EL SUBSECTOR DEL PORCINO IBERICO ES UNA INDUSTRIA QUE EN LOS ULTIMOS AÑOS HA EVOLUCIONADO MUCHO TECNICAMENTE CON EL OBJETIVO DE DISMINUIR LOS COSTES DE PRODUCCION Y OBTENER MAYORES RENDIMIENTOS. EN ESTE SENTIDO, LA INSEMINACION ARTIFICIAL (IA) CON SEMEN REFRIGERADO, MAYORITARIA EN PORCINO, ABRE LA POSIBILIDAD DE MEJORAR LA PRODUCTIVIDAD Y RENTABILIDAD DE LAS EXPLOTACIONES, PERO PLANTEA EL RETO DE LA OBTENCION DE DOSIS SEMINALES COMERCIALES DE CERDO IBERICO Y DUROC DE MAYOR CALIDAD A UN MENOR COSTE. LA CALIDAD DE LAS DOSIS SEMINALES DEPENDE DE LA CALIDAD DEL EYACULADO QUE, A SU VEZ, ESTA INFLUIDA POR DIFERENTES FACTORES, INCLUIDOS LOS AMBIENTALES. ASI, ESTA DEMOSTRADO EL EFECTO ESTACIONAL, PRINCIPALMENTE DEBIDO A LAS ALTAS TEMPERATURAS, TANTO EN LA CANTIDAD COMO EN LA CALIDAD DE LOS EYACULADOS, CON UN CONSIGUIENTE DESCENSO EN EL NUMERO DE ESPERMATOZOIDES POR EYACULADO Y EN LA VIABILIDAD Y MOVILIDAD DE LOS MISMOS. ESTO SUPONE UN DESCENSO EN EL NUMERO DE DOSIS SEMINALES OBTENIDAS POR EYACULADO Y POR VERRACO Y UN DESCENSO EN EL POTENCIAL FECUNDANTE DE LOS ESPERMATOZOIDES CON UN CLARO IMPACTO ECONOMICO EN LA INDUSTRIA PRODUCTORA DE DOSIS SEMINALES. ESTE EFECTO SE ACENTUA EN LAS REGIONES TRADICIONALES DE CRIA DEL CERDO IBERICO DONDE LAS TEMPERATURAS EN EL PERIODO ESTIVAL SOBREPASAN LOS 40ºC, SITUACION QUE CABE ESPERAR SE AGRAVE AUN MAS DEBIDO AL EFECTO DEL CAMBIO CLIMATICO. LA TEMPERATURA AMBIENTAL EXCESIVA PRODUCE EN LA CELULA UN ESTRES TERMICO QUE ACTIVA NUMEROSOS MECANISMOS PARA HACER FRENTE A ESTE, ENTRE ELLOS LA EXPRESION DE UNAS PROTEINAS DENOMINADAS PROTEINAS DE CHOQUE TERMICO (HSP) DESCRITAS EN CELULAS SOMATICAS Y GERMINALES, INCLUYENDO ESPERMATOZOIDES DE VERRACO. POR TODO ELLO, NUESTRA HIPOTESIS DE TRABAJO ES QUE LA EXPRESION Y ACTIVIDAD DE LAS HSP EN SEMEN Y ESPERMATOZOIDES DE VERRACO DECAEN DURANTE LAS EPOCAS ESTIVALES INFLUYENDO EN LA CALIDAD FINAL DEL EYACULADO Y REPERCUTIENDO EN LA CALIDAD DE LAS DOSIS SEMINALES PRODUCIDAS A PARTIR DE ESTE. EL OBJETIVO GENERAL DE ESTE PROYECTO ES PALIAR LOS EFECTOS NOCIVOS DEL INCREMENTO ESTACIONAL DE LA TEMPERATURA EN LA PRODUCCION Y CALIDAD DE LAS DOSIS SEMINALES DE CERDOS IBERICOS Y DUROC DESTINADOS A LA IA EN LA RAZA IBERICA. POR ELLO NOS PLANTEAMOS LOS SIGUIENTES OBJETIVOS ESPECIFICOS: 1) ESTUDIAR SI LA PRESENCIA DE LAS HSP EN LA MEMBRANA PLASMATICA DEL ESPERMATOZOIDE DE CERDO PUEDE USARSE COMO UN INDICADOR DE LA CALIDAD SEMINAL EN ESTA ESPECIE. 2) MEJORAR LA CALIDAD ESPERMATICA DE LAS DOSIS SEMINALES REFRIGERADAS MEDIANTE LA ADICION DE PROTEINAS RECOMBINANTES HSP. 3) COMPROBAR LA FERTILIDAD DE LAS DOSIS SEMINALES REFRIGERADAS SUPLEMENTADAS CON HSP MEDIANTE ENSAYOS DE FERTILIZACION IN VITRO (FIV). 