    DURANTE EL PROCESO DE DOMESTICACION DEL TOMATE OCURRIO UNA IMPORTANTE PERDIDA DE VARIABILIDAD GENETICA, LO QUE PROVOCO UN ESTRECHAMIENTO MUY NOTABLE DE LA BASE GENETICA. RECIENTES ESTUDIOS SOBRE LA DOMESTICACION LLEVADOS A CABO POR ESTE EQUIPO INVESTIGADOR MEDIANTE GENOTIPADO CON MILES DE MARCADORES HAN PERMITIDO PROFUNDIZAR EN ESTE PROCESO. EN ESTOS ESTUDIOS SE HA MOSTRADO QUE LA DOMESTICACION SUCEDIO EN DOS FASES, UNA EN SUDAMERICA Y OTRA POSTERIOR EN MESOAMERICA. LA PERDIDA DE VARIABILIDAD SUCEDIO DURANTE EL PASO DE SUDAMERICA A MESOAMERICA DE FORMA QUE S. LYCOPERSICUM TODAVIA CONSERVA EN EL AREA ANDINA UNA VARIABILIDAD EQUIPARABLE A LA DE UNA ESPECIE SILVESTRE. EN ESTE PROYECTO SE PRETENDE RECUPERAR PARTE DE ESTA VARIABILIDAD PERDIDA, ANTES DESCONOCIDA Y QUE POR TANTO NO HA SIDO AMPLIAMENTE UTILIZADA EN LA MEJORA DEL TOMATE. PLANTEAMOS LA CONSTRUCCION DE UNA POBLACION GENERADA A PARTIR DE OCHO PARENTALES, QUE REPRESENTAN ESTA VARIABILIDAD ANDINA PERDIDA, CUATRO DE ELLOS DE LA VARIEDAD CERASIFORME DE S. LYCOPERSICUM Y CUATRO DE LA ESPECIE SILVESTRE, S. PIMPINELLIFOLIUM. EL DESARROLLO DE LA POBLACION CONSISTE EN UNA SERIE DE GENERACIONES DE INTERCRUZAMIENTO SEGUIDAS DE AUTOFECUNDACIONES SUCESIVAS CON EL FIN DE FIJAR LOS MATERIALES OBTENIDOS. ESTAS POBLACIONES SON LAS MAS IDONEAS PARA LA CARTOGRAFIA FINA DE GENES Y QTLS, AL PRODUCIRSE VARIAS GENERACIONES DE INTERCRUZAMIENTO QUE FACILITAN LA OCURRENCIA DE MULTIPLES SOBRECRUZAMIENTOS. EN TOMATE EXISTE YA UNA POBLACION DE ESTAS CARACTERISTICAS, PERO LOS PARENTALES DE PARTIDA, TODOS ELLOS DE LA ESPECIE CULTIVADA S. LYCOPERSICUM, SON MUY DISTINTOS MORFOLOGICA Y MOLECULARMENTE A LOS ELEGIDOS PARA ESTE PROYECTO, SEGUN LAS CARACTERIZACIONES MORFOLOGICAS Y EL GENOPTIPADO REALIZADO CON 7000 SNPS Y MAS DE 1000 ENTRADAS, Y NO RECOGEN TODA LA VARIBILIDAD EXISTENTE EN ESTAS ESPECIES. LOS PARENTALES SELECCIONADOS EN ESTE PROYECTO INCLUYEN UNA GRAN VARIABILIDAD MORFOLOGICA, GENETICA Y ECOLOGICA, ENCONTRANDOSE ENTRADAS CULTIVADAS, SEMIDOMESTICADAS Y SILVESTRES. ESTA POBLACION SERVIRA PARA LA CARTOGRAFIA DE GENES IMPLICADOS EN MULTIPLES CARACTERES VEGETATIVOS, DE FLORACION Y DE FRUTO PARA LOS QUE EXISTE UNA ALTA VARIACION EN LOS OCHO PARENTALES SELECCIONADOS. OTRA CARACTERISTICA DE ELEVADO INTERES ES LA ADAPTACION A DISTINTOS TIPOS DE AMBIENTES DE LOS OCHO PARENTALES, DESDE DESIERTOS A SELVAS TROPICALES, DESDE EL NIVEL DEL MAR A 1500 M DE ALTURA Y DESDE EL ECUADOR AL NORTE DE MEXICO. ELLO POSIBILITARA LA DETECCION Y MAPEO DE GENES IMPLICADOS EN LA TOLERANCIA Y ADAPTACION A ESTRESES ABIOTICOS Y AYUDARA A DESARROLLAR MATERIALES PARA HACER FRENTE AL CAMBIO CLIMATICO. AUNQUE SE DISPONE DE DATOS PREVIOS DE FENOTIPADO DE LOS PARENTALES, SE LLEVARA A CABO, COMO SEGUNDO OBJETIVO DEL PROYECTO, UN FENOTIPADO MINUCIOSO DE ESTOS, INCLUYENDO RESISTENCIAS A VIRUS, BACTERIAS Y HONGOS, QUE SERVIRA PARA DETERMINAR LOS CARACTERES A FENOTIPAR EN LA POBLACION QUE SE VA A DESARROLLAR. FINALMENTE, LOS OCHOS GENOTIPOS FUNDACIONALES SERAN SECUENCIADOS Y ALINEADOS CON EL GENOMA DE REFERENCIA, LO QUE PERMITIRA LA LOCALIZACION DE MILES DE SNPS QUE SERVIRAN PARA LA CARTOGRAFIA DE ALTA RESOLUCION DE QTLS IMPLICADOS EN LOS CARACTERES