EN EL PRESENTE PROYECTO NOS PROPONEMOS UTILIZAR LA NANOTECNOLOGIA COMO UNA HERRAMIENTA INNOVADORA EN ESPERMATOLOGIA, UTILIZANDO NANOEMULSIONES QUE ACOMODAN LA VITAMINA E EN SU INTERIOR PROTEGIENDOLA DE LA OXIDACION Y FACILITANDO SU LIBERACION EN EL MEDIO. LOS NANOSISTEMAS PERMITEN LA LIBERACION CONTROLADA DE LAS MOLECULAS ACTIVAS QUE ENCAPSULAN, ALCANZANDO NIVELES ADECUADOS DURANTE PERIODOS PROLONGADOS DE TIEMPO. EXISTEN SOLIDAS EVIDENCIAS QUE MUESTRAN EL POTENCIAL DE LOS NANOSISTEMAS VESICULARES PARA PROMOVER EL TRANSPORTE CELULAR DE ANTIOXIDANTES. POR ELLO ES ESPERABLE QUE ESTOS SISTEMAS FACILITEN LA TERAPIA CON ANTIOXIDANTES A OTRO TIPO DE CELULAS, COMO SON LOS ESPERMATOZOIDES. LAS RAZONES QUE SUSTENTAN DICHA HIPOTESIS SERIA: I) MAYOR SOLUBILIDAD DEL ANTIOXIDANTE EN UN SISTEMA INOCUO PARA LAS CELULAS, II) MAYOR PROTECCION FRENTE A CONDICIONES DE INESTABILIDAD, III) LIBERACION CONTROLADA DEL ANTIOXIDANTE AL MEDIO EVITANDO ELEVADAS CONCENTRACIONES PROOXIDANTES. EL ANTIOXIDANTE SELECCIONADO SERA LA VITAMINA E EN PRIMER LUGAR POR SER UN POTENTE AGENTE ANTIOXIDANTE PARA LOS ESPERMATOZOIDES. ADEMAS, LA VITAMINA E PUEDE SER EL CONSTITUYENTE PRINCIPAL DE NANOSISTEMAS VESICULARES COMO LAS NANOEMULSIONES, FORMANDO EL NUCLEO OLEOSO DE LA MISMA. POR UN LADO, LA LIBERACION CONTROLADA DE LA VITAMINA E PERMITIRA LA MODULACION DE LOS RADICALES LIBRES DE OXIGENO (ROS), MINIMIZANDO LOS EFECTOS DELETEREOS DEL ESTRES OXIDATIVO COMO LA ALTERACION DE LA INTEGRIDAD DEL ADN Y LA LIPOPEROXIDACION DE MEMBRANAS, QUE LIMITA EL POTENCIAL FECUNDANTE DE ESTAS CELULAS. POR OTRO LADO, LA LIBERACION CONTROLADA PERMITIRA QUE LOS ROS DESEMPEÑEN SU PAPEL FUNDAMENTAL EN LA FISIOLOGIA ESPERMATICA, SIENDO NECESARIOS PARA PROCESOS TAN IMPORTANTES COMO EL DESARROLLO DEL PATRON DE HIPERACTIVACION DE LA MOTILIDAD DURANTE LA CAPACITACION. _x000D_ PARA ELLO NOS PROPONEMOS DESARROLLAR LOS SIGUIENTES OBJETIVOS ESPECIFICOS:_x000D_ 1. EXAMINAR LA INFLUENCIA DE LAS NANOEMULSIONES SOBRE LA VIABILIDAD ESPERMATICA._x000D_ 2. EVALUAR EL EFECTO DE DIFERENTES CONCENTRACIONES DE VITAMINA E FORMULADAS COMO NANOESTRUCTURAS DE LIBERACION CONTROLADA, EN PRESENCIA O AUSENCIA DE AGENTES OXIDATIVOS EXOGENOS, SOBRE LA FISIOLOGIA ESPERMATICA._x000D_ 3. ESTUDIAR EL EFECTO PROTECTOR DE LA VITAMINA E ENCAPSULADA EN EL TRANSPORTE/INCUBACION DE MUESTRAS ESPERMATICAS A DIFERENTES TEMPERATURAS._x000D_ 4. EVALUAR EL EFECTO DE DIFERENTES CONCENTRACIONES DE VITAMINA E FORMULADAS EN NANOEMULSIONES, EN PRESENCIA O AUSENCIA DE AGENTE OXIDATIVO EXOGENO, SOBRE LA