QUALITY CONTROL OF GAMETES IN THE CULTIVATION OF MARINE FISH REMAINS A KEY CHALLENGE FOR THE SUSTAINABILITY OF EUROPEAN AQUACULTURE. IN MALES, SPERM MOTILITY IS CONSIDERED THE MOST EFFECTIVE PARAMETER FOR ESTIMATING ITS QUALITY (I.E. ABILITY TO FERTILISE), ALTHOUGH MARINE TELEOSTEOS ARE STILL NOT WELL KNOWN TO MANY OF THE KEY MOLECULAR MECHANISMS THAT CONTROL SPERM SCOURGE ACTIVITY. THIS KNOWLEDGE IS HOWEVER ESSENTIAL TO DEVELOP ROBUST AND RELIABLE MOLECULAR BIOMARKERS TO ASSESS SPERM QUALITY, WHICH CAN BE USEFUL FOR ASSESSING THE IMPACT OF GROWING CONDITIONS, GENETIC SELECTION PROGRAMS, AND ARTIFICIAL FERTILISATION AND CRYOPRESERVATION METHODS OF GAMETES. THEREFORE, AT THE INDUSTRIAL LEVEL THERE IS AN IMPORTANT DEMAND FOR INNOVATIVE BASIC RESEARCH INTO THE PHYSIOLOGY OF THE SPERMATICA MOTILITY PROCESS IN MARINE FISH. IN PREVIOUS STUDIES ON GOLDEN SPERM (SPARUS AURATA) WE HAVE DISCOVERED A NEW MITOCHONDRIAL ANTIOXIDANT SYSTEM MEDIATED BY A WATER CHANNEL (AQUAPORINE-8BB, AQP8BB), WHICH FUNCTIONS IN THIS CASE AS A PEROXIPORINE FACILITATING THE EXIT OF H2O2 FROM MITOCHONDRIA AND THE CONTINUOUS PRODUCTION OF ENERGY (ATP) NECESSARY FOR FLAGELLAR MOTILITY. INTRACELLULAR TRANSPORT OF AQP8BB IN SPERM DURING ACTIVATION IN SEAWATER THEREFORE APPEARS AS A CRUCIAL FACTOR IN THE REGULATION OF MITOCHONDRIAL DETOXIFICATION AND MOTILITY. HOWEVER, THE INTRACELLULAR SIGNALING PATHWAYS THAT REGULATE THIS MECHANISM ARE UNKNOWN, ALTHOUGH OUR PRELIMINARY DATA SUGGEST THAT CERTAIN STRESS-ACTIVATED KINASES MAY BE INVOLVED. IN ADDITION TO AQP8BB, GOLD MOTILE SPERM EXPRESS OTHER AQUAPORINS IN THE HEAD AND ANTERIOR TAIL, WHICH SEEM TO CONTROL THE TRAJECTORY AND PATTERN OF SPERM MOVEMENT. THE MOLECULAR MECHANISMS UNDERLYING THE FUNCTION OF THESE CHANNELS HAVE NOT YET BEEN IDENTIFIED, AND WE PROPOSE THE HYPOTHESIS THAT THESE COULD BE ASSOCIATED WITH IONIC CHANNELS TO ACTIVATE LOCAL OSMOTIC RESPONSES AND ION TRANSPORT MECHANISMS, WHICH CAN MAINTAIN AND/OR DIRECT SPERM NAVIGATION. THE PURPOSE OF THIS PROJECT IS THEREFORE TO ESTABLISH THE INTRACELLULAR PATHWAYS THAT REGULATE THE MITOCHONDRIAL TRAFFIC OF AQP8BB IN THE GOLDEN SPERM, AS WELL AS TO IDENTIFY THE TYPE OF IONIC CHANNELS PRESENT IN THE SPERM AND THEIR POSSIBLE INTERACTIONS WITH THE AQUAPORINS. ALSO, SINCE WE HAVE RECENTLY DISCOVERED THAT MATURE GOLDEN SPERM ARE COMPETENT FOR THE TRANSLATION OF CERTAIN PROTEINS (SUCH AS SOME AQUAPORINS), WE ALSO PROPOSE TO DEVELOP MRNA SILENCING METHODS IN ORDER TO DEFINE THE PHYSIOLOGICAL FUNCTIONS OF WATER AND IONIC CHANNELS IN MOTILE SPERM. IN THIS WAY, THIS RESEARCH WILL CONTRIBUTE WITH NEW OXIDATIVE STRESS BIOMARKERS AND CINEMATICS FOR THE EVALUATION OF SEMEN QUALITY IN MARINE TELEOSTEOS OF COMMERCIAL INTEREST. (English)

