The objectives of this work are; describe the modification of gene expression in a group of obese treated with a Mediterranean-pattern caloric restriction after weight loss (6 months). Secondaryly assess whether there are differences in gene expression in patients with secondary improvement to weight loss of; blood pressure, LDL and HDL cholesterol and triglycerides, insulin resistance, PCR, adipocytokines vs. patients in whom this improvement is not detected in these parameters. And analyse the gene expression of responding patients (weight loss >5 %) to non-responsive patients so that alterations in gene expression and the genes involved can be identified to predict the risk of non-response to the diet. Methodology. Our study will genetically evaluate a total of 30 obese patients, determining the following variables (body mass index, waist and hip circumference, total fat mass by impedanciometry, blood pressure, glucose, insulin, insulin resistance index (HOMA-IR), triglycerides, total cholesterol, HDL cholesterol, LDL cholesterol, and C-reactive protein), as well as serum adipocytokines (leptin, adiponectin and resistin). All patients will start treatment with a caloric restriction (500 calories/day) with a Mediterranean pattern, with weekly follow-up by a dietitian-researcher, being re-evaluated within 6 months of starting the diet with the same biochemical and anthropometric determinations. A gene expression study by hybridising full transcriptoma microarrays obtained from peripheral blood cells was performed (basic time and 6 months) all. Differences in transcriptome gene levels will be compared using GeneSpring Gx ® 11.00 software, as well as biochemical functions and pathways involved in both times Ingenuity Pathway Analysis 8.5 ® (English)

    Los objetivos de este trabajo son; describir la modificación de la expresión génica en un grupo de obesos tratados con una restriccion calórica con patrón mediterráneo tras la perdida de peso (6 meses). De manera secundaria valorar si existen diferencias en la expresión génica en los pacientes que presentan una mejoría secundaria a la perdida de peso de ; tensión arterial, LDL yHDL colesterol y triglicéridos, resistencia a la insulina, PCR, adipocitoquinas vs.los pacientes en los que no se detecta esa mejoría en estos parámetros. Y analizar la expresión génica de los pacientes respondedores (perdida de peso >5%) frente a los pacientes no respondedores de modo que se puedan identificar alteraciones de la expresión génica y los genes implicados para predecir el riesgo de no respuesta a la dieta. Metodología. Nuestro trabajo evaluará genéticamente un total de 30 pacientes obesos, determinando las siguientes variables (índice de masa corporal, circunferencia cintura y cadera, masa grasa total mediante impedanciometría, tensión arterial, glucosa, insulina, índice de resistencia a la insulina (HOMA-IR), triglicéridos, colesterol total, HDL colesterol, LDL colesterol, y proteína C reactiva), así como adipocitoquinas séricas (leptina, adiponectina y resistina). Todos los pacientes iniciarán un tratamiento con una restricción calórica (500 calorías/día) con un patrón Mediterráneo, con seguimiento semanal por parte de una dietista-investigadora, siendo reevaluados a los 6 meses de iniciar la dieta con las mismas determinaciones bioquímicas y antropométricas. A todos se les realizara (tiempo basal y a los 6 meses) un estudio de expresión génica mediante la hibridación de microarrays de transcriptoma completo obtenida de células de sangre periférica. Se compararan las diferencias en los niveles génicos de transcriptoma mediante el software GeneSpring Gx ® 11.00, así como las funciones bioquímicas y rutas implicadas en ambos tiempos Ingenuity Pathway Analysis 8.5 ® (Spanish)
