COMPACT CLUSTERS RIPEN LESS EVENLY, REDUCING QUALITY AND COMPLICATING HARVESTING. IN ADDITION, THEY ARE MORE SUSCEPTIBLE TO PESTS AND DISEASES, MAINLY FUNGAL INFECTIONS. FOR ALL THIS THERE IS A GREAT INTEREST IN THE SECTOR TO KNOW THE GENETIC AND PHYSIOLOGICAL BASES THAT AFFECT THIS CHARACTER. IN THE PREVIOUS PROJECT WE ESTABLISHED THAT THE MOST DETERMINING CHARACTERS OF THIS CHARACTER ARE THE LENGTH OF THE MAIN AXES OF THE CLUSTER AND THE TOTAL NUMBER OF BERRIES, WHILE IN THE SECOND END THE SIZE OF THE BERRY APPEARED. SOME CANDIDATE GENES FOUND PROMISING ASSOCIATIONS WITH SHAFT LENGTH AND BERRY SIZE, BUT NOT WITH THE NUMBER OF BERRIES. THE FINAL NUMBER OF BERRIES IN THE CLUSTER IS DUE TO THE NUMBER OF FLOWERS IN THE INFLORESCENCE AND THE RATE OF CURD OF THE FRUIT, MAINLY RESPONSIBLE FOR REPRODUCTIVE BEHAVIOR. IN THIS PROJECT WE WILL CONTINUE THE STUDY OF THE COMPACTNESS OF THE CLUSTER BY TWO PARALLEL ROUTES: For one side, EVALUING THE PREVIOUS DETECTED ASSOCIATIONS, and for another, leading to a phenotyping more than affluent of the reproductive transport of life, RESPONSIBLE OF THE FINAL NUMERO OF RACIME BAYS, AND A NEW Selection and Sequence OF CANDIDATE GENES PERMITING CONSIGURES assocation STUDIES._x000D_ CONCRETING OBJECTIVES ARE: 1) EVALUATE AND VALIDATE THE ASSOCIATIONS DETECTED IN SELECTED GENES FOR THE LENGTH OF THE MAIN CLUSTER AXES AND THE SIZE OF THE BERRY. 2) CHARACTERISE THE REPRODUCTIVE BEHAVIOR OF THE COLLECTION OF VINE VARIETIES USED PREVIOUSLY, WITH SPECIAL EMPHASIS ON THE NUMBER OF FLOWERS AND THE RATE OF CURD OF THE FRUIT. 3) CHARACTERISE THE GENETIC AND MOLECULAR PROCESSES RESPONSIBLE FOR PHENOTYPIC DIFFERENCES BETWEEN MUTANTS THAT DIFFER IN THE REPRODUCTIVE BEHAVIOR AND COMPACTNESS OF THE CLUSTER. TO DO THIS, THEY WILL BE CHARACTERISED AT HORMONAL, TRANSCRIPTOMIC AND GENOMIC LEVEL, AND TO PERFORM ANALYSIS OF SYSTEMS WITH THE DATA SET. 4) IDENTIFY GENES INVOLVED IN THE DETERMINATION OF THE NUMBER OF FLOWERS AND THE RATE OF CURD IN RELATION TO THE COMPACTNESS OF THE CLUSTER, BASED ON THE INFORMATION GENERATED IN THE PREVIOUS OBJECTIVE. 5) IDENTIFY, IN SELECTED CANDIDATE GENES, VARIANTS ALELICAS RESPONSIBLE FOR THE OBSERVED NATURAL VARIATION. With this end, GENETIC ASSOCIATION ANALISIS will be carried out between PHENOTIPES AND POLYMORPHES OF SECUTIONS OF SELECTED VARIES._x000D_ WITH THE FIRST OBJECTIVE CONCERNING THE FIRST OBJECTIVES WITH THE FIRST OBJECTIVE OBJECTIVES AGAINST THE ASSOCIATIONS INCLUDED AND EVENTUALLY DEFERRANT POSIBLE MARCATORS FOR GENETICAL MEETING. THE SECOND VIA OF THE PROJECT WILL ALLOW TO DEFINE THE REPRODUCTIVE BEHAVIOR OF THE VINE, AND ITS RELATIONSHIP WITH THE COMPACTNESS OF THE CLUSTER. COMPARATIVE AND INTEGRATED ANALYSES OF “OMICS” DATA OBTAINED IN MUTANTS WILL ALLOW THE IDENTIFICATION OF INDIVIDUAL GENES AND NETWORKS OF RELATED GENES THAT HAVE AN IMPACT ON THE REPRODUCTIVE BEHAVIOR AND COMPACTNESS OF THE CLUSTER, AND ALSO SELECT THE GENES MOST RELEVANT FOR THE ASSOCIATION ANALYSIS. FINALLY, IT IS EXPECTED THAT THE RESULTS OF ASSOCIATION ANALYSIS CAN CONFIRM THE IMPLICATION OF SOME OF THE SELECTED CANDIDATE GENES IN THE DEFINITION OF THE FACTORS THAT CAUSE THE DIFFERENCES IN THE COMPACTNESS OF THE CLUSTER. LA COMPACIDAD DEL RACIMO ES UN RASGO FUNDAMENTAL DE LA VID (VITIS VINIFERA L.) QUE PUEDE AFECTAR TANTO A LA CALIDAD COMO AL RENDIMIENTO. LOS RACIMOS COMPACTOS MADURAN MENOS UNIFORMEMENTE, REDUCIENDO