    EL CANCER DE PANCREAS (PC) ES LA CUARTA CAUSA DE MUERTE RELACIONADA CON EL CANCER Y SIGUE SIENDO INTRATABLE, CON UNA TASA DE SUPERVIVENCIA MUY POBRE A LOS 5 AÑOS. CUANDO SE DIAGNOSTICA, MAS DEL 80% DE LOS PACIENTES TIENEN LA ENFERMEDAD AVANZADA, LO CUAL NO PERMITE SU RESECCION QUIRURGICA. ENTRE LOS FACTORES QUE CONTRIBUYEN A SU LETALIDAD SE INCLUYEN LAS CELULAS MADRE CANCERIGENAS (CSCS), UNA PEQUEÑA SUBPOBLACION DENTRO DE LOS TUMORES RESPONSABLE DE LA INICIACION DEL CANCER, EL CRECIMIENTO, LA RECIDIVA Y RESISTENCIA A LA QUIMIOTERAPIA. LAS CSC PANCREATICAS (PCSCS) SE CARACTERIZAN POR LA SOBREEXPRESION DE MARCADORES DE SUPERFICIE ESPECIFICOS CD24, CD44, ESA, C-MET O CXCR4, UNA ALTA ACTIVIDAD ALDH, LA EXCLUSION DEL COLORANTE HOECHST Y UN POTENCIAL ALTAMENTE TUMORIGENICO EN XENOINJERTOS. LAS ESTRATEGIAS TERAPEUTICAS DIRIGIDAS A LA MODULACION DE ESTAS VIAS O HACIA MARCADORES DE SUPERFICIE DE PCSCS PUEDEN TENER IMPORTANTES APLICACIONES CLINICAS. ESTO PERMITE LA SELECCION DE ESTRATEGIAS ANTITUMORALES DIRIGIDAS. ADEMAS, HEMOS DEMOSTRADO EL MECANISMO DE ACCION Y LAS PROPIEDADES ANTITUMORALES FRENTE A RUTAS DE CSC DE NUEVOS COMPUESTOS NATURALES Y SINTETICOS. TAMBIEN, EN COLABORACION CON MIEMBROS DE SUBPROYECTO 1, HEMOS DISEÑADO Y EVALUADO LA EFICACIA DE NANOCAPSULAS DE ACEITE DE OLIVA FUNCIONALIZADAS (LNCS). EN ESTE PROYECTO PROPONEMOS EVALUAR Y VALIDAR NCS DE ACEITE DE OLIVA ORALES DIRIGIDAS COMO HERRAMIENTAS TERAPEUTICAS FRENTE A PCSCS USANDO XENOINJERTOS DERIVADOS DEL PACIENTE CON PC._x000D_ NUESTRA PRIORIDAD ES LA TRASLACION DE LA INVESTIGACION BASICA A LA APLICACION CLINICA, CON UN EQUIPO INTERDISCIPLINAR COMPUESTO POR INVESTIGADORES BASICOS Y CLINICOS, Y EL APOYO DE TRES EPOS. PANCREATIC CSCS (PCSCS) ARE CHARACTERISED BY THE OVEREXPRESSION OF SPECIFIC SURFACE MARKERS CD24, CD44, THAT, C-MET OR CXCR4, A HIGH ALDH ACTIVITY, THE EXCLUSION OF HOECHST DYE AND A HIGHLY TUMORIGENIC POTENTIAL IN XENOGRAFTS. THERAPEUTIC STRATEGIES AIMED AT MODULATING THESE PATHWAYS OR TOWARDS PCSCS SURFACE MARKERS CAN HAVE IMPORTANT CLINICAL APPLICATIONS. HOWEVER, A LIMITING FACTOR IS THE DIFFICULTY OF ISOLATING AND MAINTAINING ITS CHARACTERISTICS STEM. RECENTLY, OUR RESEARCH GROUP HAS PATENTED A METHOD AND MEANS OF CULTIVATION [P201400666] OF ISOLATION AND ENRICHMENT OF SUBPOPULATIONS OF CSCS IN ESTABLISHED LINES AND PRIMARY CROPS FOR TWO WEEKS WITHOUT PERFORMING ANY ADDITIONAL PROCESSING. THIS ALLOWS THE SELECTION OF TARGETED ANTITUMOUR STRATEGIES. IN ADDITION, WE HAVE DEMONSTRATED THE MECHANISM OF ACTION AND ANTITUMOUR PROPERTIES AGAINST CSC PATHWAYS OF NEW NATURAL AND SYNTHETIC COMPOUNDS. ALSO, IN COLLABORATION WITH SUBPROJECT MEMBERS 1, WE HAVE DESIGNED AND EVALUATED THE EFFECTIVENESS OF FUNCTIONALISED OLIVE OIL NANOCAPSULAS (LNCS). Given that these LNCs allow the transport of pharmacies clinically used in more low and more toxic conditions and the union of anti-domestic people in their superfice, we start from the hypothesis of which new intelligent LNCs loaded with anti-CSC pharmacies will be biodisponable by ORAL VIA and INHIBIRAN TUMORAL GROWING AND METASTASIS in a preclinical model of PC._x000D_ OBJECTIVES: ASSESS THE IN VITRO ABILITY OF NSCL TO PASS THE GASTROINTESTINAL BARRIER AND ITS ORAL BIOAVAILABILITY IN VIVO; TO STUDY THE MOLECULAR MECHANISM OF NEW NATURAL AND SYNTHETIC COMPOUNDS VERSUS SCSCS ISOLATED FROM CELL LINES AND PATIENT SAMPLES; TO EVALUATE THE IN VITRO AND IN VIVO EFFICACY AND TOXICITY OF FUNCTIONALISED AND PHARMACO-CHARGED NSCLS AGAINST SCSCS; IV) EXAMINE BIODISTRIBUTION, SAFETY AND EFFICACY OF INTELLIGENT NSCLS IN SUBCUTANEOUS XENOGRAFTS AND ORTOTOPICOS DERIVED FROM PC PATIENTS. _x000D_ challenge: CURRENT TREATMENTS AGAINST PC ARE INEFFICIENT, TOXIC TO NORMAL CELLS AND FAIL TO SELECTIVELY KILL SCSCS. THERE ARE VERY FEW STUDIES WITH NCS USED BY ORAL VIA FOR THE SYSTEMIC RELEASE OF SELECTIVE CANCER PHARMACOS AS AN ALTERNATIVE TO INTRAVENOUS PATHWAY. In this project we propose to evaluate and valiate OIL OIL NCS DIRIGIDED AS TERAPEUTIC TOOLS FENT TO PCSCS USED XENOINJERTS DERIVED PATIENT WITH PC._x000D_Our PRIORITY is the translation of basic research into CLINICAL APPLICATION, with an INTERDISCIPLINATE TEAM BY BASIC AND CLINICAL RESEARCHERS, AND THE SUPPORT OF THREE EPOS. WE HOPE TO CREATE NEW INTERNATIONAL COLLABORATIONS THROUGH SPECIALISED NETWORKS (COST) AND APPLY TO THE H2020 FINANCING PROGRAM. (English)
