NUESTRO ESTUDIO ANALIZARA EN ESPERMATOZOIDES DE ANIMALES EXPUESTOS A LOS TOXICOS DURANTE LA VIDA EMBRIONARIA O ADULTA, LOS POTENCIALES DAÑOS EN EL GENOMA, TRANSCRIPTOMA Y EPIGENOMA, IDENTIFICANDO GENES CANDIDATOS DE DAÑO GENOTOXICO Y EPIGENOTOXICO. PARA ELLO SE EMPLEARAN TECNICAS PUESTAS A PUNTO EN NUESTRO LABORATORIO, COLABORANDO CON OTROS EQUIPOS PARA LA REALIZACION DEL RRBS. ESTOS ESTUDIOS NOS PERMITIRAN IDENTIFICAR LAS FASES MAS SENSIBLES Y LOS ELEMENTOS POTENCIALMENTE AFECTADOS POR LOS AGENTES TOXICOS, IDENTIFICANDO LOS GENES O PROMOTORES MAS VULNERABLES Y ESTABLECIENDO UN PANEL DE INDICADORES DE DAÑO EN LA LINEA GERMINAL. _x000D_ EN LA DESCENDENCIA DE LOS PROGENITORES EXPUESTOS SE EVALUARA LA MORFOGENESIS, CON ESPECIAL ATENCION A LA FORMACION DEL CORAZON, LA CAPACIDAD REPRODUCTORA, RESPUESTA INMUNE, CAPACIDAD DE REGENERACION DE TEJIDOS Y TASA DE ENVEJECIMIENTO, UTILIZANDO ESTUDIOS MORFOLOGICOS, FUNCIONALES Y DE EXPRESION GENICA. LA POSIBLE TRANSMISION DE LA INFORMACION PATERNA A LAS CELULAS DE LA LINEA GERMINAL DE LA F1, QUE PROVOCARIAN UNA HERENCIA TRANSGENERACIONAL A LA F2 SE ANALIZARA MEDIANTE EL ANALISIS DE LAS CELULAS DE LA LINEA GERMINAL DE LOS MARCADORES DE TOXICIDAD PREVIAMENTE DEFINIDOS. EN SU CASO, TAMBIEN SERA EVALUADO EL DESARROLLO DE LA F2 UTILIZANDO LOS MISMOS PARAMETROS QUE CON LA F1. SE EVALUARA ADEMAS EL POSIBLE RESCATE NUTRICIONAL DE LOS DAÑOS EPIGENOTOXICOS, QUE PODRIAN DEBERSE A UNA MENOR ACTIVIDAD DE LAS METIL TRANSFERASAS O A UNA MENOR DISPONIBILIDAD DE GRUPOS METILO, MEDIANTE LA ADMINISTRACION DE DIETAS RICAS EN FOLATOS. OUR STUDY WILL ANALYSE POTENTIAL DAMAGE TO THE GENOME, TRANSCRIPTOME, AND EPIGENOME IN ANIMALS EXPOSED TO TOXICS DURING EMBRYONIC OR ADULT LIFE, IDENTIFYING GENES FOR GENOTOXIC AND EPIGENOTOXIC DAMAGE. FOR THIS PURPOSE TECHNIQUES WILL BE USED DEVELOPED IN OUR LABORATORY, COLLABORATING WITH OTHER TEAMS FOR THE REALISATION OF THE RRBS. THESE STUDIES WILL ALLOW US TO IDENTIFY THE MOST SENSITIVE PHASES AND THE ELEMENTS POTENTIALLY AFFECTED BY TOXIC AGENTS, IDENTIFYING THE MOST VULNERABLE GENES OR PROMOTERS AND ESTABLISHING A PANEL OF DAMAGE INDICATORS IN THE GERM LINE. _x000D_ in the DESCENDENCE OF EXPUEST PROGENITORS will be evaluated morphogenesis, with special attention to HEART FORMATION, reproductive capacity, IMMUNE RESPERTY, TEJID REGENERATION CAPACITY AND Aging Tax, using MORFOLOGICAL, FUNCIONAL and GENIC EXPRESSION STUDIES. THE POSSIBLE TRANSMISSION OF PATERNAL INFORMATION TO THE CELLS OF THE GERMINAL LINE OF THE F1, WHICH PROVOCARIAN A TRANSGENERATIONAL INHERITANCE TO F2 WAS ANALYSED BY ANALYSING THE GERMLINE CELLS OF THE PREVIOUSLY DEFINED TOXICITY MARKERS. IF APPLICABLE, THE DEVELOPMENT OF F2 WILL ALSO BE EVALUATED USING THE SAME PARAMETERS AS WITH F1. THE POSSIBLE NUTRITIONAL RESCUE OF EPIGENOTOXIC DAMAGE, WHICH MAY BE DUE TO LOWER METHYL TRANSFERASE ACTIVITY OR REDUCED AVAILABILITY OF METHYL GROUPS, WILL ALSO BE EVALUATED THROUGH THE ADMINISTRATION OF FOLATE-RICH DIETS.