RESPUESTA DE LOS ESPERMATOZOIDES DESCONGELADOS AL ESTRES OXIDATIVO. IN THE PRESENT PROJECT WE PROPOSE NANOTECHNOLOGY AS AN INNOVATIVE TOOL IN SPERMATOLOGY USING NANOEMULSIONS THAT ACCOMMODATE THE VITAMIN E IN ITS INTERIOR, PROTECTING IT FROM OXIDATION AND FACILITATING ITS RELEASE IN THE MEDIUM. NANOSYSTEMS ALLOW THE CONTROLLED RELEASE OF THE ACTIVE MOLECULES THAT ENCAPSULATE, REACHING ADEQUATE LEVELS FOR PROLONGED PERIODS OF TIME. THERE IS STRONG EVIDENCE SHOWING THE POTENTIAL OF VESICULAR NANOSYSTEMS TO PROMOTE CELLULAR TRANSPORT OF ANTIOXIDANTS. IT IS THEREFORE EXPECTED THAT THESE SYSTEMS FACILITATE THERAPY WITH ANTIOXIDANTS TO OTHER TYPES OF CELLS, SUCH AS SPERMATOZOA. THE REASONS FOR THIS HYPOTHESIS WOULD BE: I) GREATER ANTIOXIDANT SOLUBILITY IN A CELL-SAFE SYSTEM, II) BETTER PROTECTION AGAINST UNSTABLE CONDITIONS, III) CONTROLLED RELEASE OF THE ANTIOXIDANT INTO THE MEDIUM AVOIDING HIGH PRO-OXIDANT CONCENTRATIONS. THE ANTIOXIDANT SELECTED WILL BE VITAMIN E PRIMARILY BECAUSE IT IS A POTENT ANTIOXIDANT AGENT FOR SPERM. IN ADDITION, VITAMIN E MAY BE THE MAIN CONSTITUENT OF VESICULAR NANOSYSTEMS SUCH AS NANOEMULSIONS, FORMING THE OILY NUCLEUS THEREOF. ON THE ONE HAND, CONTROLLED RELEASE OF VITAMIN E WILL ALLOW MODULATION OF OXYGEN FREE RADICALS (ROS), MINIMIZING THE DELETERIOUS EFFECTS OF OXIDATIVE STRESS SUCH AS ALTERED DNA INTEGRITY AND MEMBRANE LIPOPEROXIDATION, WHICH LIMITS THE POTENTIAL FOR FERTILIZATION OF THESE CELLS. ON THE OTHER HAND, CONTROLLED RELEASE WILL ALLOW THE ROS TO PLAY THEIR FUNDAMENTAL ROLE IN SPERM PHYSIOLOGY, BEING NECESSARY FOR PROCESSES AS IMPORTANT AS THE DEVELOPMENT OF THE PATTERN OF HYPERACTIVATION OF MOTILITY DURING CAPACITATION._x000D_ FOR THIS PURPOSE WE AIM TO DEVELOP THE FOLLOWING SPECIFIC OBJECTIVES:_x000D_ 1. TO EVALUATE THE EFFECT OF DIFFERENT CONCENTRATIONS OF VITAMIN E FORMULATED IN NANOEMULSIONS IN THE PRESENCE OR ABSENCE OF EXOGENOUS OXIDATIVE AGENTS ON THE RESPONSE OF THAWED SPERMATOZOA TO OXIDATIVE STRESS._x000D_ 2. EVALUATE THE EFFECT OF DIFFERENT CONCENTRATIONS OF VITAMIN E FORMULATED AS CONTROLLED RELEASE NANOSTRUCTURES, IN THE PRESENCE OR ABSENCE OF EXOGENOUS OXIDATIVE AGENTS ON SPERM PHYSIOLOGY._x000D_ 3. STUDY THE PROTECTIVE EFFECT OF ENCAPSULATED VITAMIN E IN THE TRANSPORT / INCUBATION OF SPERM SAMPLES AT DIFFERENT TEMPERATURES._x000D_ 4. EVALUATE THE EFFECT OF DIFFERENT CONCENTRATIONS OF VITAMIN E FORMULATED IN NANOEMULSIONS IN THE PRESENCE OR ABSENCE OF EXOGENOUS OXIDATIVE AGENTS ON THE RESPONSE OF THAWED SPERMATOZOA TO OXIDATIVE STRESS.
