The purpose of the project is to create and test (operational conditions) an innovative DNA digestion system from biological samples.A DNAza received in the course of the project will have features that allow it to be easily removed or inactivated after reaction and to be used in solutions with high concentrations of salt.Therefore, the project will respond to the market demand for DNAAs easily and efficiently in application.We will design and receive a fusion protein containing a modified hyperactivity sequence of human primates for the fusion of protein, which will allow protein to be assigned to the deposit after reaction.The original modifications of the sequence will result in efficient exports of the protein to a bacterial perplasma and its release into the breeding medium.The treatment of protein from a medium will enable simple and relatively cheap protein to be obtained.A key step in the research will be the optimisation of the express system in order to get the maximum yield of protein production.In the next steps we will work to improve the scale of the over-expression and purification to develop a method of obtaining a protein on a compound scale.The development will be carried out in order to test the nuclear activities in the targeted applications and to develop protocols for users.The final result of the project will be a complete protocol for obtaining on a scale production and use of the nuclease with a treatment system.Reference_reference_programme_aids:SA.41471 (2015/X) _public:Article 25 of Commission Regulation (EC) No 651/2014 of 17 June 2014 declaring certain categories of aid compatible with the internal market in the application of Article 107 and 108 of the Treaty (OJ(OJ LEU L 187/1, 26.06.2014).     Projekt ma na celu stworzenie i przetestowanie (w warunkach operacyjnych) innowacyjnego systemu wytrawiania DNA z próbek biologicznych. DNaza otrzymana w toku realizacji projektu będzie posiadać właściwości pozwalające na jej łatwe usunięcie lub inaktywację po reakcjach oraz stosowanie w roztworach o wysokim stężeniu soli. Tym samym rezultat projektu będzie odpowiedzią na zapotrzebowanie rynku na DNazy łatwe i wydajne w zastosowaniu. Zaprojektujemy i otrzymamy białko fuzyjne zawierające zmodyfikowaną sekwencję hiperaktywnej ludzkiej DNazy I. Partner fuzyjny stanowiący tag, umożliwi wiązanie białka do złoża, co będzie wykorzystane do usuwania nukleazy po reakcjach. Oryginalne modyfikacje sekwencji spowodują wydajny eksport białka do bakteryjnej periplazmy i jego uwalnianie do medium hodowlanego. Oczyszczanie białka z medium umożliwi proste i stosunkowo tanie otrzymywanie białka. Kluczowym etapem badań będzie optymalizacja systemu ekspresyjnego w celu uzyskania maksymalnej wydajności produkcji białka. W kolejnych etapach pracować będziemy nad podniesieniem skali nadekspresji i oczyszczania tak, aby opracować metodę otrzymywania białka na skalę preparatywną. Prace rozwojowe będą prowadzone celem przetestowania nukleazy w docelowych zastosowaniach i opracowania protokołów dla użytkowników. Efektem końcowym projektu będzie kompletny protokół otrzymywania na skalę produkcyjną i stosowania nukleazy z systemem oczyszczania.
