LAS PATOLOGIAS METABOLICAS CRONICAS ASOCIADOS CON LA OBESIDAD Y LOS TRASTORNOS MENTALES, COMO LA DEPRESION, REPRESENTAN LA MAYOR CAUSA DE MORBILIDAD, DISCAPACIDAD Y MORTALIDAD TEMPRANA EN TODO EL MUNDO. SIN EMBARGO, HASTA AHORA, SE HAN HECHO ESCASOS AVANCES EN EL DESARROLLO DE INTERVENCIONES EFICACES Y MEDIDAS PREVENTIVAS. ESTAS ENFERMEDADES SON ALTAMENTE COMORBIDAS Y MULTIFACTORIALES Y, PROBABLEMENTE, IMPLICAN FACTORES DE RIESGO Y MECANISMOS FISIOPATOLOGICOS COMUNES, QUE PODRIAN RELACIONARSE CON LA MICROBIOTA INTESTINAL Y SUS ALTERACIONES (DISBIOSIS). EN ESTE CONTEXTO, LA IDENTIFICACION DE FACTORES CAUSALES O QUE CONTRIBUYEN A ESTOS TRASTORNOS Y, ESPECIALMENTE, AQUELLOS QUE SON SUSCEPTIBLES DE SER MODIFICADOS POR LA DIETA Y EL ESTILO DE VIDA, COMO LA MICROBIOTA Y SUS FUNCIONES, PODRIA PERMITIR EL DESARROLLO DE MEDIDAS MAS EFICACES PARA REDUCIR SU IMPACTO SOCIAL Y ECONOMICO. LOS OBJETIVOS GENERALES DE LA PRESENTE PROPUESTA SON: (I) AVANZAR EN EL CONOCIMIENTO DE LA FUNCION QUE DESEMPEÑA LA MICROBIOTA INTESTINAL Y SUS INTERACCIONES CON FACTORES AMBIENTALES, COMO LA DIETA Y EL ESTRES, EN LA REGULACION DE LAS VIAS QUE CONTROLAN LA HOMEOSTASIS ENERGETICA Y LOS MECANISMOS DE RECOMPENSA TRAS LA INGESTA; (II) EN LAS ASOCIACIONES ENTRE LA OBESIDAD Y LOS TRASTORNOS METABOLICOS ASOCIADOS Y EMOCIONALES (DEPRESION); E (III) IDENTIFICAR BACTERIAS INTESTINALES ESPECIFICAS (INCLUIDA UNA NUEVA GENERACION DE PROBIOTICOS) Y SUS MODOS DE ACCION DE FORMA QUE PUEDAN CONSTITUIR LA BASE DEL DESARROLLO DE FUTURAS INTERVENCIONES DIETETICAS MAS EFICACES PARA REDUCIR SU INCIDENCIA. CHRONIC METABOLIC DISORDERS ASSOCIATED WITH OBESITY AND MENTAL DISORDERS, SUCH AS DEPRESSION, ACCOUNT FOR THE LARGEST BURDEN OF DISEASE, DISABILITY AND EARLY MORTALITY WORLDWIDE. SO FAR, HOWEVER, LITTLE PROGRESS HAS BEEN MADE ON THE DEVELOPMENT OF EFFECTIVE INTERVENTIONS AND PREVENTIVE MEASURES. THESE DISEASES ARE HIGHLY COMORBID AND MULTIFACTORIAL AND LIKELY INVOLVE COMMON RISK FACTORS AND PATHOPHYSIOLOGICAL MECHANISMS, WHICH COULD CROSSLINK TO THE INTESTINAL MICROBIOTA AND ITS ALTERATIONS (DYSBIOSIS). IN THIS CONTEXT, THE IDENTIFICATION OF CAUSAL AND CONTRIBUTING FACTORS TO THESE DISORDERS AND, SPECIALLY, THOSE THAT ARE MODIFIABLE BY THE DIET AND LIFESTYLE, LIKE THE MICROBIOTA AND ITS FUNCTIONS, COULD OPEN THE WAY TO DEVELOP COST-EFFECTIVE MEASURES FOR REDUCING THEIR SOCIAL AND ECONOMIC IMPACT. THE GENERAL OBJECTIVES OF THE PRESENT PROPOSAL ARE (I) TO PROGRESS IN THE UNDERSTANDING OF THE ROLE PLAYED BY THE GUT MICROBIOTA AND ITS INTERACTIONS WITH ENVIRONMENTAL FACTORS, SUCH AS DIET AND STRESS, IN THE REGULATION OF THE PATHWAYS CONTROLLING ENERGY HOMEOSTASIS AND THE FOOD REWARD SYSTEMS AND (II) IN THE LINKS BETWEEN OBESITY AND THE ASSOCIATED METABOLIC AND MOOD DISORDERS (DEPRESSION); AND (III) TO IDENTIFY SPECIFIC INTESTINAL BACTERIA (INCLUDING THE SO-CALLED NEXT GENERATION OF PROBIOTICS) AND PRODUCTS THEREOF THAT COULD CONTRIBUTE TO DEVELOPING REFINED MICROBIOME-BASED DIETARY INTERVENTIONS THAT HELP TO REDUCE THE INCIDENCE OF THESE DISORDERS.
