THE PROJECT AIMS TO ADDRESS THE CHALLENGES RELATED TO FOOD SECURITY AND THE PRODUCTIVITY AND SUSTAINABILITY OF MARINE AQUACULTURE, WHICH ARE CURRENTLY TO BE ADDRESSED IN SPAIN AND EUROPE. THE AIM IS TO DEVELOP NEW MOLECULAR TOOLS TO IMPROVE THE MANAGEMENT AND CONSERVATION OF GENETIC RESOURCES. BASED ON OUR DISCOVERIES ABOUT THE FUNDAMENTAL AND PRESERVED ROLE OF THE SPECIFIC WATER CHANNEL OF TELEOSTEOS, AQUAPORINE-1AB (AQP1AB), IN THE FORMATION OF PELAGIC EGGS (FLOATING) AND THE ACTIVATION OF THE SPERMATICA MOTILITY IN MARINE FISH, THE AIM IS TO DEVELOP NEW MOLECULAR MARKERS FOR THE MONITORING OF THE QUALITY OF GAMETES IN SPECIES OF GREAT COMMERCIAL INTEREST IN SPAIN, SUCH AS GOLDEN (SPARUS AURATA) AND SENEGALESE SOLE (SOLEA SENEGALENSIS). THE USE OF THESE GAMETE QUALITY MARKERS WILL BE OF GREAT USE FOR GENETIC SELECTION PROGRAMS CURRENTLY UNDER WAY IN GOLD AND OTHER MARINE SPECIES, IN WHICH THE LONG-TERM CONSERVATION OF GENICAS (ALLELES) VARIANTS ASSOCIATED WITH CHARACTERS OF COMMERCIAL INTEREST, INCLUDING REPRODUCTIVE CHARACTERS, IS AN ESSENTIAL ASPECT. BY STUDYING THE FUNCTION OF AQP1AB GENES IN AN EX VIVO BIOASSAY BASED ON AMPHIBIAN OOCYTES, WE HAVE RECENTLY DISCOVERED THAT THERE ARE ABERRANT ALLELES OF AQP1AB IN CULTIVATED GOLDS THAT GENERATE NON-VIABLE (NON-FLOATING) EGGS. WE HAVE ALSO IDENTIFIED A SPECIFIC MECHANISM OF TELEOSTEOS THAT CONTROLS INTRACELLULAR TRAFFIC OF AQP1AB, WHICH IS CRUCIAL FOR THE HYDRATION OF OOCYTES AND MAY ALSO BE NECESSARY DURING SPERMATIC ACTIVATION. THEREFORE OUR GOAL IS TO IDENTIFY THE VITAL FACTORS THAT REGULATE THESE MECHANISMS AS NEW MOLECULAR MARKERS OF THE QUALITY OF GAMETES. IN ADDITION, IN ORDER TO INVESTIGATE THE CAUSES RELATED TO THE DRASTIC REDUCTION IN THE NUMBER OF VIABLE EGGS BEFORE SPAWNING, WHICH OCCURS IN BREEDERS UNDER INADEQUATE CULTIVATION CONDITIONS, WE WILL EXAMINE THE DEREGULATION AND FUNCTION OF THE AQUAPORINS AS BIOMARKERS OF THE FOLLICULAR ATRESIA OVARICA, THE MAIN FACTOR THAT AFFECTS FERTILITY IN FEMALES. IN THIS WAY, THIS RESEARCH WILL CONTRIBUTE TO: (A) NEW MOLECULAR MARKERS OF GAMETE QUALITY FOR THE SELECTION OF BREEDING ANIMALS AND THEIR USE IN MARINE FISH CONSERVATION AND GENETIC SELECTION PROGRAMMES, WHICH ARE A PRIORITY FOR SPANISH AND EUROPEAN AQUACULTURE; AND (B) NEW BASIC INFORMATION ON THE MOLECULAR MECHANISMS THAT CONTROL THE PRODUCTION OF VIABLE GAMETES IN FISH. EL PROYECTO TIENE COMO OBJETIVO HACER FRENTE A LOS RETOS RELACIONADOS CON LA SEGURIDAD ALIMENTARIA Y LA PRODUCTIVIDAD Y