Objectives: (1) From a population perspective, analyse serum N-glycoma in adults and its possible variations according to demographic variables, toxic habits and lifestyle and diseases prevalent in the population. The study aims, in addition to deepening the mechanistic knowledge of frequent diseases, many of them of inflammatory basis (low-grade chronic inflammation), the establishment of biomarkers in these diseases. (2) Specific study of the glycation of certain serum proteins, especially immunoglobulins and specifically immunoglobulin E (IgE) in relation to alcohol consumption, allergic disease or helminth infection, as well as their differences in these entities and their correlation with clinical aspects. Methodology: (1) The project is implemented as part of a study based on random sampling of the general adult population of a municipality (n=2000), carried out by the same group with a view to multi-objective research into markers of frequent inflammatory diseases in the population. (2) Analysis of N-glycans released after enzymatic action (N-glycoma analysis) or glycopeptides after digestion with endoproteinases (IgE glycosylation study) using HPLC and mass spectrometry (MALDI-TOF). Objetivos: (1) Desde una perspectiva poblacional, analizar el N-glicoma sérico en adultos y sus posibles variaciones en función de variables demográficas, hábitos tóxicos y de estilo de vida y enfermedades prevalentes en la población. El estudio tiene como finalidad, además de profundizar en el conocimiento mecanístico de enfermedades frecuentes, muchas de ellas de base inflamatoria (inflamación crónica de bajo grado), el establecimiento de biomarcadores en esas enfermedades. (2) Estudio específico de la glicación de algunas proteínas séricas, especialmente las inmunoglobulinas y concretamente la inmunoglobulina E (IgE) en relación con consumo de alcohol, enfermedad alérgica o infección por helmintos, así como sus diferencias en esas entidades y su correlación con aspectos clínicos. Metodología: (1) El proyecto se implementa dentro de un estudio basado en un muestreo aleatorio de la población general adulta de un municipio (n=2000), realizado por el mismo grupo con vistas a investigación multi-objetivo en marcadores de enfermedades inflamatorias frecuentes en la población. (2) Análisis del los N-glicanos liberados tras acción enzimática (análisis del N-glicoma) o de glicopéptidos tras digestión con endoproteinasas (estudio de la glicosilación de la IgE) mediante HPLC y espectrometría de masas (MALDI-TOF).
