PUBERTY IS THE TRANSITION PROCESS IN WHICH AN IMMATURE INDIVIDUAL REACHES MATURITY FOR THE FIRST TIME AND IS ABLE TO REPRODUCE. PUBERTY CAUSES SEVERAL COMPLEX NEUROENDOCRINE MECHANISMS TO COORDINATE AND ACTIVATE THE HIPOTALAMO-PITUITARY-GONADA AXIS IN VERTEBRATES AND FISH. THE CONTROL OF PUBERTY IS AN IMPORTANT OBJECTIVE IN AQUACULTURE SINCE ITS APPEARANCE ENTAILS MANY PROBLEMS IN PRODUCTIVE AND ECONOMIC TERMS. EARLY FISH SHOW LOWER GROWTH AND CONVERSION RATE, LOW DISEASE RESISTANCE AND A DECREASE IN THE QUALITY OF THEIR MEAT. IN ADDITION, EARLY PUBERTY IS AN ENVIRONMENTAL THREAT AS IT INCREASES THE RISK OF INTERACTIONS BETWEEN WILD AND CULTIVATED POPULATIONS. IN NATURAL CONDITIONS, IN THE MEDITERRANEAN, MALES OF SEA BASS (DICENTRARCHUS LABRAX L.) REACH PUBERTY AT AGE 2 AND FEMALES AT AGE 3. HOWEVER, UNDER INTENSIVE GROWING CONDITIONS, ADVANCED PUBERTY OR PRECOCITY AFFECTS 20.30 % OF MALES IN THE 1-YEAR-OLD POPULATION BEFORE THEY REACH COMMERCIAL SIZE (~18-24 MONTHS). AS FAR AS FEMALES ARE CONCERNED, THE INFORMATION AVAILABLE IS LIMITED, BUT THERE IS EVIDENCE TO SHOW THE EXISTENCE OF PRECOCIOUS FEMALES IN CAPTIVITY. IN THE LAST FEW YEARS, MUCH WORK HAS BEEN DONE TRYING TO DECIPHER THE EFFECT OF PHOTOPERIOD ON SEXUAL MATURITY. CURRENTLY IT IS KNOWN THE EXISTENCE OF A PHOTOPERIODIC WINDOW IN WHICH THE ADMINISTRATION OF CONTINUOUS LIGHT DURING THIS PERIOD REDUCES PRECOCITY IN JUVENILE MALE SEA BASS. RECENTLY, THE DISCOVERY OF THE KISSPEPTIN SYSTEM HAS ALLOWED THE STUDY OF THE PHYSIOLOGICAL ACTION OF THESE PEPTIDES, DEMONSTRATING THAT THEY ARE ESSENTIAL REGULATORS OF GONADOTROPINS. HOWEVER, THE INFORMATION AVAILABLE ON THE GENETIC FACTORS CONTROLLING THIS FEATURE IS LIMITED. THIS REQUIRES NEW APPROACHES TO FIND DIFFERENTIAL GENES AND EXPRESSIONS RELATED TO SEXUAL MATURITY. RECENTLY, HIGH-PERFORMANCE SEQUENCING TECHNOLOGIES HAVE FACILITATED THE GENOME SEQUENCE OF THE BASS, ALLOWING LARGE-SCALE DISCOVERY OF NEW GENES ASSOCIATED WITH PRODUCTIVE CHARACTERS IN THIS AND OTHER SPECIES. THE AIM OF THE PUBERTRAIT PROJECT IS TO STUDY THE GENETIC BASES OF SEXUAL MATURITY IN SEA BASS. FOR THIS PURPOSE, THE TECHNIQUE OF RNA-SEQ AND TRANSCRIPTOMIC ANALYSIS OF THE BRAIN AND GONADA OF MALES IN DIFFERENT STATES OF MATURITY WILL BE USED TO ALLOW ALIGNMENT WITH THE SEA BASS GENOME FOR THE PURPOSE OF DETECTING AND MAPPING SNP MARKERS AND CANDIDATE GENES EXPRESSED DIFFERENTIALLY AND ASSOCIATED WITH SEXUAL MATURITY IN THIS SPECIES. ON THE OTHER HAND, THE DATA IN FEMALES ARE VERY LITTLE DOCUMENTED, AND SINCE THE MECHANISMS OF MATURATION MAY DIFFER BETWEEN SEXES, THE STUDY OF THE PRESENCE OF FEMALES OF EARLY SEA BASS WILL BE CARRIED OUT FROM A HORMONAL, MOLECULAR AND HISTOLOGIC POINT OF VIEW IN 2-YEAR SPECIMENS. IT IS THEREFORE INTENDED TO IDENTIFY NON-LETHAL MARKERS THAT REVEAL TO US THE GONADAL STATE OF DEVELOPMENT OF INDIVIDUALS AND CAN PREDICT THEIR ABILITY TO ACHIEVE CAPTIVE PUBERTY OR NOT. ALL THESE RESULTS WILL BE CRUCIAL TO KNOWING THE GENETIC PREDISPOSITION OF INDIVIDUALS OF SEA BASS AT MATURATION, AND WILL SERVE AS A GUIDE FOR THE ESTABLISHMENT OF SELECTION PROGRAMS IN THIS SPECIES. LA PUBERTAD ES EL PROCESO DE TRANSICION EN EL QUE UN INDIVIDUO INMADURO ALCANZA POR PRIMERA VEZ LA MADUREZ Y ES CAPAZ DE REPRODUCIRSE. LA