Property / summary: MIXED PLANT INFECTIONS BY TWO OR MORE PATHOGENIC VIRUSES ARE VERY COMMON IN NATURAL CONDITIONS. COMPARED TO INDIVIDUAL INFECTIONS, MULTIPLE INFECTIONS WITH MORE THAN ONE VIRUS IN THE SAME PLANT CAN CAUSE SPECIFIC COMPLEX DISEASES, AS IN THE CASE OF SYNERGISTIC REACTIONS KNOWN AMONG SOME VIRAL PATHOGENS. IN OTHER CASES, THE PRESENCE OF MIXED INFECTIONS MAY GO UNNOTICED, AND THIS MAY RESULT IN A REDUCTION IN THE EFFECTIVENESS OF SELECTIVELY APPLIED CONTROL MEASURES TO COMBAT VIRUSES INDIVIDUALLY. IN FACT, EPIDEMIOLOGIC STUDIES ARE REVEALING THE EXISTENCE OF COMPLEX SITUATIONS WITH FREQUENT MIXED INFECTIONS THAT COULD COMPROMISE THE EFFECTIVENESS OF VIROSIS CONTROL MEASURES. TO ADDRESS THIS PROBLEM, WE CONTEMPLATE TWO MAJOR GOALS THAT SEEK TO EXPLORE THE EFFECTS OF MULTIPLE VIRAL INFECTIONS IN: THE VECTOR-BORNE PARAMETERS OF THE INDIVIDUAL VIRUSES, AND II) THE RESISTANCE CHARACTERISTICS OF HOST PLANTS, WITH PARTICULAR REGARD TO THE DIFFERENT ACTIVITIES OF THE VIRAL SUPPRESSORS OF SILENCING AGAINST PLANT DEFENCE REACTIONS. BOTH OBJECTIVES WILL BE STUDIED IN PATHOSYSTEMS THAT INCLUDE DIFFERENT HOST PLANTS (CUCURBITACEOUS AND SOLANACEOUS), POTENTIAL VECTOR ORGANISMS (LEFTS AND WHITE FLIES), AND COMBINATIONS OF CRINIVIRUS WITH IPOMOVIRUS/POTYVIRUS IN MELON, OR WITH BEGOMOVIRUS IN TOMATO. THE CHOICE OF EXPERIMENTAL SYSTEMS IS BASED ON THE CURRENT SITUATION IN CROPS OF OUR ENVIRONMENT, WHILE CONSIDERING THE SCIENTIFIC AND TECHNICAL INTEREST THAT CAN CONTRIBUTE THE ANALYSIS OF THE COMBINATIONS OF VIRUSES IN BOTH HOSTS. THERE ARE ALSO ADDITIONAL SUB-OBJECTIVES FOCUSED ON DESIGNING PROACTIVE MEASURES THAT FAVOR VIRUS CONTROL, INCLUDING PROCEDURES TO GENERATE RNAI RESPONSES IN PLANT-FEEDING INSECTS AS A TECHNOLOGY WITH THE POTENTIAL TO INTERFERE WITH VIRUS-BY-VECTOR TRANSMISSION, AND THE CHARACTERISATION OF VIRAL COUNTERDEFENSE MECHANISMS AGAINST PLANT RESPONSES, IN PARTICULAR THOSE BASED ON THE COMBINED ACTIVITIES OF SILENCING SUPPRESSOR GENES THAT CAN OCCUR DURING MULTIPLE INFECTIONS. AMONG THE EXPECTED RESULTS OF THE PROJECT ARE THE RECOMMENDATIONS FOR THE CONTROL OF THE SPREAD OF THE VIRUS AND FOR THE MANAGEMENT OF RESISTANCE, BASED ON A BETTER UNDERSTANDING OF THE FREQUENT MULTIPLE INFECTIONS. LAS INFECCIONES MIXTAS DE PLANTAS POR DOS O MAS VIRUS PATOGENOS SON MUY COMUNES EN CONDICIONES NATURALES. EN COMPARACION CON LAS INFECCIONES INDIVIDUALES, LAS