IN PARTICULAR, PEST CONTROL THROUGH THE USE OF PESTICIDES INCREASES THE COST OF PRODUCTION AND ALSO REPRESENTS A RISK FACTOR FOR CONSUMERS AND THE ENVIRONMENT. THE DEVELOPMENT OF PEST-RESISTANT VARIETIES IS THEREFORE A MAJOR CHALLENGE IN THE CONTEXT OF SUSTAINABLE AGRICULTURE. UNLIKE OTHER PLANT SPECIES, THE TOMATO, ALONG WITH OTHER SPECIES OF THE GENUS SOLANUM, HAS AN ATYPICAL COMPOSITION IN THE CONTENT AND PROFILE OF STEROLS, BEING TO HIGHLIGHT THE EXTRAORDINARILY HIGH LEVELS OF CONJUGATED STEROLS. MOREOVER, DURING THE RIPENING OF THE FRUIT THERE ARE VERY IMPORTANT CHANGES IN THE LEVELS AND RELATIVE PROPORTIONS OF THE FREE AND CONJUGATED STEROLS. THE BIOLOGICAL AND EVOLUTIONARY SIGNIFICANCE OF THE PECULIAR METABOLISM OF STEROLS IN TOMATO IS TOTALLY UNKNOWN. RECENT STUDIES INDICATE THAT STEROLS PLAY AN ESSENTIAL ROLE IN PLANT GROWTH AND DEVELOPMENT AND IN THEIR RESPONSE TO DIFFERENT TYPES OF STRESS. BASED ON THESE OBSERVATIONS, AND TAKING INTO ACCOUNT THAT THE KNOWLEDGE OF ENZYMES INVOLVED IN THE SYNTHESIS OF CONJUGATED STEROLS IS STILL VERY LIMITED, A PROJECT AIMED AT STUDYING THE METABOLISM OF THESE COMPOUNDS IN TOMATO IS PROPOSED. ON THE ONE HAND, IT IS PROPOSED TO IDENTIFY AND CHARACTERISE THE ENZYMES INVOLVED IN THE SYNTHESIS OF CONJUGATED STEROLS AND TO CLONE THE CORRESPONDING GENES. THIS WILL ALLOW TO ADDRESS GENETIC APPROACHES BASED ON THE GENERATION OF TRANSGENIC PLANTS AND THE SELECTION OF MUTANT PLANTS AND WITH ALELICAS VARIANTS OF THE GENES OF INTEREST (OBTAINED BY TILLING AND ECOTILLING) WITH CONTENTS AND PROFILES OF MODIFIED STEROLS. THE AVAILABILITY OF THESE PLANTS WILL ALLOW TO EVALUATE THE ROLE OF SPECIFIC STEROLS IN PROCESSES RELATED BOTH TO THE GROWTH AND DEVELOPMENT OF THE PLANT AND ITS RESPONSE TO STRESS AND TO THE MATURATION OF THE FRUIT, THE RESPONSE TO COLD DAMAGE AND SUSCEPTIBILITY TO THE ATTACK BY PATHOGENS.