LA CALIDAD Y COMPLICANDO LA VENDIMIA. ADEMAS SON MAS SUSCEPTIBLES A PLAGAS Y ENFERMEDADES, PRINCIPALMENTE INFECCIONES POR HONGOS. POR TODO ELLO HAY UN GRAN INTERES EN EL SECTOR POR CONOCER LAS BASES GENETICAS Y FISIOLOGICAS QUE AFECTAN A ESTE CARACTER. EN EL PROYECTO ANTERIOR ESTABLECIMOS QUE LOS CARACTERES MAS DETERMINANTES DE ESE CARACTER SON LA LONGITUD DE LOS EJES PRINCIPALES DEL RACIMO Y EL NUMERO TOTAL DE BAYAS, MIENTRAS QUE EN SEGUNDO TERMINO APARECIA EL TAMAÑO DE LA BAYA. EN ALGUNOS GENES CANDIDATOS SE DETECTARON ASOCIACIONES PROMETEDORAS CON LA LONGITUD DE LOS EJES Y EL TAMAÑO DE BAYA, PERO NO CON EL NUMERO DE BAYAS. EL NUMERO FINAL DE BAYAS EN EL RACIMO ES CONSECUENCIA DEL NUMERO DE FLORES EN LA INFLORESCENCIA Y DE LA TASA DE CUAJADO DEL FRUTO, PRINCIPALES RESPONSABLES DEL COMPORTAMIENTO REPRODUCTIVO. EN ESTE PROYECTO SE VA A CONTINUAR EL ESTUDIO DE LA COMPACIDAD DEL RACIMO POR DOS VIAS PARALELAS: POR UN LADO, EVALUANDO LAS ASOCIACIONES PREVIAMENTE DETECTADAS, Y POR OTRO, LLEVANDO A CABO UN FENOTIPADO MAS FINO DEL COMPORTAMIENTO REPRODUCTIVO DE LA VID, RESPONSABLE DEL NUMERO FINAL DE BAYAS DEL RACIMO, Y UNA NUEVA SELECCION Y SECUENCIACION DE GENES CANDIDATOS QUE PERMITAN LOS CONSIGUIENTES ESTUDIOS DE ASOCIACION._x000D_ LOS OBJETIVOS CONCRETOS SON: 1) EVALUAR Y VALIDAR LAS ASOCIACIONES DETECTADAS EN GENES SELECCIONADOS PARA LA LONGITUD DE LOS EJES PRINCIPALES DEL RACIMO Y EL TAMAÑO DE LA BAYA. 2) CARACTERIZAR EL COMPORTAMIENTO REPRODUCTIVO DE LA COLECCION DE VARIEDADES DE VID EMPLEADA ANTERIORMENTE, CON ESPECIAL ENFASIS EN EL NUMERO DE FLORES Y LA TASA DE CUAJADO DEL FRUTO. 3) CARACTERIZAR LOS PROCESOS GENETICOS Y MOLECULARES RESPONSABLES DE LAS DIFERENCIAS FENOTIPICAS ENTRE MUTANTES QUE SE DIFERENCIAN EN EL COMPORTAMIENTO REPRODUCTIVO Y LA COMPACIDAD DEL RACIMO. PARA ELLO SE VAN A CARACTERIZAR A NIVEL HORMONAL, TRANSCRIPTOMICO Y GENOMICO, Y A REALIZAR ANALISIS DE SISTEMAS CON EL CONJUNTO DE DATOS. 4) IDENTIFICAR GENES INVOLUCRADOS EN LA DETERMINACION DEL NUMERO DE FLORES Y LA TASA DE CUAJADO EN RELACION CON LA COMPACIDAD DEL RACIMO, EN BASE A LA INFORMACION GENERADA EN EL ANTERIOR OBJETIVO. 5) IDENTIFICAR, EN GENES CANDIDATOS SELECCIONADOS, VARIANTES ALELICAS RESPONSABLES DE LA VARIACION NATURAL OBSERVADA. CON ESTE FIN SE REALIZARAN ANALISIS DE ASOCIACION GENETICA ENTRE FENOTIPOS Y POLIMORFISMOS DE SECUENCIA DE LAS VARIEDADES SELECCIONADAS._x000D_ CON LA CONSECUCION DEL PRIMER OBJETIVO SE VALIDARAN ALGUNAS DE LAS ASOCIACIONES ENCONTRADAS, Y EVENTUALMENTE SE PODRAN DEFINIR POSIBLES MARCADORES PARA MEJORA GENETICA. LA SEGUNDA VIA DEL PROYECTO PERMITIRA DEFINIR EL COMPORTAMIENTO REPRODUCTIVO DE LA VID, Y SU RELACION CON LA COMPACIDAD DEL RACIMO. LOS ANALISIS COMPARATIVO E INTEGRADO DE LOS DATOS "OMICS" OBTENIDOS EN MUTANTES PERMITIRAN LA IDENTIFICACION DE GENES INDIVIDUALES Y DE REDES DE GENES RELACIONADOS QUE TIENEN UN IMPACTO SOBRE EL COMPORTAMIENTO REPRODUCTIVO Y LA COMPACIDAD DEL RACIMO, Y TAMBIEN SELECCIONAR LOS GENES MAS RELEVANTES PARA EL ANALISIS DE ASOCIACION. FINALMENTE, SE ESPERA QUE LOS RESULTADOS DE ANALISIS DE ASOCIACION PUEDAN CONFIRMAR LA IMPLICACION DE ALGUNOS DE LOS GENES CANDIDATOS SELECCIONADOS EN LA DEFINICION DE LOS FACTORES QUE CAUSAN LAS DIFERENCIAS EN LA COMPACIDAD DEL RACIMO. THE COMPACTNESS OF THE CLUSTER IS A FUNDAMENTAL FEATURE OF THE VINE (VITIS VINIFERA L.) THAT CAN AFFECT BOTH QUALITY AND YIELD.
