SE HA OBSERVADO QUE LAS INTERACCIONES ENTRE LOS VIRUS Y LA PLANTA HUESPED SIGUEN A MENUDO IMPORTANTES CAMBIOS A LO LARGO DEL TIEMPO, Y EL NIVEL DE DAÑO CAUSADO SE PUEDE MODULAR HASTA INCLUSO INVERTIRSE DESDE UN SINERGISMO INICIAL CON EXACERBACION DE SINTOMAS A UNA ATENUACION DEL EFECTO COMBINADO EN TIEMPOS TARDIOS. LA HIPOTESIS DE PARTIDA ES QUE ESTA RESPUESTA DINAMICA PREVISIBLEMENTE SEA LA RESULTANTE DE INTERACCIONES A DOS NIVELES: I) INTERNAMENTE, POR LA MODIFICACION DE LAS RESPUESTAS DEFENSIVAS EN LA PLANTA (INCLUYENDO SILENCIAMIENTO GENICO Y RESPUESTAS DEPENDIENTES DE SEÑALIZACION HORMONAL); Y II) EXTERNAMENTE, POR LA ALTERACION DEL METABOLISMO DE LA PLANTA Y SU EFECTO EN LA ACTIVIDAD DE LOS VECTORES COMO ELEMENTOS CLAVES EN LA DISEMINACION ESPACIAL Y PERVIVENCIA TEMPORAL DE LOS VIRUS PATOGENOS. LOS CAMBIOS MENCIONADOS EN LAS INFECCIONES MIXTAS PUEDEN COMPROMETER LA EFECTIVIDAD DE LAS MEDIDAS DE CONTROL APLICABLES CONTRA VIRUS INDIVIDUALES, POR LO QUE SU CONOCIMIENTO PODRIA TENER IMPLICACIONES PRACTICAS EN EL MANEJO DE ENFERMEDADES VIRALES. IT HAS BEEN OBSERVED THAT THE INTERACTIONS BETWEEN THE VIRUS AND THE HOST PLANT OFTEN FOLLOW IMPORTANT CHANGES OVER TIME, AND THE LEVEL OF DAMAGE CAUSED CAN BE MODULATED TO EVEN REVERSE FROM AN INITIAL SYNERGISM WITH EXACERBATION OF SYMPTOMS TO AN ATTENUATION OF THE COMBINED EFFECT IN LATE TIMES. THE PROCESSES RELEVANT TO THIS DYNAMIC RESPONSE HAVE BEEN LITTLE STUDIED, BUT THEY COULD HELP EXPLAIN THE PREVALENCE OF MIXED INFECTIONS IF AS A CONSEQUENCE THEY FAVOR THE SPREAD OF VIROSIS, SO WE PROPOSE TO INCORPORATE THEM INTO THE ONGOING WORK. THE STARTING HYPOTHESIS IS THAT THIS DYNAMIC RESPONSE IS LIKELY TO RESULT FROM INTERACTIONS AT TWO LEVELS: INTERNALLY, BY MODIFYING DEFENSIVE RESPONSES IN THE PLANT (INCLUDING GENIC SILENCING AND HORMONAL SIGNALING-DEPENDENT RESPONSES); AND II) EXTERNALLY, DUE TO THE ALTERATION OF THE METABOLISM OF THE PLANT AND ITS EFFECT ON THE ACTIVITY OF THE VECTORS AS KEY ELEMENTS IN THE SPATIAL DISSEMINATION AND TEMPORAL SURVIVAL OF PATHOGEN VIRUSES. THE CHANGES MENTIONED IN MIXED INFECTIONS MAY COMPROMISE THE EFFECTIVENESS OF THE CONTROL MEASURES APPLICABLE AGAINST INDIVIDUAL VIRUSES, SO THEIR KNOWLEDGE COULD HAVE PRACTICAL IMPLICATIONS FOR THE MANAGEMENT OF VIRAL DISEASES. THE PROJECT ENVISAGES AS GENERAL OBJECTIVES THE EXPLORATION OF CHANGES OVER TIME IN DIFFERENT PATHOSYSTEMS OF MIXED VIRAL INFECTIONS, ASSESSING HOW THEY AFFECT VIRUSES (OBJECTIVE 1), AND THE ASSOCIATED RESPONSES IN HOST PLANTS, INCLUDING THE CONSEQUENCES OF SUCH CHANGES IN VECTOR DISSEMINATION (OBJECTIVE 2). THE COMPARISONS WERE MADE IN PARALLEL IN THE PATHOSYSTEMS WHOSE STUDY HAS ALREADY BEGUN IN THE PREVIOUS PROJECT, FOCUSING ON CUCURBITACEA AND SOLANACEA SUSCEPTIBLE TO CRINIVIRUS, IN COMBINATION WITH POTYVIRUS IN MELON AND WITH BEGOMOVIRUS IN TOMATO, USING WHITE FLIES AND APHIDS AS VECTORS. THE VALIDATION OF THE HYPOTHESES WILL EXTEND TO THE USE OF DIFFERENT VIRUSES AND EXPERIMENTAL HOSTS, INCLUDING THE CASE OF THE VIRAL DISEASE OF THE SWEET POTATO (HOST PLANT CONVOLVULACEA + CRINIVIRUS + POTYVIRUS), WHERE IT IS PROPOSED TO DEEPEN THE MECHANISMS OF REGULATING THE EXPRESSION OF GENES BY PHENOMENES RECENTLY CHARACTERISED BY SLIPPING OF POLYMERASE, AND WHICH GENERATE VARIANTS OF RNA EDITED AND CAPABLE OF TRANSLATING INTO QUIMERICAS PROTEINS IN MORE THAN ONE READING PHASE, AND WHICH HAS BEEN SEEN TO CHANGE IN SITUATION OF COINFECCION. SUB-OBJECTIVES WITH MULTIPARTICULATED VIRUSES SUCH AS CRINIVIRUS ARE ALSO INCLUDED, EXPLORING POSSIBLE CHANGES IN GENIC EXPRESSION (AND ITS ASSOCIATED VIRAL PHENOTYPES) DUE TO ALTERATION OF THE GENOMIC FORMULA IN MIXED INFECTIONS. THE PARTICIPANTS, BELONGING TO TWO INSTITUTIONS, HAVE EXPERIENCE IN THE AFOREMENTIONED PATHOSYSTEMS, AND ARE ALREADY COLLABORATING WITH DIFFERENT APPROACHES AND METHODOLOGIES THAT ADD COMPLEMENTARITY TO THE JOINT WORK. THE EXPECTED RESULTS OF THE PROJECT WILL SERVE TO COMPLETE THE KNOWLEDGE OF MIXED INFECTIONS, AND WILL FOCUS ON UPDATING THE RECOMMENDATIONS FOR CONTROL AND MANAGEMENT OF VIROSIS IN CROPS. (English)