PUBERTAD HACE QUE VARIOS MECANISMOS NEUROENDOCRINOS COMPLEJOS SE COORDINEN Y ACTIVEN AL EJE HIPOTALAMO- PITUITARIA-GONADA EN VERTEBRADOS Y EN PECES. EL CONTROL DE LA PUBERTAD ES UN OBJETIVO IMPORTANTE EN ACUICULTURA DADO QUE SU APARICION ACARREA MUCHOS PROBLEMAS EN TERMINOS PRODUCTIVOS Y ECONOMICOS. LOS PECES PRECOCES MUESTRAN UN CRECIMIENTO Y TASA DE CONVERSION MENORES, RESISTENCIA BAJA A ENFERMEDADES Y UNA MERMA DE LA CALIDAD DE SU CARNE. ASIMISMO, LA PUBERTAD TEMPRANA ES UNA AMENAZA MEDIOAMBIENTAL DADO QUE INCREMENTA EL RIESGO DE LAS INTERACCIONES ENTRE LAS POBLACIONES SALVAJES Y CULTIVADAS. EN CONDICIONES NATURALES, EN EL MEDITERRANEO, LOS MACHOS DE LUBINA (DICENTRARCHUS LABRAX L.) ALCANZAN LA PUBERTAD A LOS 2 AÑOS Y LAS HEMBRAS A LOS 3 AÑOS. SIN EMBARGO, BAJO CONDICIONES INTENSIVAS DE CULTIVO LA PUBERTAD ADELANTADA O PRECOCIDAD AFECTA AL 20¿30% DE LOS MACHOS DE LA POBLACION DE 1 AÑO DE EDAD ANTES DE QUE ESTOS ALCANCEN EL TAMAÑO COMERCIAL (~18-24 MESES). EN CUANTO A LAS HEMBRAS LA INFORMACION DISPONIBLE ES LIMITADA, PERO HAY EVIDENCIAS QUE DEMUESTRAN LA EXISTENCIA DE HEMBRAS PRECOCES EN CAUTIVIDAD. EN LOS ULTIMOS AÑOS SE HA TRABAJADO MUCHO INTENTANDO DESCIFRAR EL EFECTO QUE TIENE EL FOTOPERIODO SOBRE LA MADUREZ SEXUAL. ACTUALMENTE SE CONOCE LA EXISTENCIA DE UNA VENTANA FOTOPERIODICA EN LA QUE LA ADMINISTRACION DE LUZ CONTINUA DURANTE ESTE PERIODO REDUCE LA PRECOCIDAD EN JUVENILES MACHOS DE LUBINA. RECIENTEMENTE, EL DESCUBRIMIENTO DEL SISTEMA KISSPEPTINA HA PERMITIDO EL ESTUDIO DE LA ACCION FISIOLOGICA DE ESTOS PEPTIDOS, DEMOSTRANDOSE QUE SON REGULADORES ESENCIALES DE LAS GONADOTROPINAS. SIN EMBARGO, LA INFORMACION DISPONIBLE DE LOS FACTORES GENETICOS QUE CONTROLAN ESTE CARACTER ES LIMITADA. ESTO REQUIERE DE NUEVOS ENFOQUES CON LA FINALIDAD DE ENCONTRAR GENES Y EXPRESIONES GENICAS DIFERENCIALES RELACIONADOS CON LA MADUREZ SEXUAL. RECIENTEMENTE, LAS TECNOLOGIAS DE SECUENCIACION DE ALTO RENDIMIENTO HAN FACILITADO LA SECUENCIA DEL GENOMA DE LA LUBINA, LO QUE PERMITIRA A GRAN ESCALA EL DESCUBRIMIENTO DE NUEVOS GENES ASOCIADOS A CARACTERES PRODUCTIVOS EN ESTA Y OTRAS ESPECIES. EL OBJETIVO DEL PROYECTO PUBERTRAIT ES EL ESTUDIO DE LAS BASES GENETICAS DE LA MADUREZ SEXUAL EN LA LUBINA. PARA ELLO SE USARA LA TECNICA DE RNA-SEQ Y ANALISIS TRANSCRIPTOMICO DE CEREBRO Y GONADA DE MACHOS EN DISTINTOS ESTADOS DE MADUREZ QUE PERMITIRA EL ALINEAMIENTO CON EL GENOMA DE LUBINA CON EL PROPOSITO DE DETECTAR Y MAPEAR MARCADORES SNP Y GENES CANDIDATOS EXPRESADOS DE MANERA DIFERENCIAL Y ASOCIADOS A LA MADUREZ SEXUAL EN ESTA ESPECIE. POR OTRA PARTE LOS DATOS EN HEMBRAS ESTAN MUY POCO DOCUMENTADOS, Y PUESTO QUE LOS MECANISMOS DE MADURACION PUEDEN DIFERIR ENTRE SEXOS, EL ESTUDIO DE LA PRESENCIA DE HEMBRAS DE LUBINA PRECOCES SE LLEVARA A CABO DESDE UN PUNTO DE VISTA HORMONAL, MOLECULAR E HISTOLOGICO EN EJEMPLARES DE 2 AÑOS. ASI PUES SE PRETENDE IDENTIFICAR MARCADORES NO-LETALES QUE NOS REVELEN EL ESTADO DE DESARROLLO GONADAL DE LOS INDIVIDUOS Y PUEDAN PREDECIR SU CAPACIDAD PARA ALCANZAR O NO LA PUBERTAD EN CAUTIVIDAD. TODOS ESTOS RESULTADOS SERAN CRUCIALES PARA CONOCER LA PREDISPOSICION GENETICA DE LOS INDIVIDUOS DE LUBINA A LA MADURACION, Y SERVIRA DE GUIA PARA EL ESTABLECIMIENTO DE PROGRAMAS DE SELECCION EN ESTA ESPECIE.
