DURANTE LA INFECCION POR PATOGENOS, LAS CELULAS HUESPED SUFREN UNA REPROGRAMACION QUE PERMITE A LAS PLANTAS RELOCALIZAR RECURSOS DEDICADOS AL CRECIMIENTO PARA SU DEFENSA. DICHO EQUILIBRIO ENTRE DESARROLLO Y DEFENSA ES CRUCIAL PARA EL ENTENDIMIENTO DE LA INTERACCION PLANTA-PATOGENO. LA RELACION ENTRE CRECIMIENTO Y DEFENSA ESTA BIEN ESTUDIADA, PERO LOS MECANISMOS MOLECULARES SUBYACENTES AL IMPACTO DE LA RESPUESTA DE DEFENSA SOBRE EL DESARROLLO SON AMPLIAMENTE DESCONOCIDOS. UNA ESTRATEGIA COMUN EMPLEADA POR LOS PATOGENOS IMPLICA LA PRODUCCION DE PROTEINAS EFECTORAS QUE DESMANTELAN LA VIA DE SEÑALIZACION POR SRNA (ARNS DE PEQUEÑO TAMAÑO, EN SUS SIGLAS INGLESAS). COMO CONSECUENCIA, ESOS EFECTORES PROTEICOS NO SOLO AFECTAN A UN SISTEMA DEFENSIVO, SINO TAMBIEN A PROCESOS DE DESARROLLO AL LIBERAR A LOS TRANSCRITOS DEL HUESPED DE SU REGULACION POR SRNAS. LA AUSENCIA DE SRNA FUNCIONALES, Y EL CONSIGUIENTE AUMENTO DE LOS TRANSCRITOS A LOS QUE REGULAN, PODRIAN FACILITAR LA REPROGRAMACION CELULAR DEL HUESPED PARA PREVENIR EL PROGRESO DE LA INFECCION. DICHA REPROGRAMACION SUELE MANIFESTARSE CON SINTOMAS QUE AFECTAN AL DESARROLLO DEL HUESPED. _x000D_ LOS MIRNAS (MICROARNS, EN SUS SIGLAS INGLESAS), UNA CLASE PARTICULAR DE SRNAS, CONTROLAN DE FORMA PREDOMINANTE LA EXPRESION DE TFS (FACTORES TRANSCRIPCIONALES, EN SUS SIGLAS INGLESAS). LOS TFS DIANAS DE MIRNAS PARTICIPAN EN COMPLEJOS REGULADORES CON UN PAPEL FUNDAMENTAL EN EL CONTROL PROGRAMAS DE DESARROLLO. POR LO TANTO, LOS TFS REGULADOS POR MIRNAS SON CANDIDATOS ESENCIALES PARA EL INICIO DE LA REPROGRAMACION CELULAR DEL HUESPED BAJO EL ATAQUE DE PATOGENOS. EL OBJETIVO PRINCIPAL DE ESTA PROPUESTA ES DESCUBRIR LOS MECANISMOS MOLECULARES SUBYACENTES A LA RELACION ENTRE EL DESARROLLO Y LA DEFENSA VEGETAL. PARTIENDO DE RESULTADOS DE MI INVESTIGACION ACTUAL, PROPONGO LA CARACTERIZACION DEL PAPEL DE TFS REGULADOS POR MIRNAS QUE PARTICIPAN EN EL CONTROL DEL DESARROLLO, EN EL CONTEXTO DE PROCESOS DE DEFENSA VEGETAL. PARA LLEVAR A CABO DICHA INVESTIGACION, SE EMPLEARAN MODERNAS TECNICAS, ENTRE LAS QUE SE ENCUENTRAN TECNICAS DE SECUENCIACION DE ULTIMA GENERACION, JUNTO CON GENETICA, BIOLOGIA CELULAR, BIOLOGIA MOLECULAR Y FISIOLOGIA. DURING PATHOGEN INFECTION, GUEST CELLS UNDERGO A REPROGRAMMING THAT ALLOWS PLANTS TO RELOCATE RESOURCES DEDICATED TO GROWTH FOR THEIR DEFENSE. SUCH A BALANCE BETWEEN DEVELOPMENT AND DEFENSE IS CRUCIAL FOR THE UNDERSTANDING OF PLANT-PATHOGEN INTERACTION. THE RELATIONSHIP BETWEEN GROWTH AND DEFENSE IS WELL STUDIED, BUT THE MOLECULAR MECHANISMS UNDERLYING THE IMPACT OF THE DEFENSE RESPONSE ON DEVELOPMENT ARE WIDELY UNKNOWN. A COMMON STRATEGY EMPLOYED BY PATHOGENS INVOLVES THE PRODUCTION OF EFFECTOR PROTEINS THAT DISMANTLE THE SRNA SIGNALING WAY (SMALL-SIZED RNAS). AS A RESULT, THESE PROTEIN EFFECTORS NOT ONLY AFFECT A DEFENSIVE SYSTEM, BUT ALSO DEVELOPMENTAL PROCESSES BY FREEING THE TRANSCRITES OF THE HOST FROM THEIR REGULATION BY SRNAS. THE ABSENCE OF FUNCTIONAL SRNA, AND THE CONSEQUENT INCREASE IN TRANSCRIPTS TO THOSE THEY REGULATE, COULD FACILITATE THE CELLULAR REPROGRAMMING OF THE HOST TO PREVENT THE PROGRESS OF THE INFECTION. SUCH REPROGRAMMING USUALLY MANIFESTS ITSELF WITH SYMPTOMS THAT AFFECT THE DEVELOPMENT OF THE GUEST. _x000D_ the miRNAs (MICROARNS, IN YOUR ENGLISH SIGLS), A PARTICULAR CLASS OF SRNAs, CONTROLAN DE FORMMA PREDOMINANT THE EXPRESSION OF TFS (transcriptional FACTORS, IN YOUR INGLESS SIGLAS). MIRNAS TARGET TFS PARTICIPATES IN REGULATORY COMPLEXES WITH A KEY ROLE IN CONTROLLING DEVELOPMENT PROGRAMS. THEREFORE, THE TFS REGULATED BY MIRNAS ARE ESSENTIAL CANDIDATES FOR THE START OF CELLULAR REPROGRAMMING OF THE HOST UNDER THE ATTACK OF PATHOGENS. THE MAIN OBJECTIVE OF THIS PROPOSAL IS TO DISCOVER THE MOLECULAR MECHANISMS UNDERLYING THE RELATIONSHIP BETWEEN DEVELOPMENT AND PLANT DEFENSE. BASED ON THE RESULTS OF MY CURRENT RESEARCH, I PROPOSE THE CHARACTERISATION OF THE ROLE OF TFS REGULATED BY MIRNAS INVOLVED IN THE CONTROL OF DEVELOPMENT, IN THE CONTEXT OF PLANT DEFENSE PROCESSES. TO CARRY OUT THIS RESEARCH, MODERN TECHNIQUES WILL BE USED, AMONG WHICH ARE TECHNIQUES OF SEQUENCING OF THE LAST GENERATION, ALONG WITH GENETICA, CELL BIOLOGY, MOLECULAR BIOLOGY AND PHYSIOLOGY.
