The POLYPEPPHARM Project aims to develop 3 supergenic polypeptide drugs, namely Interferon α2a (INFα2a), Erythropoietin α (EPOa), and Interleukin 2 (IL-2), using methyltrophic fungi that add polysaccharides to excreted proteins. The recombinant proteins that will be synthesised will be subjected to an antigenicity and immunogenicity test, an analysis of their structure and stability, a biochemical characterisation and a series of functionality tests, and eventually integrated into advanced pharmacoforms, i.e. gel-type implants and microspheres type implants. It is expected that the developed polypeptide drugs will have improved characteristics compared to the corresponding, commercially available medicines mainly due to better stability and solubility, specific activity, and bioavailability. Finally, techno-economic and market research will be carried out to identify the best strategy for exploiting the results. Το Έργο POLYPEPPHARM στοχεύει στην ανάπτυξη 3 υπεργενόσημων πολυπεπτιδικών φαρμάκων, συγκεκριμένα την Ιντερφερόνη α2a (INFα2a), την Ερυθροποιητίνη α (EPOα), και την Ιντερλευκίνη 2 (IL-2), χρησιμοποιώντας μεθυλοτροφικούς μύκητες που προσθέτουν πολυσακχαρίτες στις εκκρινόμενες πρωτεΐνες. Οι ανασυνδυασμένες πρωτεΐνες που θα συντεθούν θα υποβληθούν σε έλεγχο αντιγονικότητας και ανοσογονικότητας, σε ανάλυση της δομής και της σταθερότητάς τους, σε βιοχημικό χαρακτηρισμό και σε μια σειρά τεστ λειτουργικότητας και, τελικά, θα ενσωματωθούν σε προηγμένες φαρμακομορφές, δηλαδή σε εμφυτεύματα τύπου πηκτής και σε εμφυτεύματα τύπου μικροσφαιριδίων. Είναι αναμενόμενο ότι τα πολυπεπτιδικά φάρμακα που θα αναπτυχθούν θα έχουν βελτιωμένα χαρακτηριστικά σε σχέση με τα αντίστοιχα, εμπορικά διαθέσιμα φάρμακα κυρίως λόγω της καλύτερης σταθερότητας και διαλυτότητας, της ειδικής ενεργότητας, και της βιοδιαθεσιμότητας. Τέλος, θα διεξαχθεί τεχνοοικονομική μελέτη και έρευνα αγοράς για να προσδιοριστεί η βέλτιστη στρατηγική αξιοποίησης των αποτελεσμάτων. Το Έργο POLYPEPPHARM συνεπάγεται πολύ μικρό ρίσκο και αναμένεται να έχει τεράστιο οικονομικό όφελος. (Greek)
