    La endometriosis es enfermedad crónica y recurrente que afecta a la mujer en edad fértil produciendo dolores pélvicos y siendo la primera causa de infertilidad. La endometriosis representa el problema ginecológico más frecuente, pues afecta al 5-10% de las mujeres en etapa reproductiva. Su etiopatogenia es aún desconocida. La hipótesis básica de este proyecto es que la endometriosis es producida por las células endometriales estromales (EnSC) (Hipótesis 1), células que derivan de las células madres mesenquimales (MSC), cuyo establecimiento en los focos de endometriosis es favorecido por una situación inflamatoria previa (Hip. 4). En los focos, las EnSC proliferan y también derivan en células epiteliales (Hip. 2) y debido a la secreción de quimioquinas, las EnSC atraen a leucocitos, fundamentalmente macrófagos (Hip. 3). Al estar relacionadas con las MSC, las EnSC humanas tienen la capacidad de sobrevivir como trasplantes xenogénicos en ratones no inmunodeprimidos, lo que ha permitido desarrollar un modelo murino para estudiar la formación de endometriomas in vivo (Hip. 5) y el efecto terapéutico de distinta drogas que actúan sobre rutas sobreactivadas en la endometriosis (metilación de ADN, rutas ERK y Akt). Para el desarrollo de este proyecto obtendremos líneas de EnSC de sangre menstrual (EnSC) y de quistes de chocolate de endometrioma ovárico (fEnSC), Se harán estudios fenotípicos y funcionales de ambos tipos de líneas (Objetivo 1). Se estudiará la diferenciación epitelial de las líneas (Obj. 2). Mediante la inoculación de las líneas celulares en ratones no inmunodeprimidos se inducirán focos de endometriosis en los que será estudiada su composición (Obj. 3) y se analizará el efecto in vivo e in vitro de las drogas (Obj , 4). Endometriosis represents the most common gynecological problem, affecting 5-10 % of women in reproductive stages. His ethiopathogeny is still unknown. The basic hypothesis of this project is that endometriosis is produced by endometrial stromal cells (ENSC) (hypothesis 1), cells derived from mesenchymal stem cells (MSC), whose establishment in the foci of endometriosis is favored by a prior inflammatory situation (Hip. 4). In the foci, NSCs proliferate and also derive in epithelial cells (Hip. 2) and due to the secretion of chemokines, ENSC attracts leukocytes, mainly macrophages (Hip. 3). Since they are related to CSM, human CNS have the ability to survive as xenogenic transplants in non-immunodepressed mice, which has allowed the development of a murine model to study the formation of endometriomas in vivo (Hip. 5) and the therapeutic effect of different drugs acting on overactivated routes in endometriosis (DNA methylation, ERK and Akt routes). For the development of this project we will obtain lines of menstrual blood ENSC (ENSC) and ovarian endometrioma chocolate cysts (fEnSC), Phenotypic and functional studies of both types of lines will be performed (Objective 1). Epithelial differentiation of lines will be studied (Obj. 2). By inoculation of cell lines in non-immunodepressed mice, foci of endometriosis will be induced in which their composition will be studied (Obj. 3) and the in vivo and in vitro effect of drugs will be analysed (Obj, 4).
