    Le durcissement des artères et l’infarctus sont des pathologies associées au vieillissement. Nous travaillons sur les mécanismes de ces pathologies et plus particulièrement sur des protéines intervenant sur la contraction des artères et du cœur et dans la formation du caillot. Ces protéines appartiennent à une grande famille des petites protéines G. Dans cette famille, nous avons choisi d’étudier un facteur d’échange d’information entre des protéines G qui est le facteur d’échange Arhgef1. Pour cette étude nous utiliserons un modèle de souris génétiquement modifiée et qui porte ou non à l’intérieur des cellules des vaisseaux cette protéine. Des animaux jeunes et vieux feront partie de cette étude et l’objectif est de démontrer que ces facteurs jouent un rôle important dans les anomalies des artères au cours du vieillissement pour ultérieurement mettre au point des médicaments capables de restaurer les propriétés de ce facteur d’échange. Cette étape chez l’animal est indispensable avant de tester ces molécules chez l’homme. (French)

These proteins belong to a large family of small G proteins. In this family, we have chosen to study an information exchange factor between G proteins which is the Arhgef1 exchange factor. For this study, we will use a model of genetically modified mice that carries or does not carry this protein inside the cells of the vessels. Young and old animals will be included in this study and the objective is to demonstrate that these factors play an important role in artery abnormalities during aging, and subsequently develop drugs capable of restoring the properties of this exchange factor. This step in animals is essential before testing these molecules in humans. (English)
