Property / summary: THE MAIN OBJECTIVE OF THE SUBPROJECT IS TO INVESTIGATE THE PHYSIOLOGICAL MECHANISMS INVOLVED IN THE REGULATION OF FISH FEED AND AS VARIOUS FACTORS RELATED TO AQUACULTURE PRODUCTION THAT ALTER WELL-BEING CAN INTERFERE WITH THEM AND THEREFORE ALTER FOOD INTAKE. WE INTEND TO CONTINUE THE STUDIES CARRIED OUT TO DATE BY THE RESEARCH GROUP THAT HAVE HELPED TO LAY THE SCIENTIFIC BASIS FOR THE IMPACT OF NUTRIENT SENSOR SYSTEMS, THE CIRCADIAN SYSTEM AND THE STRESS SIGNALS ON THE INTEGRATED RESPONSE OF FISH FEED. SPECIFICALLY, USING RAINBOW TROUT AS AN EXPERIMENTAL MODEL, IT IS HOPED TO OBTAIN RESULTS IN THIS PROJECT THAT WILL SERVE TO CLARIFY AS THE INFORMATION ON THE ENERGETIC HOMEOSTASIS OF THE ANIMAL, THE ACTIVITY OF THE MONOAMINERGICOS SYSTEMS AND THE CIRCADIAN ACTIVITY MODULATE THE INTAKE OF FOOD. AT THE LEVEL OF THE NUTRIENT SENSOR SYSTEMS, OUR OBJECTIVE AT THE CENTRAL LEVEL (MAINLY IN HYPOTHALAMUS) IS TO CHARACTERISE THE SPECIFIC LOCATION IN NEURONAL NUCLEI OF THE SENSOR SYSTEMS OF GLUCOSE AND FATTY ACIDS ALREADY DESCRIBED, AS WELL AS TO CHARACTERISE SENSOR SYSTEMS NOT PREVIOUSLY STUDIED IN FISH INVOLVED IN GLUCOSE DETECTION (BASED ON CHREB AND NA+, K±ATPASA), LIPIDS (BASED ON LPL, TASTE AND SPECIFIC PUFA RECEPTORS, IN THE LATTER CASE USING SOLE AS AN EXPERIMENTAL MODEL), AMINO ACIDS AND CETONICAL BODIES. WE WILL ALSO STUDY THE IMPLICATION OF CERAMIDES AND THE REGULATORY ROLE OF N-ACYLETHANOLAMINES ON LIPIDIC HOMEOSTASIS AND ITS IMPLICATION IN THE REGULATION OF INTAKE, AS WELL AS THE MECHANISMS THAT ALLOW INTEGRATING THE METABOLIC INFORMATION OF DIFFERENT PATHWAYS TO MODULATE THE INTAKE THROUGH CHANGES IN THE EXPRESSION OF NEUROPEPTIDES. AT THE LEVEL OF MONOAMINERGICOS SYSTEMS, WE WILL STUDY THE ROLE OF INDOLAMINS AND CATECHOLAMINES IN THE INTEGRATION OF STRESS SIGNALS, PHARMACOLOGICALLY CHARACTERISING THE RECEPTORS INVOLVED AND THEIR PARTICIPATION IN THE NEURAL PATHWAYS OF STRESS AND INGESTION REGULATION, AS WELL AS THEIR PARTICIPATION IN THE NEURAL REWARD CIRCUITS, USING FOOD AS REWARD AGENT. FINALLY, IN THE STUDY OF THE CIRCADIAN SYSTEM WE WILL DEEPEN THE CHARACTERISATION OF MOLECULAR MECHANISMS AT THE CENTRAL LEVEL AND EVALUATE AS DIFFERENT ENDOCRINE FACTORS AND RELATED TO INTAKE AND METABOLISM CONDITION THE CIRCADIAN RHYTHMIC ACTIVITY AT CENTRAL AND PERIPHERIC LEVEL. LIKEWISE, THE RHYTHMITY OF THE NEUROTRANSMITTERS MONOAMINERGICOS BRAINS AND THEIR ABILITY TO MODULATE THE RHYTHMS OF CLOCK GENES TO THE HYPOTHALAMIC LEVEL, ALSO DEFINING THEIR IMPLICATION IN THE EFFECTS OF STRESS ON THE CIRCADIAN SYSTEM, WILL BE STUDIED. EL OBJETIVO PRINCIPAL DEL SUBPROYECTO ES INVESTIGAR LOS MECANISMOS FISIOLOGICOS QUE PARTICIPAN EN LA