4) ESTUDIAR LA CALIDAD DE LOS EMBRIONES PRODUCIDOS POR FIV MEDIANTE EL ANALISIS DE LA EXPRESION DE GENES ESPECIFICOS. IN THIS SENSE, ARTIFICIAL INSEMINATION (AI) WITH CHILLED SEMEN, MOSTLY IN PIGS, OPENS THE POSSIBILITY OF IMPROVING THE PRODUCTIVITY AND PROFITABILITY OF FARMS, BUT POSES THE CHALLENGE OF OBTAINING COMMERCIAL SEMINAL DOSES OF IBERICO AND DUROC PIGS OF HIGHER QUALITY AT A LOWER COST. THE QUALITY OF THE SEMINAL DOSES DEPENDS ON THE QUALITY OF THE EJACULATE WHICH, IN TURN, IS INFLUENCED BY DIFFERENT FACTORS, INCLUDING ENVIRONMENTAL FACTORS. THUS, THE SEASONAL EFFECT IS DEMONSTRATED, MAINLY DUE TO THE HIGH TEMPERATURES, BOTH IN THE QUANTITY AND QUALITY OF THE EJACULATED, WITH A CONSEQUENT DECREASE IN THE NUMBER OF SPERMATOZOA PER EJACULATE AND IN THE VIABILITY AND MOBILITY OF THE SAME. THIS IMPLIES A DECREASE IN THE NUMBER OF SEMINAL DOSES OBTAINED BY EJACULATE AND BOAR AND A DECREASE IN THE FECUNDANT POTENTIAL OF SPERM WITH A CLEAR ECONOMIC IMPACT ON THE SEMINAL DOSE-PRODUCING INDUSTRY. THIS EFFECT IS ACCENTUATED IN THE TRADITIONAL BREEDING REGIONS OF THE IBERIAN PIG WHERE TEMPERATURES IN THE SUMMER PERIOD EXCEED 40 °C, A SITUATION THAT CAN BE EXPECTED TO WORSEN EVEN MORE DUE TO THE EFFECT OF CLIMATIC CHANGE. THE EXCESSIVE AMBIENT TEMPERATURE PRODUCES IN THE CELL A THERMAL STRESS THAT ACTIVATES NUMEROUS MECHANISMS TO COPE WITH IT, INCLUDING THE EXPRESSION OF PROTEINS CALLED THERMAL SHOCK PROTEINS (HSPS) DESCRIBED IN SOMATIC AND GERM CELLS, INCLUDING BOAR SPERM. THEREFORE, OUR WORKING HYPOTHESIS IS THAT THE EXPRESSION AND ACTIVITY OF THE HSP IN SEMEN AND SPERMATOZOA OF VERRACO FALL DURING THE SUMMER SEASONS INFLUENCING THE FINAL QUALITY OF THE EJACULATE AND HAVING AN IMPACT ON THE QUALITY OF THE SEMINAL DOSES PRODUCED FROM IT. THE GENERAL OBJECTIVE OF THIS PROJECT IS TO MITIGATE THE HARMFUL EFFECTS OF SEASONAL TEMPERATURE INCREASE IN THE PRODUCTION AND QUALITY OF SEMINAL DOSES OF IBERIAN AND DUROC PIGS INTENDED FOR AI IN THE IBERICA BREED. THAT IS WHY WE SET THE FOLLOWING SPECIFIC OBJECTIVES: 1) TO STUDY WHETHER THE PRESENCE OF PSCS IN THE PLASMA MEMBRANE OF PORK SPERM CAN BE USED AS AN INDICATOR OF SEMINAL QUALITY IN THIS SPECIES. 2) IMPROVE THE SPERMATIC QUALITY OF CHILLED SEMINAL DOSES BY ADDING HSP RECOMBINANT PROTEINS. 3) CHECK THE FERTILITY OF THE CHILLED SEMINAL DOSES SUPPLEMENTED WITH HSP BY MEANS OF IN VITRO FERTILISATION (IVF) ASSAYS. 4) TO STUDY THE QUALITY OF EMBRYOS PRODUCED BY IVF BY ANALYSING THE EXPRESSION OF SPECIFIC GENES.