INFECCIONES MULTIPLES CON MAS DE UN VIRUS EN LA MISMA PLANTA PUEDEN CAUSAR ENFERMEDADES COMPLEJAS ESPECIFICAS, COMO OCURRE EN EL CASO DE LAS REACCIONES SINERGICAS QUE SE CONOCEN ENTRE ALGUNOS PATOGENOS VIRALES. EN OTROS CASOS, LA PRESENCIA DE INFECCIONES MIXTAS PUEDE PASAR INADVERTIDA, Y ESTO PUEDE TENER COMO CONSECUENCIA QUE SE REDUZCA LA EFICACIA DE LAS MEDIDAS DE CONTROL APLICADAS SELECTIVAMENTE PARA COMBATIR A LOS VIRUS DE FORMA INDIVIDUAL. DE HECHO, LOS ESTUDIOS EPIDEMIOLOGICOS ESTAN REVELANDO LA EXISTENCIA DE SITUACIONES COMPLEJAS CON FRECUENTES INFECCIONES MIXTAS QUE PODRIAN COMPROMETER LA EFECTIVIDAD DE LAS MEDIDAS DE CONTROL DE VIROSIS. PARA HACER FRENTE A ESTA PROBLEMATICA, SE CONTEMPLAN DOS GRANDES OBJETIVOS QUE BUSCAN EXPLORAR QUE EFECTOS TIENEN LAS INFECCIONES VIRALES MULTIPLES EN: I) LOS PARAMETROS DE TRANSMISION POR VECTORES DE LOS VIRUS INDIVIDUALES, Y II) LAS CARACTERISTICAS DE RESISTENCIA DE LAS PLANTAS HUESPED, CON ESPECIAL ATENCION A LAS DIFERENTES ACTIVIDADES DE LOS SUPRESORES VIRALES DE SILENCIAMIENTO FRENTE A LAS REACCIONES DE DEFENSA DE LAS PLANTAS. AMBOS OBJETIVOS SE ESTUDIARAN EN PATOSISTEMAS QUE INCLUYEN DIFERENTES PLANTAS HUESPED (CUCURBITACEAS Y SOLANACEAS), POTENCIALES ORGANISMOS VECTORES (PULGONES Y MOSCAS BLANCAS), Y COMBINACIONES DE CRINIVIRUSES CON IPOMOVIRUS/POTYVIRUS EN MELON, O CON BEGOMOVIRUS EN TOMATE. LA ELECCION DE LOS SISTEMAS EXPERIMENTALES SE BASA EN LA SITUACION ACTUAL EN CULTIVOS DE NUESTRO ENTORNO, A LA VEZ QUE SE HA CONSIDERADO EL INTERES CIENTIFICO-TECNICO QUE PUEDE APORTAR EL ANALISIS DE LAS COMBINACIONES DE VIRUS EN AMBOS HUESPEDED. TAMBIEN SE CONTEMPLAN SUBOBJETIVOS ADICIONALES ENFOCADOS A DISEÑAR MEDIDAS PROACTIVAS QUE FAVOREZCAN EL CONTROL DE VIRUS, INCLUYENDO PROCEDIMIENTOS PARA GENERAR RESPUESTAS DE RNAI EN INSECTOS QUE SE ALIMENTAN DE PLANTAS COMO UNA TECNOLOGIA CON POTENCIAL PARA INTERFERIR CON LA TRANSMISION DE VIRUS POR VECTORES, Y LA CARACTERIZACION DE LOS MECANISMOS VIRALES DE CONTRADEFENSA FRENTE A LAS RESPUESTAS DE LAS PLANTAS, EN PARTICULAR LOS BASADOS EN LAS ACTIVIDADES COMBINADAS DE GENES SUPRESORES DE SILENCIAMIENTO QUE PUEDEN DARSE DURANTE LAS INFECCIONES MULTIPLES. ENTRE LOS RESULTADOS ESPERABLES DEL PROYECTO SE PUEDEN MENCIONAR LAS RECOMENDACIONES PARA EL CONTROL DE LA PROPAGACION DEL VIRUS Y PARA EL MANEJO DE RESISTENCIAS, BASADAS EN UN MEJOR CONOCIMIENTO DE LAS FRECUENTES INFECCIONES MULTIPLES.