REGULACION DE LA ALIMENTACION EN PECES Y COMO DIVERSOS FACTORES RELACIONADOS CON LA PRODUCCION ACUICOLA QUE ALTERAN EL BIENESTAR PUEDEN INTERFERIR CON ELLOS Y POR TANTO ALTERAR LA INGESTA DE ALIMENTO. PRETENDEMOS CONTINUAR LOS ESTUDIOS REALIZADOS HASTA LA FECHA POR EL GRUPO DE INVESTIGACION QUE HAN PERMITIDO CONTRIBUIR A SENTAR LAS BASES CIENTIFICAS DEL IMPACTO DE LOS SISTEMAS SENSORES DE NUTRIENTES, EL SISTEMA CIRCADIANO Y LAS SEÑALES DE ESTRES SOBRE LA RESPUESTA INTEGRADA DE LA ALIMENTACION EN PECES. CONCRETAMENTE, UTILIZANDO COMO MODELO EXPERIMENTAL LA TRUCHA ARCO IRIS, SE ESPERAN OBTENER RESULTADOS EN EL PRESENTE PROYECTO QUE SERVIRAN PARA ESCLARECER COMO LA INFORMACION SOBRE LA HOMEOSTASIS ENERGETICA DEL ANIMAL, LA ACTIVIDAD DE LOS SISTEMAS MONOAMINERGICOS Y LA ACTIVIDAD CIRCADIANA MODULAN LA INGESTA DE ALIMENTO. A NIVEL DE LOS SISTEMAS SENSORES DE NUTRIENTES NUESTRO OBJETIVO A NIVEL CENTRAL (FUNDAMENTALMENTE EN HIPOTALAMO) ES CARACTERIZAR LA LOCALIZACION ESPECIFICA EN NUCLEOS NEURONALES DE LOS SISTEMAS SENSORES DE GLUCOSA Y ACIDOS GRASOS YA DESCRITOS, ASI COMO CARACTERIZAR SISTEMAS SENSORES NO ESTUDIADOS PREVIAMENTE EN PECES IMPLICADOS EN LA DETECCION DE GLUCOSA (BASADOS EN CHREB Y NA+,K+-ATPASA), LIPIDOS (BASADOS EN LPL, RECEPTORES GUSTATIVOS Y ESPECIFICOS DE PUFA, EN ESTE ULTIMO CASO USANDO COMO MODELO EXPERIMENTAL EL LENGUADO), AMINOACIDOS Y CUERPOS CETONICOS. ESTUDIAREMOS TAMBIEN LA IMPLICACION DE LAS CERAMIDAS Y EL PAPEL REGULADOR DE LAS N-ACILETANOLAMINAS SOBRE LA HOMEOSTASIS LIPIDICA Y SU IMPLICACION EN LA REGULACION DE LA INGESTA, ASI COMO LOS MECANISMOS QUE PERMITEN INTEGRAR LA INFORMACION METABOLICA DE DIFERENTES VIAS PARA MODULAR LA INGESTA A TRAVES DE CAMBIOS EN LA EXPRESION DE NEUROPEPTIDOS. A NIVEL DE LOS SISTEMAS MONOAMINERGICOS, ESTUDIAREMOS EL PAPEL DE LAS INDOLAMINAS Y CATECOLAMINAS EN LA INTEGRACION DE SEÑALES DE ESTRES, CARACTERIZANDO FARMACOLOGICAMENTE LOS RECEPTORES IMPLICADOS Y SU PARTICIPACION EN LAS VIAS NEURALES DEL ESTRES Y DE REGULACION DE INGESTA, ASI COMO SU PARTICIPACION EN LOS CIRCUITOS NEURONALES DE RECOMPENSA, UTILIZANDO EL ALIMENTO COMO AGENTE DE RECOMPENSA. FINALMENTE, EN EL ESTUDIO DEL SISTEMA CIRCADIANO PROFUNDIZAREMOS EN LA CARACTERIZACION DE LOS MECANISMOS MOLECULARES A NIVEL CENTRAL Y EVALUAREMOS COMO DISTINTOS FACTORES ENDOCRINOS Y RELACIONADOS CON INGESTA Y EL METABOLISMO CONDICIONAN LA ACTIVIDAD RITMICA CIRCADIANA A NIVEL CENTRAL Y PERIFERICO. ASIMISMO, SE ESTUDIARA LA RITMICIDAD DE LOS NEUROTRANSMISORES MONOAMINERGICOS CEREBRALES Y SU CAPACIDAD PARA MODULAR LOS RITMOS DE LOS GENES RELOJ A NIVEL HIPOTALAMICO, DEFINIENDO ADEMAS SU IMPLICACION EN LOS EFECTOS DEL ESTRES SOBRE EL SISTEMA CIRCADIANO.
