UNA DE LAS ESTRATEGIAS DE CONTROL DE PPH MAS DESEABLES ES EL USO DE VARIEDADES DE JUDIA RESISTENTES. LOS AISLADOS DE PPH SE CLASIFICAN EN 9 RAZAS EN FUNCION A SUS INTERACCIONES CON 8 VARIEDADES DIFERENTES DE JUDIA, QUE SE HAN EXPLICADO POR LA INTERACCION DE 5 PARES EN RELACION GEN A GEN, AUNQUE ES POSIBLE REALIZAR UNA MAYOR DIFERENCIACION EN HASTA 14 RAZAS. POR UN LADO, AUNQUE PPH ES UN PATOVAR GENETICAMENTE HOMOGENEO, LOS DIFERENTES AISLADOS MUESTRAN DIFERENCIAS EN SU VIRULENCIA, ASPECTO QUE REFLEJA LA ESPECIFICIDAD DE RAZA DE LOS MISMOS. LA DISPONIBILIDAD DE LOS GENOMAS CERRADOS DE LAS CEPAS MODELO PPH 1448A Y PSV NCPPB 3335, ASI COMO LAS HERRAMIENTAS GENETICAS Y FUNCIONALES DESARROLLADAS DURANTE LA EJECUCION DE NUESTROS PROYECTOS ANTERIORES, CONVIERTEN A ESTAS BACTERIAS EN MODELOS IDEALES PARA EL ANALISIS DE LA ESPECIFICIDAD DE HUESPED Y LA EVOLUCION DE LA VIRULENCIA. ASI, EL ANALISIS GENOMICO COMPARATIVO Y FUNCIONAL DE CEPAS BACTERIANAS FILOGENETICAMENTE RELACIONADAS FACILITARA LA IDENTIFICACION DE DETERMINANTES DE PATOGENICIDAD A NIVEL DE PATOVAR Y DE RAZA. LOS OBJETIVOS PRINCIPALES DE ESTE PROYECTO SON: 1) IDENTIFICAR Y CARACTERIZAR FUNCIONALMENTE DIFERENCIAS EN EL CONJUNTO DE GENES DE VIRULENCIA DE LOS PATOVARES DE PSV Y LAS RAZAS DE PPH RELACIONADAS CON LA ESPECIFICIDAD DE HUESPED; Y 2) APLICAR LA INFORMACION OBTENIDA MEDIANTE GENOMICA COMPARATIVA Y FUNCIONAL PARA EL DESARROLLO DE HERRAMIENTAS DE DIAGNOSTICO DE LAS RAZAS DE PPH Y DEL AISLADO EMERGENTE DE PSV EN MANDEVILLA. LOS OBJETIVOS ESPECIFICOS SON: 1) ASIGNAR UN PATOVAR A LAS CEPAS DE P. SAVASTANOI AISLADAS DE M. SANDERI. 2) SECUENCIAR Y ANOTAR LOS GENOMAS DE CEPAS REPRESENTATIVAS DE LOS PATOVARES Y LAS RAZAS DE P. IN ADDITION, PSV HAS RECENTLY BEEN REVEALED AS AN EMERGING PATHOGEN IN THE ORNAMENTAL SPECIES MANDEVILLA SANDERI (DIPLADENIA). P. SAVASTANOI PV. PHASEOLICOLA (PPH) IS A PATHOGEN OF ECONOMIC IMPORTANCE IN JUDIA AND ONE OF THE MOST RELEVANT RESEARCH MODELS IN PHYTOBACTERIOLOGY. ONE OF THE MOST DESIRABLE PPH CONTROL STRATEGIES IS THE USE OF RESISTANT JEWISH VARIETIES. PPH ISOLATES ARE CLASSIFIED INTO 9 BREEDS ACCORDING TO THEIR INTERACTIONS WITH 8 DIFFERENT JEWISH VARIETIES, WHICH HAVE BEEN EXPLAINED BY THE INTERACTION OF 5 PAIRS IN GENE-TO-GEN RELATIONSHIP, ALTHOUGH IT IS POSSIBLE TO MAKE A GREATER DIFFERENTIATION IN UP TO 14 BREEDS. BOTH PATHOSYSTEMS OFFER AN UNPRECEDENTED OPPORTUNITY TO STUDY THE MOLECULAR BASES OF GUEST SPECIFICITY. ON THE ONE HAND, ALTHOUGH PPH IS A GENETICALLY HOMOGENOUS PATOVAR, THE DIFFERENT ISOLATES SHOW DIFFERENCES IN THEIR VIRULENCE, AN ASPECT THAT REFLECTS THEIR RACE SPECIFICITY. ON THE OTHER HAND, THE DIFFERENT PATOVARES OF P. SAVASTANOI INDUCE SIMILAR SYMPTOMS (TUMORS) IN BOTH WOODY AND HERBACEOUS HOSTS. THE AVAILABILITY OF THE CLOSED GENOMES OF THE PPH 1448A AND PSV NCPPB 3335 STRAINS, AS WELL AS THE GENETIC AND FUNCTIONAL TOOLS DEVELOPED DURING THE EXECUTION OF OUR PREVIOUS PROJECTS, MAKE THESE BACTERIA IDEAL MODELS FOR THE ANALYSIS OF GUEST SPECIFICITY AND THE EVOLUTION OF VIRULENCE. THUS, THE COMPARATIVE AND FUNCTIONAL GENOMIC ANALYSIS OF PHYLOGENETICALLY RELATED BACTERIAL STRAINS WILL FACILITATE THE IDENTIFICATION OF PATHOGENICITY DETERMINANTS AT THE PATOVAR AND BREED LEVEL. THE MAIN OBJECTIVES OF THIS PROJECT ARE: 1) FUNCTIONALLY IDENTIFY AND CHARACTERISE DIFFERENCES IN THE SET OF VIRULENCE GENES OF PSV PATHOVARS AND PPH BREEDS RELATED TO HOST SPECIFICITY; AND 2) APPLY THE INFORMATION OBTAINED BY COMPARATIVE AND FUNCTIONAL GENOMIC FOR THE DEVELOPMENT OF DIAGNOSTIC TOOLS FOR THE PPH BREEDS AND THE EMERGING ISOLATED PSV IN MANDEVILLA. THE SPECIFIC OBJECTIVES ARE: 1) ASSIGN A PATOVAR TO P. SAVASTANOI STRAINS ISOLATED FROM M. SANDERI. 2) SEQUENCE AND RECORD GENOMES OF REPRESENTATIVE STRAINS OF PATOVARS AND BREEDS OF P. SAVASTANOI. 3) PERFORM A COMPARATIVE GENOMIC ANALYSIS AND A PHYLOGENETIC ANALYSIS OF PSV PATHOVARS AND PPH BREEDS. 4) STUDY THE BIOLOGY AND ROLE IN THE SPECIFICITY OF HOST OF THE PLASMAIDS OF THESE BACTERIAL STRAINS. 5) TO DEEPEN THE ROLE IN THE GUEST SPECIFICITY OF THE SECRETION SYSTEMS, OF THE EFFECTORS OF THE TYPE III SECRETION SYSTEM AND, 6) OF OTHER VIRULENCE FACTORS. 7) TO STUDY THE ROLE OF GENIC EXPRESSION IN THE SPECIFICITY OF GUESTS. 8) DEVELOP SPECIFIC DIAGNOSTIC TOOLS FOR MANDEVILLA ISOLATES AND PPH BREEDS.
