Reference_reference_programme_aids:SA.42799 (2015/X), aid_de_de minimis:§ 42 of the Regulation of the Minister of Infrastructure and Development of 10 July 2015 on the granting of financial assistance by the Polish Agency for Enterprise Development in the framework of the OP Smart Growth 2014-2020.The project concerns the reception of an inactivated precursor of isolated eggs from bee eggs and the development of a testing system to allow in vitro testing of selected active substances without an animal model.The work planned in the framework of the project is to:1.Harvesting of the starting material, i.e. honey bee eggs (Apis mellifera).2.Processing and characterisation of the starting material received.3.Enzinmatotino — mechanical insulation of precursor cells, adhesion, proliferation of morphological type similar to fibroblabellike, and optimisation of the conditions of their rearing.4.Morphological characteristics and ultra-structural adhesion cells, and/or fibrobentials, similar.5.A culture with a viral manufacturer for immortisation of the line received.6.The morphological, structural and molecular characterisation of the killed cell line (as defined by the European Collection of Authentic Cell Cultures).7.The banking of the cells obtained at -120 degrees and storage testing of unlethal lines with their viability after deep-freezing time 8.The development of a system using bee cells for in vitro testing of active compounds, to the exclusion of the animal model, will result in the reception of an inactivated, stable cellular line isolated from the bees and a test system based on those cells allowing the active substances to be tested in vitro breeding conditions.The solutions developed will be the first ones commercially available on the world market.     Numer_referencyjny_programu_pomocowego: SA.42799(2015/X), pomoc_de_minimis: §42 rozporządzenia Ministra Infrastruktury i Rozwoju z dnia 10 lipca 2015 r. w sprawie udzielania przez Polską Agencję Rozwoju Przedsiębiorczości pomocy finansowej w ramach Programu Operacyjnego Inteligentny Rozwój 2014–2020. Przedmiotem projektu jest otrzymanie unieśmiertelnionych progenitorowych komórek izolowanych z jaj pszczelich oraz opracowanie systemu badawczego pozwalającego w warunkach hodowli in vitro testować wybrane substancje czynne z pominięciem modelu zwierzęcego. Zaplanowane w ramach projektu prace przewidują: 1. Pozyskanie materiału wyjściowego, tj. jaj pszczoły miodnej (Apis mellifera). 2. Preparatykę i charakterystykę otrzymanego materiału wyjściowego. 3. Enzymatyczno - mechaniczna izolację komórek progenitorowych, adherentnych, proliferujących o morfotypie zbliżonym do fibroblastopodobnego oraz optymalizację warunków ich hodowli. 4. Charakterystykę morfologiczną oraz ultrastrukturalną komórek o charakterze adherentnym, i/lub fibroblasto podobnym. 5. Hodowlę z konstruktem wirusowym służącym do unieśmiertelniania otrzymanej linii. 6. Charakterystykę morfologiczna, ultrastrukturalna oraz molekularna unieśmiertelnionej linii komórkowej (zgodnie z wytycznymi European Collection of Authenticated Cell Cultures). 7. Bankowanie otrzymanych komórek w temperaturze -120 stopni oraz testy przechowalnicze otrzymanych nieśmiertelnych linii wraz z kontrolą ich żywotności po okresie głębokiego mrożenia 8. Opracowanie systemu wykorzystującego komórki pszczele do testowania in vitro związków czynnych z pominięciem modelu zwierzęcego Rezultatem zaplanowanych prac będzie otrzymanie unieśmiertelnionej, stabilnej linii komórkowej izolowanej z jaj pszczelich oraz system badawczy oparty na tych komórkach, pozwalający w warunkach hodowli in vitro testować substancje czynne. Opracowane rozwiązania będą pierwszymi tego typu dostępnymi komercyjnie na rynku światowym. (Polish)
