LA DINAMICA EN EL TIEMPO DE LOS PROCESOS DE IMPULSIVIDAD Y COMPULSIVIDAD EN EL DESARROLLO DE ADICCIONES COMPONEN UN CICLO DE ADICCION A DROGAS DE ABUSO Y AZUCAR QUE DIFERENCIA TRES FASES PROGRESIVAS (KOOB AND LE MOAL, 1997), SIENDO LA INICIAL UNA FASE DE CONSUMO EN ¿ATRACON/INTOXICACION, GUIADO POR IMPULSIVIDAD Y QUE SE CARACTERIZA POR CONSUMO ESPONTANEO EN ATRACON EN EPISODIOS REPETIDOS, USUALMENTE INTERMITENTES, ASOCIADOS A UN ESTADO MOTIVACIONAL POSITIVO. EN GENERAL, LA INVESTIGACION CIENTIFICA EN EL MARCO DEL CICLO DE ADICCION (VOLKOW & KOOB 2010) ) HA CENTRADO SUS ESFUERZOS MAYORITARIAMENTE EN LA CARACTERIZACION NEUROBIOLOGICA DE LOS PROCESOS TARDIOS DE DEPENDENCIA Y EN ESTADOS POST-DEPENDIENTES Y POCO SABEMOS SOBRE LOS MECANISMOS NEUROQUIMICOS QUE SUBYACEN AL CONSUMO INTENSIVO PROPIO DE LAS ETAPAS INICIALES O SOBRE LOS PROCESOS NEUROBIOLOGICOS DE TRANSICION HACIA ESTADOS ADICTIVOS, YA SEA DE DROGAS O DE COMIDA HEDONICA. EN ESTE CONTEXTO, NUESTRA PROPUESTA SE CENTRARA EN EL ESTUDIO DE LAS POTENCIALES RELACIONES BIDIRECCIONALES EXISTENTES ENTRE OREXINAS Y EN EL CONSUMO DE ALCOHOL Y AZUCAR EN LAS FASES INICIALES DE LA ADICCION, DE CONSUMO EN ATRACON, EN ORGANISMOS VULNERABLES. ADICIONALMENTE, EN ESTE CONTEXTO ABORDAMOS EXPLICITAMENTE LA IDENTIFICACION DE ENDOFENOTIPOS CONDUCTUALES Y MOLECULARES DE VULNERABILIDAD A TRAVES DE LA CARACTERIZACION DE SUB-POBLACIONES QUE DE FORMA ESPONTANEA MUESTRAN UN ALTO CONSUMO EN ATRACON DE REFORZADORES ALTAMENTE PREFERIDOS (ALCOHOL Y SACAROSA). PARA EL PROYECTO UTILIZAREMOS EL PROCEDIMIENTO DE DRINKING IN THE DARK (DID), QUE MODELA CONSUMOS EN ATRACON PARA ALCOHOL Y AZUCAR, CON ALTA EFECTIVIDAD EN ANIMALES NO DEPENDIENTES Y SIN NECESIDADES CALORICAS. AÑADIREMOS LA VARIABLE SEXO TRANSVERSALMENTE A LOS ESTUDIOS E INCLUIMOS UNA SERIE QUE ABORDA POR PRIMERA VEZ Y CON CLARO VALOR TRASLACIONAL, EL IMPACTO DE LA EXPOSICION A ENRIQUECIMIENTO AMBIENTAL, COMO TERAPIA PREVENTIVA DEL DESARROLLO DE DESORDENES DEL CONSUMO EN ATRACON EN MODELOS PRE-CLINICOS. THE DYNAMICS OVER TIME OF THE PROCESSES OF IMPULSIVENESS AND COMPULSIVITY IN THE DEVELOPMENT OF ADDICTIONS MAKE UP A CYCLE OF ADDICTION TO DRUGS OF ABUSE AND SUGAR THAT DIFFERENTIATES THREE PROGRESSIVE PHASES (KOOB AND LE MOAL, 1997), THE INITIAL BEING A PHASE OF CONSUMPTION IN ATRACON/INTOXICATION, GUIDED BY IMPULSIVITY AND CHARACTERISED BY SPONTANEOUS CONSUMPTION IN BERTH IN REPEATED EPISODES, USUALLY INTERMITTENT, ASSOCIATED WITH A POSITIVE MOTIVATIONAL STATE. In GENERAL, SCIENTIFIC RESEARCH IN THE COMMITTEE OF THE ADDITION CICLE (Volkow & KOOB 2010)) has CENTRATED YOUR ESFUERZES mostly IN THE neurobiologic CHARACTERISATION OF THE PROCESSING TARDS OF DEPENDENCE AND POST-DEPENDENT STATES AND MAY KNOW ON THE NEUROQUIMIC MECANISMS underlie the INTENSIONAL CONSUMPTION OF THE INICIAL ETAPAS OR ON Neurobiologic PROCESSES OF TRANSICTION HAS ADICTIVE STATES, AND WE ARE OF DROGS or HODICAL COMMITTEE. IN THIS CONTEXT, OUR PROPOSAL WILL FOCUS ON THE STUDY OF THE POTENTIAL BIDIRECTIONAL RELATIONSHIPS BETWEEN OREXINS AND ALCOHOL CONSUMPTION AND SUGAR IN THE EARLY STAGES OF ADDICTION, OF CONSUMPTION IN BERTH, IN VULNERABLE ORGANISMS. ADDITIONALLY, IN THIS CONTEXT, WE EXPLICITLY ADDRESS THE IDENTIFICATION OF BEHAVIORAL AND MOLECULAR ENDOPHENOTYPES OF VULNERABILITY THROUGH THE CHARACTERISATION OF SUB-POPULATIONS THAT SPONTANEOUSLY SHOW A HIGH CONSUMPTION OF HIGHLY PREFERRED BOOSTERS (ALCOHOL AND SUCROSE). FOR THE PROJECT WE WILL USE THE DRINKING IN THE DARK (DID) PROCEDURE, WHICH MODELS CONSUMPTIONS IN BERTH FOR ALCOHOL AND SUGAR, WITH HIGH EFFECTIVENESS IN NON-DEPENDENT ANIMALS AND WITHOUT CALORIC NEEDS. WE WILL ADD THE VARIABLE SEX TRANSVERSALLY TO THE STUDIES AND INCLUDE A SERIES THAT ADDRESSES FOR THE FIRST TIME AND WITH CLEAR TRANSLATIONAL VALUE, THE IMPACT OF EXPOSURE TO ENVIRONMENTAL ENRICHMENT, AS PREVENTIVE THERAPY FOR THE DEVELOPMENT OF DISORDERS OF CONSUMPTION IN ATRACON IN PRE-CLINIC MODELS.
