been proposed:_x000D_ _x000D_ 1. Determine POLI-P PRODUCTION AND THEIR POLIMERISATION GRADE, INTRAcellular POLI-P AS A CCUMULATED MEASURE IN CULTIVO, OF A COLLECTION OF LACTOBACILLUS CEPAS._x000D_ _x000D_ 2. Use L. CASEI AND L. plantarum as models for the physiologic PAP STUDY OF THE POLI-P physiologic PAPER FOR THE CONSTRUCTION OF DEFICIENT MUTANTS IN THE GEN OF THE CHINASE POLIOSFATE (PPK), THE RESPONSIBLE ENZIMA OF POLI-P SINTESIS AND CEPAS that overexpress PPK and establish its effectiveness in the accumulation of POLI-P._x000D_ _x000D_ 3. Determine the protective effect of POLI-P FENT to INFLAMATORY STIMULES in a CULTIVE MODEL OF INTESTINAL EPITELIAL CELLS, Using as much as Lactobacillus CEPAS WITH BEST POLI-P PRODUCTION AS THE TWO ANTERIAL MODELS, including your MUTANTS, with the end of correlating the observed EFFECT with the content in POLI-P._x000D_ _x000D_ 4. DETERMINE THE LEVELS OF METAL TOXICITY IN THE SELECTED LACTOBACILLUS STRAINS AND THEIR MODIFIED DERIVATIVES AND THE CAPACITY OF UNION (INTRACELLULAR ADSORPTION AND TRANSPORT) USING MERCURY AS A MODEL. The DATA OBTAINED WITH POLI-P CONTENTS will be correlated. EVENTUALLY TESTS WITH OTHER METALS (CADMIO AND PLOMO) will be carried out to deter the POLI-P POLI-P POLI-P POTOXIFICATION POTENTIAL IN YOUR DETOXIFICATION._x000D_ _x000D_ MEDIANTING these OBJECTIVES ASSESSMENT OF THE POLI-P ACTION AND QUANTIFICATE THE POLI-P’s ACTION as a PROBOSIOTIC-Effective Molecula. IN TURN, THE BASES FOR THE SELECTION OF AGENTS WITH POTENTIAL IN THE DETOXIFICATION OF METALS AT THE INTESTINAL LEVEL WILL BE CREATED. (English)
