    Objetivos: Identificar un panel de biomarcadores de genes cuyo patrón coordinado de metilación y/o la metilación de las regiones reguladoras de la expresión de genes específicos (ICR) se asocie con el alteraciones adversas en desarrollo, crecimiento fetal y neurodesarrollo a los 12 meses edad. Así como evaluar la asociación entre la exposición prenatal a plaguicidas organofosforados y neonicotinoides y el patrón de metilación. Metodología: Cohorte de nacimiento prospectiva de 344 binomios madre-hijo/a. Las mujeres se captarán durante el primer trimestre de embarazo y se les efectuará un seguimiento durante el segundo y tercer trimestre de embarazo, el parto y al año del nacimiento del/a hijo/a. Se tomarán muestras de orina durante el embarazo y al niño al año, asimismo se tomaránmuestras de placenta en el parto. Se recogerá información sobre variables relacionadas con desarrollo y crecimiento fetal y se aplicará a los niños/as la prueba Bayley-II de neurodesarrollo a los 12 meses. El ADN será extraído y purificado a partir de lasmuestras de placenta utilizando el QIAamp DNA Mini Kit. El análisis de metilación del genoma completo de células de placenta se realizarán con Infinium® HumanMethylation450 BeadChip de Illumina y de genes específicos por pirosecuenciación. Sedeterminarán los niveles de plaguicidas organofosforados y neonicotinoides en las muestras de orina. Utilizando semisupervised recursively partitioned mixture models (SS-RPMM) se identificará el grupo de genes cuyos patrones de metilación resultan más asociados a las variables de efecto. Se construirán modelos de regresión logística y lineal multivariantes según corresponda, sobre las variables de efectos en relación a los cluster de los patrones de metilación. (Spanish)

    Objectives: Identify a panel of gene biomarkers whose coordinated methylation pattern or methylation of specific gene expression regulatory regions (GRIs) is associated with adverse alterations in development, foetal growth and neurodevelopment at 12 months of age. As well as evaluating the association between prenatal exposure to organophosphorus and neonicotinoid pesticides and the methylation pattern. Methodology: Prospective birth cohort of 344 mother-child binomies. Women will be captured during the first trimester of pregnancy and monitored during the second and third trimesters of pregnancy, delivery and the year of birth of the child. Urine samples will be taken during pregnancy and the child per year, placenta samples will also be taken at birth. Information on variables related to foetal growth and development will be collected and the Bayley-II neurodevelopment test will be applied to children at 12 months. DNA will be extracted and purified from placenta samples using the QIAamp DNA Mini Kit. The methylation analysis of the complete genome of placenta cells will be performed with Illumina’s Infinium® HumanMethylation450 BeadChip and specific genes by pyrossequencing. They will determine the levels of organophosphorus and neonicotinoid pesticides in urine samples. Using semi-supervised Recursively partitioned mixture models (SS-RPMM) will identify the group of genes whose methylation patterns are most associated with effect variables. Multivariate logistic and linear regression models shall be constructed, as appropriate, on the effects variables in relation to the cluster of methylation patterns. (English)