DE INTERES, ASI COMO PARA REALIZAR ESTUDIOS COMPARATIVOS DE SU GENOMA. EL FENOTIPADO Y GENOTIPADO DE LA POBLACION RESULTANTE SERA OBJETO DE OTRO PROYECTO POSTERIOR, YA QUE EL DESARROLLO DE LA POBLACION SEGREGANTE AQUI Y EL GENOTIPADO DE LOS PARENTALES SE CONSIDERA SUFICIENTE TRABAJO PARA ESTE PROYECTO. THE LOSS OF VARIABILITY OCCURRED DURING THE PASSAGE FROM SOUTH AMERICA TO MESOAMERICA SO THAT S. LYCOPERSICUM STILL RETAINS A VARIABILITY COMPARABLE TO THAT OF A WILD SPECIES IN THE ANDEAN AREA. THIS PROJECT AIMS TO RECOVER SOME OF THIS PREVIOUSLY UNKNOWN LOST VARIABILITY THAT HAS NOT BEEN WIDELY USED IN TOMATO IMPROVEMENT. WE PROPOSE THE CONSTRUCTION OF A POPULATION GENERATED FROM EIGHT PARENTS, WHICH REPRESENT THIS LOST ANDEAN VARIABILITY, FOUR OF THEM FROM THE VARIETY CERASIFORME S. LYCOPERSICUM AND FOUR FROM THE WILD SPECIES, S. PIMPINELLIFOLIUM. THE DEVELOPMENT OF THE POPULATION CONSISTS OF A SERIES OF GENERATIONS OF INTERCROSSING FOLLOWED BY SUCCESSIVE SELF-FERTILISATIONS IN ORDER TO FIX THE OBTAINED MATERIALS. THESE POPULATIONS ARE THE MOST IDEAL FOR THE FINE MAPPING OF GENES AND QTLS, WITH SEVERAL GENERATIONS OF INTERCROSSING THAT FACILITATE THE OCCURRENCE OF MULTIPLE CROSSOVERS. IN TOMATO THERE IS ALREADY A POPULATION OF THESE CHARACTERISTICS, BUT THE PARENTS OF DEPARTURE, ALL OF THEM OF THE CULTIVATED SPECIES S. LYCOPERSICUM, ARE VERY DIFFERENT MORPHOLOGICALLY AND MOLECULARLY TO THOSE CHOSEN FOR THIS PROJECT, ACCORDING TO THE MORPHOLOGICAL CHARACTERISATIONS AND THE GENOPTIPADO MADE WITH 7000 SNPS AND MORE THAN 1000 ENTRIES, AND DO NOT COLLECT ALL THE VARIBILITY EXISTING IN THESE SPECIES. THE PARENTS SELECTED IN THIS PROJECT INCLUDE A GREAT MORPHOLOGIC, GENETIC AND ECOLOGICAL VARIABILITY, FINDING CULTIVATED, SEMI-DOMESTICATED AND WILD ENTRANCES. THIS POPULATION WILL SERVE FOR THE MAPPING OF GENES INVOLVED IN MULTIPLE VEGETATIVE, FLOWERING AND FRUIT CHARACTERS FOR WHICH THERE IS A HIGH VARIATION IN THE EIGHT PARENTS SELECTED. ANOTHER FEATURE OF HIGH INTEREST IS THE ADAPTATION TO DIFFERENT TYPES OF ENVIRONMENTS OF THE EIGHT PARENTS, FROM DESERTS TO RAINFORESTS, FROM SEA LEVEL TO 1500 M HIGH AND FROM ECUADOR TO THE NORTH OF MEXICO. THIS WILL ENABLE THE DETECTION AND MAPPING OF GENES INVOLVED IN TOLERANCE AND ADAPTATION TO ABIOTIC STRESS AND HELP DEVELOP MATERIALS TO COPE WITH CLIMATIC CHANGE. ALTHOUGH PREVIOUS DATA ON PHENOTYPING OF PARENTS ARE AVAILABLE, A DETAILED PHENOTYPING OF THESE, INCLUDING RESISTANCE TO VIRUSES, BACTERIA AND FUNGI, WILL BE CARRIED OUT AS A SECOND OBJECTIVE OF THE PROJECT, WHICH WILL SERVE TO DETERMINE THE CHARACTERS TO BE PHENOTYPED IN THE POPULATION TO BE DEVELOPED. FINALLY, THE EIGHT FOUNDING GENOTYPES WILL BE SEQUENCED AND ALIGNED WITH THE REFERENCE GENOME, WHICH WILL ALLOW THE LOCATION OF THOUSANDS OF SNPS THAT WILL SERVE FOR THE HIGH RESOLUTION MAPPING OF QTLS INVOLVED IN THE CHARACTERS OF INTEREST, AS WELL AS TO CARRY OUT COMPARATIVE STUDIES OF THEIR GENOME. THE PHENOTYPING AND GENOTYPING OF THE RESULTING POPULATION WILL BE THE SUBJECT OF ANOTHER SUBSEQUENT PROJECT, SINCE THE DEVELOPMENT OF THE SEGREGATING POPULATION HERE AND THE GENOTYPING OF PARENTS IS CONSIDERED SUFFICIENT WORK FOR THIS PROJECT.
