    Hipótesis: El sexo y las hormonas sexuales influyen en la fisiopatología, presentación clínica y curso a largo plazo de los trastornos metabólicos crónicos de etiología multifactorial compleja. Objetivos: 1- Estudiar la influencia del sexo y de las hormonas sexuales, y su interacción con la obesidad, en la fisiopatología de la disfunción del tejido adiposo y sus comorbilidades. 2- Estudiar la influencia del sexo y de las hormonas sexuales en la patogenia, presentación clínica, respuesta al tratamiento y pronóstico de la obesidad grave (mórbida) y su comorbilidades 3- Estudiar la influencia del tratamiento de la disfunción gonadal (exceso androgénico en la mujer y deficiencia de andrógenos en el varón) sobre la disfunción del tejido adiposo y sus comorbilidades. 4- Estudiar la influencia del sexo y de las hormonas sexuales sobre el transcriptoma, proteoma, metaboloma y microbioma de varios fluidos, secreciones y tejidos relacionados con los trastornos descritos en los objetivos anteriores. 5- Identificar biomarcadores diagnósticos y pronósticos de los trastornos metabólicos crónicos específicos para cada sexo. 6- Desarrollar y proponer guías clínicas para el diagnóstico y manejo de estos trastornos específicas para cada sexo. Metodología: cuatro paquetes de trabajo (WPs) complementarios: WP1: ensayo clínico en ratas (intervención con androgenización prenatal y/u obesidad inducida por dieta rica en grasa) y dos estudios prospectivos en humanos (evolución de disfunción gonadal tras pérdida de peso y evolución metabólica tras tratamiento hormonal de disfunción gonadal); WP2: desarrollo de biomarcadores pronósticos y de guías clínicas específicas para cada sexo; WP3: Descubrimiento de biomarcadores mediante técnicas “ómicas” no dirigidas; y WP4: Validación de biomarcadores mediante técnicas dirigidas.
