Background: changes in foetal growth and low birth weight are associated with an increased risk of disease in adult life. Maternal diet and nutritional status, and exposure to environmental pollutants, are considered the main determinants of foetal growth, as they alter the availability of nutrients among which are compounds that play a key role in early development. Objectives: the overall objective of this study is to characterise maternal urinary metaboloma throughout pregnancy, in order to identify metabolomic profiles associated with foetal growth alterations considering sources of variation of metaboloma based on lifestyles and exposure to organophosphate pesticides. Methodology: prospective birth cohort of 646 mother-child binomies. Women will be captured during the first trimester of pregnancy and monitored during pregnancy and birth of the child. A non-directed metabolomic analysis in urine from the first and third trimester of gestation will be performed using liquid chromatography and mass spectrometry (UHPLC-UHR-QTOF System) to identify variations in metabolite levels associated with fetal growth alterations and anthropometry at birth. Multivariate analyses will be performed to explore the association between biomarkers and indicators of metabolomic alterations and the weight of children at birth, considering as covariables the lifestyle, diet and exposure to contaminants of the pregnant women selected for the study. Antecedentes: las alteraciones en el crecimiento fetal y el bajo peso al nacer están asociadas con un mayor riesgo de enfermedades en la vida adulta. La dieta y el estado nutricional maternos, y la exposición a contaminantes ambientales, se consideran los principales determinantes del crecimiento fetal, ya que alteran la disponibilidad de nutrientes entre los que se encuentran compuestos con un papel clave en el desarrollo temprano. Objetivos: el objetivo general de este estudio es caracterizar el metaboloma urinario materno a lo largo del embarazo, para identificar perfiles metabolómicos asociados con alteraciones del crecimiento fetal considerando fuentes de variación del metaboloma basadas en estilos de vida y exposición a plaguicidas organofosforados. Metodología: cohorte de nacimiento prospectiva de 646 binomios madre-hijo/a. Las mujeres se captarán durante el primer trimestre de embarazo y se les efectuará un seguimiento durante el embarazo y el nacimiento del/a hijo/a. Se realizará un análisis metabolómico no dirigido en orina del primer y tercer trimestre de gestación, mediante cromatografía líquida y espectrometría de masas (UHPLC-UHR-QTOF System), para identificar variaciones de los niveles de metabolitos asociadas con alteraciones en el crecimiento fetal y antropometría al nacer. Se realizarán análisis multivariantes para explorar la asociación entre biomarcadores e indicadores de alteraciones metabolómicas y el peso de los niños/as al nacer, considerando como covariables el estilo de vida, dieta y exposición a contaminantes de las gestantes selecionadas para el estudio.
