Las células madre del tejido adiposo (ASCs) intervienen en los procesos de expansibilidad de dicho tejido y en los fenómenos inmunomoduladores que resultan determinantes en la obesidad y sus co-morbilidades. La plasticidad celular de las ASCs puede estar influenciada por metafactores sistémicos modulados por la microbiota intestinal (MI). Los cambios observados en la funcionalidad de estas células en la obesidad y la diabetes pueden persistir latentes a pesar de la reducción de peso (RP). OB GENERAL:Evaluar la reversibilidad de los cambios en la funcionalidad de las ASCs observados en pacientes obesos y con T2D tras la RP. Hallar un patrón predictivo del comportamiento de las ASCs pre- y post-RP, mediante un análisis comparativo integral de los mecanismos moleculares que regulan la funcionalidad de las ASCs y el análisis de elementos modificables relacionados con la MI.OB SP1:1a: Analizar el efecto de la RP sobre las funciones precursoras, inmunoreguladoras y memoria epigenéticade ASCs en obesos mórbidos tras cirugía bariátrica (CB);1b: Identificar y caracterizar la población de ASCs circulantes pre- y post-CB. Evaluar la capacidad de estas ASCs circulantes de reflejar el patrón de metilación de las ASCsdel SAT pre- y post-RP;1c: Identificación de firmas moleculares específicas asociadas a la reversión total o parcial del comportamiento disfuncional de las ASCs de SAT tras la RP;1d: Evaluar los cambios en diversos factores sistémicos y la composición de la MI, pre- y post-CB;1e: Establecer un patrón predictivo integrando todos los datos que pueda determinar aquellos sujetos susceptibles de revertir la disfuncionalidad de las ASCs. METODOLOGÍA:Estudio prospectivo en pacientes obesos mórbidos sometidos a CB. Estudio de variables analíticas. Estudio de permeabilidad y MI. Estudio funcional y de ómicas de las ASCs pre- y post-RP. Find a predictive pattern of the behavior of pre- and post-RP ASCs, through a comprehensive comparative analysis of the molecular mechanisms that regulate the functionality of ASCs and the analysis of modifiable elements related to MI.OB SP1:1a: Analyse the effect of PR on precursor, immunoregulatory and epigenetic memory functions of ASCs in obese morbids after bariatric surgery (CB);1b: Identify and characterise the population of pre- and post-CB circulating ASCs. Assess the ability of these circulating ASCs to reflect the methylation pattern of pre- and post-RP ASCs;1c: Identification of specific molecular signatures associated with the total or partial reversal of dysfunctional behaviour of SAT ASCs after PR;1d: Evaluate changes in various systemic factors and the composition of MI, pre- and post-CB;1e: Establish a predictive pattern by integrating all the data that can determine those subjects likely to reverse the dysfunctionality of the ASCs.