EL CONTROL DE LA CALIDAD DE LOS GAMETOS EN EL CULTIVO DE PECES MARINOS SIGUE REPRESENTANDO UN DESAFIO FUNDAMENTAL PARA LA SOSTENIBILIDAD DE LA ACUICULTURA EUROPEA. EN LOS MACHOS, LA MOTILIDAD DEL ESPERMA SE CONSIDERA EL PARAMETRO MAS EFICAZ PARA ESTIMAR SU CALIDAD (I.E. CAPACIDAD DE FERTILIZACION), A PESAR DE QUE EN TELEOSTEOS MARINOS TODAVIA NO SE CONOCEN BIEN MUCHOS DE LOS MECANISMOS MOLECULARES CLAVE QUE CONTROLAN LA ACTIVIDAD DEL FLAGELO DE LOS ESPERMATOZOIDES. ESTE CONOCIMIENTO ES SIN EMBARGO ESENCIAL PARA DESARROLLAR BIOMARCADORES MOLECULARES ROBUSTOS Y FIABLES PARA EVALUAR LA CALIDAD DEL ESPERMA, LOS CUALES PUEDAN SER UTILES PARA VALORAR EL IMPACTO DE LAS CONDICIONES DE CULTIVO, LOS PROGRAMAS DE SELECCION GENETICA, Y LOS METODOS DE FECUNDACION ARTIFICIAL Y CRIOPRESERVACION DE GAMETOS. POR TANTO, A NIVEL INDUSTRIAL EXISTE UNA IMPORTANTE DEMANDA DE INVESTIGACION BASICA INNOVADORA SOBRE LA FISIOLOGIA DEL PROCESO DE MOTILIDAD ESPERMATICA EN PECES MARINOS. EN ESTUDIOS PREVIOS SOBRE EL ESPERMA DE DORADA (SPARUS AURATA) HEMOS DESCUBIERTO UN NUEVO SISTEMA ANTIOXIDANTE MITOCONDRIAL MEDIADO POR UN CANAL DE AGUA (ACUAPORINA-8BB, AQP8BB), EL CUAL FUNCIONA EN ESTE CASO COMO UNA PEROXIPORINA FACILITANDO LA SALIDA DE H2O2 DE LA MITOCONDRIA Y LA PRODUCCION CONTINUADA DE ENERGIA (ATP) NECESARIA PARA LA MOTILIDAD FLAGELAR. EL TRANSPORTE INTRACELULAR DE LA AQP8BB EN LOS ESPERMATOZOIDES DURANTE SU ACTIVACION EN AGUA DE MAR APARECE POR TANTO COMO UN FACTOR CRUCIAL PARA LA REGULACION DE LA DESINTOXICACION MITOCONDRIAL Y LA MOTILIDAD. SIN EMBARGO, LAS VIAS DE SEÑALIZACION INTRACELULAR QUE REGULAN ESTE MECANISMO SON DESCONOCIDAS, AUNQUE NUESTROS DATOS PRELIMINARES SUGIEREN QUE CIERTAS QUINASAS ACTIVADAS POR ESTRES PODRIAN ESTAR IMPLICADAS. ADEMAS DE LA AQP8BB, LOS ESPERMATOZOIDES MOTILES DE DORADA EXPRESAN OTRAS ACUAPORINAS EN LA CABEZA Y COLA ANTERIOR, LAS CUALES PARECEN CONTROLAR LA TRAYECTORIA Y EL PATRON DE MOVIMIENTO ESPERMATICO. LOS MECANISMOS MOLECULARES QUE SUBYACEN A LA FUNCION DE ESTOS CANALES NO HAN SIDO TODAVIA IDENTIFICADOS, Y PLANTEAMOS LA HIPOTESIS DE QUE ESTOS PODRIAN ASOCIARSE CON CANALES IONICOS PARA ACTIVAR RESPUESTAS OSMOTICAS LOCALES Y MECANISMOS DE TRANSPORTE DE IONES, LOS CUALES PUEDEN MANTENER Y/O DIRIGIR LA NAVEGACION DE LOS ESPERMATOZOIDES. LA FINALIDAD DE ESTE PROYECTO ES POR TANTO ESTABLECER LAS VIAS INTRACELULARES QUE REGULAN EL TRAFICO MITOCONDRIAL DE AQP8BB EN LOS ESPERMATOZOIDES DE DORADA, ASI COMO IDENTIFICAR EL TIPO DE CANALES IONICOS PRESENTES EN LOS ESPERMATOZOIDES Y SUS POSIBLES INTERACCIONES CON LAS ACUAPORINAS. ASIMISMO, YA QUE RECIENTEMENTE HEMOS DESCUBIERTO QUE LOS ESPERMATOZOIDES MADUROS DE DORADA SON COMPETENTES PARA LA TRASLACION DE CIERTAS PROTEINAS (COMO ALGUNAS ACUAPORINAS), PROPONEMOS TAMBIEN DESARROLLAR METODOS DE SILENCIAMIENTO DE MRNAS CON EL FIN DE DEFINIR LAS FUNCIONES FISIOLOGICAS DE LOS CANALES DE AGUA E IONICOS EN ESPERMATOZOIDES MOTILES. DE ESTE MODO, ESTA INVESTIGACION CONTRIBUIRA CON NUEVOS BIOMARCADORES DE ESTRES OXIDATIVO Y CINEMATICOS PARA LA EVALUACION DE LA CALIDAD DEL ESPERMA EN TELEOSTEOS MARINOS DE INTERES COMERCIAL. (English)
