IN PARTICULAR, WE WILL INVESTIGATE THE MECHANISMS BY WHICH THE SECRETORY PROTEINS ARE PROCESSED AND TRANSPORTED THROUGH THE GOLGI COMPLEX AND HOW THESE MEMBRANES ARE COMPARTMENTALISED IN A FUNCTIONAL WAY. Finally, we hope that we can get to buy better the mechanisms that underlie the diseases related to the very functioning of the GOLGI COMPLEMENT._x000D_ to overwhelm the risks of this project, we use a new MULTIDISCIPLINAL BIOLOGY TOOLS TOOLS OF MOLECULAR AND CELL BIOLOGY, super-resolution nanoscopy and biophysic modelling. WE WILL FOCUS ON THREE MAIN OBJECTIVES: (1) TO ELUCIDATE THE CONTENTS OF THE INTRA-GOLGI TRANSPORT VESICLES; (2) STUDY THE LATERAL COMPARTMENTALISATION OF GOLGI MEMBRANES; And (3) DEVELOPING A QUANTITATIVE biophysical model of the intra-GOLGI TRAFIC based on our EXPERIMENTAL RESULTS._x000D_ THE NEWS OF THIS PROPOSES CONSISTE IN ASPECTS: PROVIDE CLEAR EXPERIMENTAL RESULTS ON THE CONTENT AND DIRECTIONALITY OF INTRA-GOLGI TRANSPORT VESICLES; THE USE OF SUPER-RESOLUTION NANOSCOPY TECHNIQUES TO STUDY THE LATERAL ORGANISATION OF GOLGI MEMBRANES; AND C) COMBINE ALL THIS INFORMATION TO PROPOSE AND CHARACTERISE IN A BIOPHYSICAL WAY A CONSENSUS MODEL FOR INTRA-GOLGI MEMBRANE TRAFFIC.
