MORE SPECIFICALLY, IT SEEMS THAT WITH OBESITY DECREASES THE SENSITIVITY TO THE SWEET TASTE AND THE STRENGTHENING POWER OF FOODS CONTAINING SUGARS AND THAT ALL THIS COULD BE MEDIATED BY LEPTIN, THE HORMONE ANOREXINEGIC REGULATING APPETITE. RECENT DATA FROM OUR RESEARCH GROUP SHOW THAT IN AN ANIMAL MODEL OF OBESITY INDUCED BY COFFEE DIET, PHYSICAL EXERCISE DECREASES THE CIRCULATING LEVELS OF LEPTIN, WHICH SUGGESTS THAT PHYSICAL EXERCISE COULD ALSO MODULATE THE RESISTANCE TO THE SWEET TASTE AND THE DIMINUCTION OF THE REINFORCING VALUE OF SWEET FOODS THAT APPEARS IN OBESITY. THIS IS THE STARTING POINT AND THE MAIN SCENARIO OF THIS PROJECT. THE TEAM IS INTERDISCIPLINARY AND SOME OF ITS MEMBERS HAVE WORKED CLOSELY ON ONE OF THE BOOMING STRATEGIES FOR THE TREATMENT OF OBESITY BASED ON THE USE OF NATURALLY OCCURRING BIOACTIVE INGREDIENTS. MORE SPECIFICALLY, WE HAVE DEMONSTRATED THE BENEFICIAL EFFECTS OF POLYPHENOLS AGAINST OBESITY AND RELATED PATHOLOGIES, EVEN ADDRESSING THE PROBLEM OF OBESITY FROM THE OPTIC OF METABOLIC PROGRAMMING. THUS, THE GENERAL OBJECTIVE OF THE PROJECT IS TO DEVELOP AN INTERVENTION BASED ON PHYSICAL EXERCISE, A MILD DIET AND AN INGREDIENT OF NATURAL ORIGIN, OLEUROPEIN, A POLYPHENOL FOUND IN EXTRA VIRGIN OLIVE OIL. WE INTEND TO EVALUATE THE EFFECTS OF A MILD DIET, A MILD DIET+EXERCISE, A MILD DIET+OLEUROPEIN, AND GENTLE DIET+EXERCISE+OLEUROPEIN ADMINISTERED TO AN ANIMAL MODEL OF INDUCED OBESITY. THESE EXPERIMENTAL GROUPS WILL BE COMPARED WITH ANIMALS FED NORMOLIPIDICA DIET AND ANOTHER GROUP FED COFFEE DIET THROUGHOUT THE EXPERIMENT. THIS DESIGN WILL MAKE IT POSSIBLE TO EVALUATE THE EFFECTS OF THE DIFFERENT INTERVENTIONS ON THE SENSITIVITY TO THE SWEET TASTE, THE REINFORCING VALUE OF SWEET FOODS, AND THE REGULATORY FUNCTIONS OF LEPTIN. FOR ALL THIS WE WILL EVALUATE IN MALE AND FEMALE RATS LICKING BEHAVIORS AND PREFERABLY BY SWEET SOLUTIONS OF TWO TYPES, A NATURAL COMPOUND, THE SUCROSE, AND AN ARTIFICIAL NON CALORIC SWEETENER, THE SUCRALOSE. WEIGHT, INTAKE AND BLOOD SAMPLING WILL BE MONITORED TO STUDY THE EVOLUTION OF CLASSICAL MARKERS (GLUCOSE, TRIGLYCERIDES, CHOLESTEROL, INSULIN). IN ADDITION, TO DETERMINE THE ANXIETY LEVELS OF OBESE ANIMALS WE WILL EVALUATE THEIR ANXIOUS BEHAVIOR AND ANALYSE THE HORMONAL RESPONSE OF THE HPA AXIS ONCE THE COFFEE DIET HAS BEEN REMOVED. THUS, WE INTEND TO PROVIDE SOLID SCIENTIFIC BASES AND EVALUATE A MULTIFACETED STRATEGY FOR STABILISING THE TREATMENT OF OBESITY, WHILE STUDYING THE IMPLICATION OF LEPTIN IN ITS EFFECTS. (English)
