LA PLASTICIDAD CORTICAL ES UN FENOMENO COMPLEJO QUE SE DA EN DIFERENTES ETAPAS DEL DESARROLLO ASI COMO EN LA VIDA ADULTA LIGADA A FENOMENOS COMO APRENDIZAJE, DAÑO NEURONAL O COMO CONSECUENCIA DE DIFERENTES PATOLOGIAS. EN RELACION CON LA PATOLOGIA, LA PLASTICIDAD NEURONAL SE PROPONE COMO EL MECANISMO BASICO QUE SUBYACE AL FENOMENO DE REORGANIZACION CORTICAL QUE SE HA DESCRITO DESPUES DE UNA LESION MEDULAR. LA REORGANIZACION CORTICAL DESPUES DE LESION MEDULAR SE HA ESTUDIADO BIEN DESDE UNA PERSPECTIVA GLOBAL DEL FENOMENO MEDIANTE TECNICAS NO INVASIVAS COMO LA FMRI, O BIEN MEDIANTE TECNICAS MUY ESPECIFICAS QUE HAN LLEVADO A RESULTADOS DEMASIADO REDUCCIONISTAS COMO EL REGISTRO NEURONAL MUY LOCALIZADO EN CAPAS DE LA CORTEZA CEREBRAL. EN ESTE MARCO DE ESTUDIO, EXISTE POCA CONCORDANCIA ENTRE LOS RESULTADOS EN RELACION A MECANISMOS, LOCALIZACION Y EVOLUCION TEMPORAL DEL PROCESO DE REORGANIZACION CORTICAL DESPUES DE LESION MEDULAR. ESTA FALTA DE CONCORDANCIA SE PUEDE DEBER A QUE EN LOS ESTUDIOS PREVIOS NO SE HA TENIDO EN CUENTA LA DOBLE HETEROGENEIDAD DEL PROCESO DE PLASTICIDAD NEURONAL, ESPACIAL EN LAS CAPAS CORTICALES Y EN DIFERENTES VENTANAS DE TIEMPO._x000D_ NUESTRO GRUPO DE INVESTIGACION HA ABIERTO EN LOS ULTIMOS AÑOS LINEAS DE TRABAJO CUYOS RESULTADOS HAN SIDO CLAVES PARA ENTENDER EL INICIO DE LOS PROCESOS DE CAMBIOS FUNCIONALES QUE SUCEDEN EN LA CORTEZA CEREBRAL Y EL TALAMO SOMATOSENSORIAL INMEDIATAMENTE DESPUES DE UNA LESION MEDULAR (AGUILAR ET AL 2010; YAGUE ET AL 2011; HUMANES-VALERA ET AL 2013; ALONSO-CALVIÑO ET AL 2016). ESTE PROYECTO SE PROPONE COMO LA CONTINUACION DEL ESTUDIO DEL PROCESO INMEDIATO YA DESCRITO, Y REALIZAR EL ESTUDIO TEMPORAL QUE DETERMINA LOS CAMBIOS EN LA EXCITABILIDAD CORTICAL EN DOS MODELOS DIFERENTES DE LESION MEDULAR A NIVEL TORACICO T9-T10 (LESION COMPLETA VS CONTUSION SEVERA). SIENDO UN OBJETIVO DEFINIR EL PERFIL TEMPORAL DE LA EXCITABILIDAD CORTICAL DESDE QUE SUCEDE UNA LESION MEDULAR HASTA QUE SE ESTABILIZA EN EL TIEMPO. ADEMAS, NUESTROS DATOS ACTUALES INDICAN QUE EXISTE UNA HETEROGENEIDAD ESPACIAL QUE SE REFLEJA EN LOS CAMBIOS DE EXCITABILIDAD QUE SUCEDEN EN LAS DIFERENTES CAPAS DE LA CORTEZA CEREBRAL, SIMILAR A OTROS PROCESOS DE PLASTICIDAD. POR LO TANTO, NOS PROPONEMOS COMO SEGUNDO OBJETIVO CARACTERIZAR LOS CAMBIOS NEUROFISIOLOGICOS DE LAS DIFERENTES CAPAS DE LA CORTEZA CEREBRAL SOMATOSENSORIAL EN FUNCION DEL TIPO DE LESION MEDULAR Y DEL PUNTO TEMPORAL EN EL QUE SE REALICE EL ESTUDIO._x000D_ EL PROYECTO SE LLEVARA A CABO MEDIANTE LA EJECUCION DE DOS SUBPROYECTOS QUE PERMITIRAN ABORDAR LOS DOS OBJETIVOS DE FORMA COMPLEMENTARIA. SUPROYECTO 1: DEFINICION DEL PERFIL TEMPORAL DE LA EXCITABILIDAD CORTICAL EN ANIMALES CON LESION CRONICA Y REGISTRO CRONICO DE EEG Y LFP. SUBPROYECTO 2: DEFINICION DEL PERFIL DE CAMBIOS FISIOLOGICOS DE LAS DIFERENTES CAPAS CORTICALES DE LA CORTEZA SOMATOSENSORIAL DESAFERENTADA POR LESION, MEDIANTE EL REGISTRO CON ELECTRODOS MULTICONTACTO QUE ABARCAN TODAS LAS CAPAS EN SENTIDO VERTICAL PARA OBTENER REGISTROS DE LFP Y DE NEURONAS INDIVIDUALES. _x000D_ LOS RESULTADOS SERVIRAN PARA COMPRENDER MEJOR LA HETEROGENEIDAD TEMPORAL Y ESPACIAL DEL PROCESO DE REORGANIZACION CORTICAL DESPUES DE LESION MEDULAR, Y FACILITAR CON ELLO EL DESARROLLO DE