SOSTENIBILIDAD DE LA ACUICULTURA MARINA LOS CUALES DEBEN ABORDARSE ACTUALMENTE EN ESPAÑA Y EUROPA. PARA ELLO SE PRETENDEN DESARROLLAR NUEVAS HERRAMIENTAS MOLECULARES PARA MEJORAR LA GESTION Y CONSERVACION DE LOS RECURSOS GENETICOS. A PARTIR DE NUESTROS DESCUBRIMIENTOS SOBRE EL PAPEL FUNDAMENTAL Y CONSERVADO DEL CANAL DE AGUA ESPECIFICO DE TELEOSTEOS, ACUAPORINA-1AB (AQP1AB), EN LA FORMACION DE LOS HUEVOS PELAGICOS (FLOTANTES) Y LA ACTIVACION DE LA MOTILIDAD ESPERMATICA EN PECES MARINOS, EL OBJETIVO ES DESARROLLAR NUEVOS MARCADORES MOLECULARES PARA LA MONITORIZACION DE LA CALIDAD DE LOS GAMETOS EN ESPECIES DE GRAN INTERES COMERCIAL EN ESPAÑA, COMO LA DORADA (SPARUS AURATA) Y EL LENGUADO SENEGALES (SOLEA SENEGALENSIS). EL USO DE ESTOS MARCADORES DE CALIDAD DE GAMETOS SERA DE GRAN UTILIDAD PARA PROGRAMAS DE SELECCION GENETICA ACTUALMENTE EN CURSO EN LA DORADA Y OTRAS ESPECIES MARINAS, EN LOS CUALES LA CONSERVACION A LARGO PLAZO DE LAS VARIANTES GENICAS (ALELOS) ASOCIADAS CON CARACTERES DE INTERES COMERCIAL, INCLUIDOS CARACTERES REPRODUCTIVOS, ES UN ASPECTO ESENCIAL. MEDIANTE EL ESTUDIO DE LA FUNCION DE LOS GENES AQP1AB EN UN BIOENSAYO EX VIVO BASADO EN OOCITOS DE ANFIBIO, HEMOS DESCUBIERTO RECIENTEMENTE QUE EXISTEN ALELOS ABERRANTES DE AQP1AB EN DORADAS CULTIVADAS QUE GENERAN HUEVOS NO VIABLES (NO FLOTANTES). ASIMISMO, HEMOS IDENTIFICADO UN MECANISMO ESPECIFICO DE TELEOSTEOS QUE CONTROLA EL TRAFICO INTRACELULAR DE LA AQP1AB, EL CUAL ES CRUCIAL PARA LA HIDRATACION DE LOS OOCITOS Y PODRIA SER TAMBIEN NECESARIO DURANTE LA ACTIVACION ESPERMATICA. POR TANTO NUESTRO OBJETIVO ES IDENTIFICAR LOS FACTORES VITALES QUE REGULAN ESTOS MECANISMOS COMO NUEVOS MARCADORES MOLECULARES DE LA CALIDAD DE LOS GAMETOS. ADEMAS, CON EL FIN DE INVESTIGAR LAS CAUSAS RELACIONADAS CON LA DRASTICA REDUCCION DEL NUMERO DE HUEVOS VIABLES ANTES DEL DESOVE, LO CUAL OCURRE EN REPRODUCTORES BAJO INADECUADAS CONDICIONES DE CULTIVO, EXAMINAREMOS LA DESREGULACION Y LA FUNCION DE LAS ACUAPORINAS COMO BIOMARCADORES DE LA ATRESIA FOLICULAR OVARICA, PRINCIPAL FACTOR QUE AFECTA LA FECUNDIDAD EN HEMBRAS. DE ESTE MODO, ESTA INVESTIGACION CONTRIBUIRA CON: (A) NUEVOS MARCADORES MOLECULARES DE CALIDAD DE GAMETOS PARA LA SELECCION DE REPRODUCTORES Y SU USO EN PROGRAMAS DE CONSERVACION Y SELECCION GENETICA DE PECES MARINOS, LOS CUALES SON UNA PRIORIDAD PARA LA ACUICULTURA ESPAÑOLA Y EUROPEA; Y (B) NUEVA INFORMACION BASICA SOBRE LOS MECANISMOS MOLECULARES QUE CONTROLAN LA PRODUCCION DE GAMETOS VIABLES EN PECES.