TERAPIAS DIRIGIDAS A LA MODULACION DE LA ACTIVIDAD CORTICAL. CORTICAL PLASTICITY IS A COMPLEX PHENOMENON THAT OCCURS AT DIFFERENT STAGES OF DEVELOPMENT AS WELL AS IN ADULT LIFE LINKED TO PHENOMENA SUCH AS LEARNING, NEURONAL DAMAGE OR AS A CONSEQUENCE OF DIFFERENT PATHOLOGIES. IN RELATION TO PATHOLOGY, NEURONAL PLASTICITY IS PROPOSED AS THE BASIC MECHANISM THAT UNDERLIES THE PHENOMENON OF CORTICAL REORGANISATION THAT HAS BEEN DESCRIBED AFTER A SPINAL INJURY. THE CORTICAL REORGANISATION AFTER SPINAL INJURY HAS BEEN STUDIED EITHER FROM A GLOBAL PERSPECTIVE OF PHENOMENE BY MEANS OF NON-INVASIVE TECHNIQUES SUCH AS FMRI, OR BY MEANS OF VERY SPECIFIC TECHNIQUES THAT HAVE LED TO TOO REDUCTIONIST RESULTS SUCH AS THE NEURONAL RECORD VERY LOCALISED IN LAYERS OF THE CEREBRAL CORTEX. IN THIS FRAMEWORK OF STUDY, THERE IS LITTLE AGREEMENT BETWEEN THE RESULTS IN RELATION TO MECHANISMS, LOCATION AND TEMPORAL EVOLUTION OF THE PROCESS OF CORTICAL REORGANISATION AFTER MEDULLARY INJURY. This CONCORDANCE FALTE may not have been taken into account in the anticipated statistics of the double heterogeneity of the NEURONAL PLASTICITY PROCESS, SPACIAL in the cortical capacity and in the Venentan TIME DIFERENTS._x000D_ OUR RESEARCH GROUP IN THE ULTIMES LINE WORKING YEARS WHICH RESULTS BEING KEYS TO BE FROM FUNCTIONAL CHANGE PROCESSES IN THE CEREBRAL CORTEZA AND THE SOMATOSENSORY TALAME IMMEDIATELY AVAILABLE TO A MEDULAR LESION (AGUIL ET by 2010; YAGUE ET AL 2011; HUMANES-VALERA ET AL 2013; ALONSO-CALVIÑO ET AL 2016). THIS PROJECT IS PROPOSED AS THE CONTINUATION OF THE STUDY OF THE IMMEDIATE PROCESS ALREADY DESCRIBED, AND TO CARRY OUT THE TEMPORARY STUDY THAT DETERMINES THE CHANGES IN CORTICAL EXCITABILITY IN TWO DIFFERENT MODELS OF SPINAL INJURY AT THORACIC LEVEL T9-T10 (COMPLETE LESION VS SEVERE CONTUSION). IT IS AN OBJECTIVE TO DEFINE THE TEMPORAL PROFILE OF CORTICAL EXCITABILITY FROM THE OCCURRENCE OF A MEDULLARY INJURY UNTIL IT STABILISES OVER TIME. IN ADDITION, OUR CURRENT DATA INDICATE THAT THERE IS A SPATIAL HETEROGENEITY THAT IS REFLECTED IN THE CHANGES OF EXCITABILITY THAT OCCUR IN THE DIFFERENT LAYERS OF THE CEREBRAL CORTEX, SIMILAR TO OTHER PROCESSES OF PLASTICITY. Therefore, we PROPONE AS SECOND OBJECTIVES to characterise the Neurophysiologic CHANGES OF THE DIFFERENT CAPAS OF CEREBRAL CORTEZA somatosensorial IN FUNCTION OF THE MEDULAR LESION TYPE AND THE TEMPORAL POINT IN THE STUDY OF THE STUDY._x000D_ THE PROJECT WILL BE READY TO CABO by means of the implementation of two sub-projects that will allow the two complementary forms of training to be opened. PROJECT 1: DEFINITION OF THE TEMPORAL PROFILE OF CORTICAL EXCITABILITY IN ANIMALS WITH CHRONIC LESION AND CHRONIC RECORD OF EEG AND LFP. SUB-PROJECT 2: DEFINITION OF THE PROFILE OF PHYSIOLOGICAL CHANGES OF THE DIFFERENT CORTICAL LAYERS OF THE SOMATOSENSORIAL CORTEX DISAFENTED BY INJURY, BY RECORDING WITH MULTICONTACT ELECTRODES COVERING ALL LAYERS VERTICALLY TO OBTAIN RECORDS OF LFP AND INDIVIDUAL NEURONS. _x000D_ the RESULTS shall serve to buy better the temporal and spatial heterogeneity of the CORTICAL REORGANISATION PROCESS DEVELOPMENT OF MEDULAR LESIONS, AND FACILITIT WITH THE DEVELOPMENT OF TERAPIES DIRIGID TO THE MODULATION OF CORTICAL ACTIVITY.
